Synergistic improvement of N-acetylglucosamine production by engineering transcription factors and balancing redox cofactors.
Deng Chen,Xueqin Lv,Jianghua Li,Hongzhi Zhang,Yanfeng Liu,Guocheng Du,Rodrigo Ledesma Amaro,Long Liu +7 more
TL;DR: In this article, a global transcription machinery engineering (gTME) and redox cofactors engineering was used to balance the central carbon/nitrogen metabolism of Corynebacterium glutamicum S9114 to promote the production of the nutraceutical Nacetylglucosamine (GlcNAc).

Metabolic engineering of Corynebacterium glutamicum S9114 based on whole-genome sequencing for efficient N-acetylglucosamine synthesis.
TL;DR: This is the first report concerning the metabolic engineering of C. glutamicum, and the results of this study provide a good starting point for further metabolic engineering to achieve industrial-scale production of GlcNAc.

Efficient expression of cyclodextrin glycosyltransferase from Geobacillus stearothermophilus in Escherichia coli by promoter engineering and downstream box evolution
TL;DR: The results obtained here indicate that in addition to promoter type, the spacer sequence of the promoter and the downstream box are important target elements for improved gene expression.

Enzyme Engineering and Industrial Bioprocess
Deng Chen,Huang Tingting,Jiang Zhu,Xueqin Lv,Long Liu,Jian Chen,Guocheng Du +6 more
- 01 Jan 2019
TL;DR: This chapter mainly introduces enzyme engineering research from the following three aspects: the use of genetic engineering technology to produce large amounts of enzyme preparations; through molecular site-directed mutagenesis and in vitro molecular-directed evolution of natural enzyme protein molecular modification; finally, the industrial application of enzymes examples.

Corynebacterium glutamicum for producing N-acetylglucosamine and application of corynebacterium glutamicum
Liu Long,Du Guocheng,Chen Jian,Jianghua Li,Deng Chen +4 more
- 21 Aug 2018
TL;DR: In this paper, the authors used a corynebacterium glutamicum strain S9114 as an original strain, and a key enzyme gene in a synthesis way of acetylglucosamine was expressed by using a shuttleexpression vector pJYW-4 of the corynbacteriam and escherichia coli; with glucosamine-6-bacillus stearothermophilus phosphotransacetylase GNAl derived from caenorhabditis elegans, the synthesis way was enhanced by inserting repeated pal
