The forced swim test (FST) measures coping strategy to an acute inescapable stress and thus provides unique insight into the neural limb of the stress response. Stress, particularly chronic stress, is a contributing factor to depression in humans and depression is associated with altered response to stress. In addition, drugs that are effective antidepressants in humans typically promote active coping strategy in the FST. As a consequence, passive coping in the FST has become loosely equated with depression and is often referred to as “depression-like” behavior. This terminology oversimplifies complex biology and misrepresents both the utility and limitations of the FST. The FST provides little construct- or face-validity to support an interpretation as “depression-like” behavior. Many mammals spontaneously rear on their hind legs in response to novelty. The current paper is the first review of rearing behaviour, and is intended to collate findings from different perspectives that are not usually brought together. We suggest that rearing is a useful marker of environmental novelty, that the hippocampal formation is a crucial component of the system controlling rearing in novel environments, and that rearing is one of several ethological measures that can profitably be used to assess hippocampal learning and memory. Consideration is given to the following topics: the possible functions of rearing in information-gathering and escape behaviour; the modulation of rearing by various factors such as anxiety/ fear emotionality; comparative perspectives on rearing; neuroanatomical circuits involved in rearing with particular reference to the hippocampal formation and its afferents and efferents; and the role of the hippocampal formation in uncharted and mismatch environmental novelty. The review concludes with testable predictions about rearing, environmental novelty and the hippocampus.

Effect of different doses of corticosterone on depression-like behavior and HPA axis responses to a novel stressor

This experiment examined the effect of repeated corticosterone injections on anxiety and depression-like behavior in male and female rats. Rats received either corticosterone or vehicle injections for 21 consecutive days prior to behavioral testing in the forced swim, open-field, and predator odor tests. The corticosterone injections significantly increased depression-like behavior in the forced swim test in both male and female rats but had no significant effect on anxiety in the open-field test. In the predator odor test, the corticosterone injections significantly increased a subset of defensive behaviors in the male rats. These results suggest that repeated exposure to corticosterone increases depression-like behavior, with some effects on anxiety, and that male rats may be more affected than female rats by this manipulation.
