Assessing skin sensitization hazard in mice and men using non-animal test methods.
Daniel Urbisch,Annette Mehling,Katharina Guth,Tzutzuy Ramirez,Naveed Honarvar,Susanne N. Kolle,Robert Landsiedel,Joanna Jaworska,Petra S. Kern,Frank Gerberick,Andreas Natsch,Roger Emter,Takao Ashikaga,Masaaki Miyazawa,Hitoshi Sakaguchi +14 more
TL;DR: Results of the single test methods for the simple '2 out of 3' ITS for 213 substances have been compiled and qualitatively compared to both animal and human data, and the non-animal test methods exhibited good predictivities when compared to local lymph node assay (LLNA) data and even better predictivitiesWhen compared to human data.

Assessment of Pre- and Pro-haptens Using Nonanimal Test Methods for Skin Sensitization.
Daniel Urbisch,Matthias Becker,Naveed Honarvar,Susanne N. Kolle,Annette Mehling,Wera Teubner,Britta Wareing,Robert Landsiedel +7 more
TL;DR: This analysis shows that the combination of in chemico and in vitro test methods is suitable to identify pre-haptens and the majority of pro- haptens, exhibiting a similar sensitivity as for directly acting haptens.

TL;DR: Based on the relevance of the mechanisms and effects as well as the strengths and limitations of the experimental systems used to identify them, rules and principles are defined for deriving skin sensitization in silico assessments.

Peptide reactivity associated with skin sensitization: The QSAR Toolbox and TIMES compared to the DPRA.
Daniel Urbisch,Naveed Honarvar,Susanne N. Kolle,Annette Mehling,Tzutzuy Ramirez,Wera Teubner,Robert Landsiedel +6 more
TL;DR: Using the '2 out of 3' prediction model with in vitro data derived from LuSens and h-CLAT, the model identified 90% of human sensitizers and non-sensitizers; using in silico results (including abiotic and biotic activations) instead of DPRA results led to a comparable high predictivity.

Towards a science-based testing strategy to identify maternal thyroid hormone imbalance and neurodevelopmental effects in the progeny - part II: how can key events of relevant adverse outcome pathways be addressed in toxicological assessments?
Sue Marty,Manon Beekhuijzen,Alex Charlton,Nina Hallmark,Bethany R. Hannas,Sylvia Jacobi,Stephanie Melching-Kollmuss,Ursula G. Sauer,Larry Sheets,Volker Strauss,Daniel Urbisch,Philip A. Botham,Bennard van Ravenzwaay +12 more
TL;DR: The current understanding of thyroid-related adverse outcome pathways (AOPs) with adverse neurodevelopmental outcomes in mammals has been reviewed in this article, which served to establish if standard rodent toxic...
