Immunology of the tonsils

Pathogenic Yersinia strains were isolated between December 1998 and April 1999 from 37 wild animals: rabbit (Lepus europeus), boar (Sus scrofa scrofa), asiatic jackal (Canis aureus), red fox (Vulpes vulpes), mouflon (Ovis musimon), european river otter (Lutra lutra), beech marten (Martes foina), polecat (Mustela putorius) and wild cat (Felis silvestris). It was established that among the wild animals Y. enterocolitica strains of serotype 0:3 predominated, accompanied by Y. pseudotuberculosis strains of serotype 0:3. In one sample from asiatic jackal and one sample from rabbit, Y. enterocolitica serotype 0:8 was isolated. Yersinia enterocolitica and Y. pseudotuberculosis strains were isolated from tonsils and tongues as well as from the viscera – lung, liver, heart, spleen, kidney and lymph nodes, mainly in young animals (1–2 years of age). The results showed that wild animals are a possible natural reservoir for pathogenic Y. enterocolitica and Y. pseudotuberculosis and are included in the epidemiological chain of yersinioses.

An experimental oral pig model was used to assess the pathogenic and immunogenic potential of Yersinia enterocolitica serotype O:8 wild-type strain 8081-L2 and its lipopolysaccharide (LPS) mutant derivatives: a spontaneous rough mutant 8081-R2, strain 8081-ΔwzzGB expressing O-antigen with uncontrolled chain lengths, and strain 8081-wbcEGB expressing semirough LPS with only one O-unit. Microbiological and immunological parameters of the infected pigs were followed from day 7 to 60 postinfection. The wild-type and all LPS mutant strains persisted in the lymphoid tissue of tonsils and small intestines, causing asymptomatic infection without any pathological changes. Although the pig is known as a reservoir of Yersiniae , a precise analysis of pathogenic and immunogenic parameters based on different in vitro tests (hematological response, killing ability of leukocytes and blood sera, antibody response, hydrogen peroxide production by macrophages, classical and alternative pathways of complement activation), revealed significant attenuation in the pathogenicity of the LPS mutant strains but not the loss of immunogenic potential. In comparison with the other strains, strain 8081-ΔwzzGB demonstrated more continuous leucocytosis with monocytosis, higher invasive potential, significant activation of hydrogen peroxide production by macrophages and an effective immunoglobulin G immune response accompanied by relevant histological immunomorphological rearrangements.

Experimental pig yersiniosis to assess attenuation of Yersinia enterocolitica O:8 mutant strains.

Yersinia enterocolitica, an enteropathogenic species belonging to the genus Yersinia, is widely distributed in nature and affects both humans and animals. In the past 10 years, this microorganism has been extensively studied in numerous publications, focusing on molecular biology, basic genetics, virulence, epidemiology and genomics. This chapter provides a comprehensive overview of all the biological and medical aspects relating to this bacterial pathogen, relating well-accepted data to recent information. Due to the versatile nature of Y. enterocolitica, a thorough knowledge of this species is not achievable and it is expected that future research will bring further enlightenment.

Chapter 73 – Yersinia enterocolitica

Summary The cellular immune response after an experimental oral infection with Yersinia enterocolitica (serotype 0:3, biotype 4, harbouring the virulence plasmid — pYV) was studied in pigs. A maximal stimulation of the T cell population in the thymus, spleen and peripheral blood was stated on the 15th day post infection (p.i.) by the rosette forming cell (RFC) test. The hemolysins (produced by B cells and detected by the plaque forming cell test — PFC) were significantly increased on the 15th da p.i. among the thymus and spleen lymphocytes and on the 25th day p.i. among the blood lymphocytes. Blood and thymus lymphocytes were activated faster by the infectious agent in comparison to the spleen cells. The electronmicroscopic studies revealed an intracellular presence of the bacteria in alveolar macrophages (aMa) and peritoneal macrophages (pMa) as well as in Peyer's patches and tonsils as early as on the 4th day p.i. Extracellularly located bacteria were observed, too. The results have shown that inspite of the activation of T and B cell immune response, this infectious agent persisted in the porcine organism.

Immunological and Electronmicroscopic Studies in Pigs Infected with Yersinia enterocolitica 0:3

The pathogenesis and the cell immune response (CIR) of guinea pigs after mixed infection with Y. enterocolitica and L. monocytogenes was investigated. The guinea pigs were infected per os with 1.1 x 10(9) CFU Y. enterocolitica 0:3, (pYV+) and four days later with 1.1 x 10(9) CFU L. monocytogenes 4B. Clinical, paraclinical and morphological findings attending the infectious process were followed in dynamics up to the 28th day post infection (p.i.) with L. monocytogenes. The phagocyting activity of alveolar macrophages (aMa) was suppressed against Y. enterocolitica, in contrast to peritoneal macrophages (pMa) engulfing yersiniae more actively at the end of the study. Moreover, the tendency of augmented entering in both phagocytes of L. monocytogenes cells was well demonstrated, starting at the earlier intervals of examination. Histopathological studies showed a purulent meningoencephalitis and a catarrhal pneumonie, non-reactive micronecroses in the spleen and lymphadenitis catarrhalis in the mesenteric lymph nodes. Analysis of the T-cell immune response (T-CIR) showed maximal values in the spleen lymphocytes after Y. enterocolitica and L. monocytogenes mixed infection. The B-CIR occurred early (at the 7th day p.i.) and was maximal at the 28th day p.i. in blood lymphocytes. The results obtained demonstrated that the mixed infection of guinea pigs with Y. enterocolitica and L. monocytogenes runs has a non lethal, generalized illness with a dominant role of L. monocytogenes cells.
