Reducing tau aggregates with anle138b delays disease progression in a mouse model of tauopathies
Jens Wagner,Sybille Krauss,Song Shi,Sergey Ryazanov,Julia Steffen,Carolin Miklitz,Andrei Leonov,Alexander Kleinknecht,Bettina Göricke,Jochen H. Weishaupt,Daniel Weckbecker,Anne Reiner,Wolfgang Zinth,Johannes Levin,Dan Ehninger,Stefan Remy,Hans A. Kretzschmar,Christian Griesinger,Armin Giese,Martin Fuhrmann +19 more
TL;DR: It is shown that the di-phenyl-pyrazole anle 138b binds to aggregated tau and inhibits tau aggregation in vitro and in vivo and that reducing tau aggregates with anle138b may represent an effective and promising approach for the treatment of human tauopathies.
[ 11 C]MODAG-001-towards a PET tracer targeting α-synuclein aggregates
Laura Kuebler,Sabrina Buss,Andrei Leonov,Sergey Ryazanov,Felix Schmidt,Andreas Maurer,Daniel Weckbecker,Anne M. Landau,Thea P. Lillethorup,Daniel Bleher,Ran Sing Saw,Bernd J. Pichler,Christian Griesinger,Armin Giese,Kristina Herfert +14 more
TL;DR: In this paper, a target-specific tracer to detect pathological aggregates of α-synuclein (αSYN) remains lacking, and a positron emission tomography (PET) tracer based on anle138b, a compound shown to have therapeutic activity in animal models of neurodegenerative diseases.
Tau-induced mitochondrial membrane perturbation is dependent upon cardiolipin.
Angelique Camilleri,Stephanie Ghio,Mario Caruana,Daniel Weckbecker,Felix Schmidt,Frits Kamp,Andrei Leonov,Sergey Ryazanov,Christian Griesinger,Armin Giese,Ruben J. Cauchi,Neville Vassallo +11 more
TL;DR: A scenario in which toxic cytosolic entities of tau protein would target mitochondrial organelles by associating with their CL-rich membrane domains, leading to membrane poration and compromised mitochondrial structural integrity is advanced.

Biogenesis of Mitochondrial Proteins
TL;DR: This chapter gives an overview on the mitochondrial protein translocases and the mechanisms by which they drive the transport and assembly of mitochondrial proteins.

Safety, tolerability and pharmacokinetics of the oligomer modulator anle138b with exposure levels sufficient for therapeutic efficacy in a murine Parkinson model: A randomised, double-blind, placebo-controlled phase 1a trial
Johannes Levin,Nand Sing,Sue Melbourne,Amber Morgan,Carla Mariner,Maria Grazia Spillantini,Michal Wegrzynowicz,Jeffrey W. Dalley,Simon Langer,Sergey Ryazanov,Andrei Leonov,Christian Griesinger,Felix Schmidt,Daniel Weckbecker,Kai Prager,Torsten Matthias,Armin Giese +16 more
TL;DR: Anle138b was studied in a single-centre, double-blind, randomised, placebo-controlled single ascending dose and multiple ascending dose (MAD) study in healthy subjects as mentioned in this paper .
