Ascorbic acid enhances tet-mediated 5-methylcytosine oxidation and promotes DNA demethylation in mammals
Ruichuan Yin,Shi-Qing Mao,Bailin Zhao,Zechen Chong,Ying Yang,Ying Yang,Chao Zhao,Dapeng Zhang,Hua Huang,Juan Gao,Zheng Li,Yan Jiao,Cuiping Li,Shengquan Liu,Danni Wu,Weikuan Gu,Yun-Gui Yang,Yun-Gui Yang,Guoliang Xu,Hailin Wang +19 more
TL;DR: The data establish a direct link among AA, Tet, and DNA methylation, thus revealing a role of AA in the regulation of DNA modifications.

Redox-active quinones induces genome-wide DNA methylation changes by an iron-mediated and Tet-dependent mechanism
Bailin Zhao,Ying Yang,Xiaoli Wang,Zechen Chong,Ruichuan Yin,Shuhui Song,Chao Zhao,Cuiping Li,Hua Huang,Bao-Fa Sun,Danni Wu,Kang Xuan Jin,Maoyong Song,Ben-Zhan Zhu,Guibin Jiang,Jannie M. Rendtlew Danielsen,Guoliang Xu,Yun-Gui Yang,Hailin Wang +18 more
TL;DR: Redox-active quinones, a ubiquitous class of chemicals found in natural products, cancer therapeutics and environment, stimulate the conversion of 5mC to 5hmC in vivo, and increase 5hm C in 5751 genes in cells, suggesting an iron-regulated Tet-dependent DNA demethylation mechanism mediated by redox- active biomolecules.
Detection of 8-hydroxydeoxyguanosine (8-OHdG) as a biomarker of oxidative damage in peripheral leukocyte DNA by UHPLC-MS/MS.
TL;DR: It is demonstrated that the level of leukocyte 8-OHdG of the cancer patients group ( n=46) is significantly higher than that of the health control (n=75) and the developed UHPLC-MS/MS method was developed and validated to detect 8- OHdG in human peripheral leukocytes.

Detection of 1,N2-propano-2′-deoxyguanosine adducts in genomic DNA by ultrahigh performance liquid chromatography-electrospray ionization-tandem mass spectrometry in combination with stable isotope dilution
TL;DR: A sensitive ultrahigh performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UHPLC-ESI-MS/MS) method for accurate quantification of two diastereomers of ProdG adducts and can be detected in untreated human MRC5 cells with a frequency of 2.4-3.5Adducts per 10(8) nucleotides.

UHPLC-Q-TOF/MS detection of UV-induced TpT dimeric lesions in genomic DNA.
TL;DR: It is demonstrated that only are TpT dimeric lesions detectable in genomic DNA of HCT116 cells upon UVC irradiation, and the proposed UHPLC-Q-TOF/MS method is promising for accurate identification and quantitative detection of UV-induceddimeric lesions in cellular DNA.
