Dose Optimization to Mitigate the Risk of CRS with Elranatamab in Multiple Myeloma
Mohamed Elmeliegy,Andrea Viqueira,Erik Vandendries,Anne Hickman,Jennifer E. Hibma,Hoi-Kei Lon,Joseph Piscitelli,Pooneh Soltantabar,Diane Wang,Sibo Jiang,G. Finn,Akos Czibere +11 more
TL;DR: In this article , the authors presented a pooled analysis of CRS across 4 clinical studies leading to the selection and validation of the optimal priming regimen for elranatamab as a 2 step-up priming strategy.

A Systematic Meta-Analysis of Cytokine Release Syndrome Incidence in B-Cell Maturation Antigen-Targeting Chimeric Antigen Receptor T-Cell Therapy and Bispecific Antibodies for Patients with Relapsed and/or Refractory Multiple Myeloma
Sheena Sharma,Diane Wang,Hoi-Kei Lon,Pooneh Soltantabar,Andrea Viqueira,Akos Czibere,Anne Hickman,Jane Liang White,Mohamed Elmeliegy +8 more
TL;DR: In this paper , the authors used a meta-analysis approach to compare CRS profile in BCMA-targeting CAR-Ts vs. BsAbs in patients with relapsed and/or refractory multiple myeloma.

Impact of Treatment Modality and Route of Administration on Cytokine Release Syndrome in Relapsed or Refractory Multiple Myeloma: A Meta-Analysis.
Pooneh Soltantabar,Sheena Sharma,Diane Wang,Hoi-Kei Lon,Akos Czibere,Anne Hickmann,Mohamed Elmeliegy +6 more
TL;DR: Meta-analysis comparing cytokine release syndrome (CRS) profiles in BCMA-targeting CAR-T and BsAb therapies in relapsed or refractory multiple myeloma (RRMM) suggests that CAR-T therapies are associated with higher CRS incidence and severity compared with BsAbs. The s.c. route of BsAb administration may be associated with a lower risk of CRS grade ≥ 3 compared with the i.v. route.

Elranatamab Exposure-Efficacy Analysis in Patients with Relapsed or Refractory Multiple Myeloma: Insights from Pooled Magnetismm Studies
Hoi Kei Lon,Jennifer Hibma,Jason Williams,Diane Wang,David Gifondorwa,Wei An Ma,Lindsay King,Umberto Conte,Akos Czibere,Mohamed Elmeliegy +9 more
TL;DR: The efficacy endpoint ORR demonstrated E-R relationships with total and free ELRA exposures and lower baseline sBCMA level was associated with higher ORR.

Abstract 5063: Optimizing benefit/risk in oncology: Review of post marketing dose optimization
TL;DR: In this article , a review of post-marketing dose optimization studies for oncology molecularly targeted (MTAs) and immune-oncology (IO) agents is presented.
