A Phase I Study Evaluating the Safety, Pharmacokinetics, and Clinical Response of a Human IL-12 p40 Antibody in Subjects with Plaque Psoriasis
Catharine L. Kauffman,Nancy Aria,Eiko Toichi,Thomas S. McCormick,Kevin D. Cooper,Alice B. Gottlieb,Daniel E. Everitt,Bart Frederick,Yaowei Zhu,Martin A. Graham,Charles Pendley,Mary Ann Mascelli +11 more
TL;DR: Dose-dependent associations with both the rate and extent of clinical response were observed across the four dose groups, and significant and sustained concentration-dependent improvements in psoriatic lesions were observed in most subjects.

An Anti-IL-12p40 Antibody Down-Regulates Type 1 Cytokines, Chemokines, and IL-12/IL-23 in Psoriasis
Eiko Toichi,Gisela Torres,Thomas S. McCormick,Timothy Chang,Mary Ann Mascelli,Catharine L. Kauffman,Nancy Aria,Alice B. Gottlieb,Daniel E. Everitt,Bart Frederick,Charles Pendley,Kevin D. Cooper,Kevin D. Cooper +12 more
TL;DR: In a human Th1-mediated disease, blockade of APC cytokines by anti-IL-12p40 down-regulates expression of type 1 cytokines and chemokines that are downstream of IL- 12/IL-23, and also IL-12/ILs23 themselves, with a pattern indicative of coordinated deactivation of APCs and Th1 cells.

A phase I study assessing the safety, clinical response, and pharmacokinetics of an experimental infliximab formulation for subcutaneous or intramuscular administration in patients with rheumatoid arthritis.
Rene Westhovens,Frédéric Houssiau,Johan Joly,Daniel E. Everitt,Yaowei Zhu,Deborah Sisco,Bart van Hartingsveldt,Mary Ann Mascelli,Martin A. Graham,Patrick Durez,Esther Bouman-Thio +10 more
TL;DR: SC and IM treatment with this experimental infliximab formulation was well tolerated and was associated with a favorable ACR response.

A phase 1, double-blind, placebo-controlled study evaluating single subcutaneous administrations of a human interleukin-12/23 monoclonal antibody in subjects with plaque psoriasis
Alice B. Gottlieb,Kevin D. Cooper,Thomas S. McCormick,Eiko Toichi,Daniel E. Everitt,Bart Frederick,Yaowei Zhu,Charles Pendley,Martin A. Graham,Mary Ann Mascelli +9 more
TL;DR: A single SC administration of IL‑12/23 mAb was well tolerated and showed clinical response in subjects with moderate-to-severe psoriasis and interpretation of results is limited due to the small sample size.
A phase I trial of an interleukin-12/23 monoclonal antibody in relapsing multiple sclerosis.
Lloyd H. Kasper,Daniel E. Everitt,Thomas Leist,Kathleen A. Ryan,Mary Ann Mascelli,Kenneth P. Johnson,Aparna Raychaudhuri,Timothy Vollmer +7 more
TL;DR: Single subcutaneous administrations of IL‐12/23 mAb in this first study of relapsing MS were generally well tolerated, and safety of the agent will need to be tested in a study of longer duration and involving a larger cohort of subjects.
