Three-dimensional technologies in presurgical planning of bone surgeries: current evidence and future perspectives

This proof-of-concept research reports a label and bio-active free electrochemical testosterone (hormone essential regulating body function) sensing at low and in physiological range using molecularly imprinted polymers (MIPs)-based electrochemical sensing platform. State-of-the-art testosterone testing is clinical-based involving expertise, sophisticated laboratory set-up, and time-consuming testing and analysis. These consequences are limiting testosterone testing regularly suitable for an average person and a wide population. To manage these challenges, o-phenylenediamine (oPD) electropolymerized to poly(o-phenylenediamine) i.e., PoPD using cyclic voltammetry within minutes for fabricating MIP onto miniaturized screen-printed-electrode (SPE) for electrical testosterone sensing application. PoPD-MIPs/SPE sensor was responsive to varied concentrations of testosterone and exhibited a detection limit (LoD) of 1 ng/dL and a wide detection ranging from 1 to 25 ng/dL. The observed LoD correlates with the salivary testosterone of a healthy male. Therefore, PoPD-MIPs/SPE sensor, interfaced with a miniaturized potentiostat (M−P) and smartphone, has the potential of point-of-care (POC) testosterone sensing. 

Label and bio-active free electrochemical detection of testosterone hormone using MIP-based sensing platform

Testosterone is a steroid hormone that plays an indispensable role in the normal metabolism of organisms. However, exogenous testosterone, even as low as nmol L-1, will harm the human body due to accumulation. In this study, we developed an unlabeled fluorescent sensor for testosterone based on SYBR Green I. SYBR Green I is a fluorescent dye that can be embedded into the G-quadruplex of the testosterone aptamer T5. The fluorescence quenching effect is utilized to achieve quantitative detection, which occurs by the competition between testosterone and SYBR Green I for the T5 aptamer binding sites. In this work, we optimized the detection conditions to make the fluorescent sensor more sensitive and verify the specificity, linear range, and detection ability in the buffer and real water samples. The sensor's LOD and LOQ values were 0.27 nmol L-1 and 0.91 nmol L-1, respectively, while the detection range was linear from 0.91 nmol L-1 to 2000 nmol L-1. According to the results, the sensor shows high specificity and good performance even in real sample detection such as tap water and river water, providing an alternative method for the quantitative detection of testosterone in the environment, which is more convenient and efficient.

A label-free fluorescent aptamer sensor for testosterone based on SYBR Green I.

This work describes the development and evaluation of a novel electrochemical aptasensor for testosterone detection. The sensor utilizes a specifically designed DNA immobilized on a screen-printed gold electrode (SPGE) modified with a conductive hydrogel and gold nanoparticles (HG/NP) composite. The aptasensor exhibited a dose-dependent response to testosterone (0.05 to 50 ng/mL) with a detection limit of 0.14 ng/mL and a good sensitivity of 0.23 μA ng-1 mL cm-2. The sensor displayed excellent selectivity towards testosterone compared to structurally similar steroid hormones. Importantly, the incorporation of HG/NP not only improved the sensor's conductivity but also acted as an antifouling layer, minimizing signal interference from non-specific biomolecule interactions in complex biological samples like human serum. The results obtained from the aptasensor showed good correlation with a standard ELISA method, demonstrating its effectiveness in real-world scenarios.

DNA Aptamer Integrated Hydrogel Nanocomposites on Screen Printed Gold Electrodes for Point-of-Care Detection of Testosterone in Human Serum.

Nasal defects are facial defects caused by trauma, tumors, or congenital diseases that seriously damage a patient’s physical and mental health. Nasal defects, from skin defects to total nasal defects, require surgical repair and reconstruction to restore the appearance and function of the nose, which have always been challenges for rhinoplasty. The development of digital technology has increased the possibility of nasal reconstruction. Digital technology is involved in the preoperative, intraoperative, and postoperative stages of nasal construction and is of great significance in improving the effect of this surgery. This article reviews the application of major digital technologies, including three-dimensional（3D） imaging technology, computer-assisted surgical navigation, and 3D printing, in nasal reconstruction and discusses the shortcomings of the current application of digital technology.

Objective The purpose of this study is to propose a surgical plan based on augmented reality (AR) and guide template technology for restoration of nasal deformities, and evaluate its feasibility and clinical efficacy. Methods Patients were scanned with a FaceScan to obtain the three-dimensional (3D) facial model, and computed tomography was also performed to obtain the maxillofacial computed tomography images while wearing the artificial marker. The mirroring tool and database searching and matching technology were employed to restore the nasal deformities for a normal nose (preoperative planning model). The design of guide template for deciding the incision area was based on the preoperative planning model, which was also imported into the AR image guidance system named HuaxiAR1.0 for reconstruction of the nose contour. One week after the surgery, the postoperative 3D facial model was obtained. Then, the clinical efficacy was evaluated by comparing the difference between the preoperative planning and postoperative 3D facial model. Results The patients obtained satisfactory nasal shapes after surgery. Comparison of the difference between the preoperative and postoperative 3D model revealed that the maximum error was ranging from 2.24 mm to 3.10 mm with the mean error from 0.54 mm to 0.65 mm. Conclusion The combined application of AR and guide template technology provides a new approach for the treatment of nasal deformities, and has a certain significance in realizing the precise repair of other craniofacial soft tissue deformities.
