Antidiabetic plants and their active constituents.

The Department of Medicinal Chemistry and Molecular Pharmacology’s laboratory has adopted four “bench top” bioassays which do not require higher animals to screen and direct the fractionation of botanical extracts in drug discovery efforts. These are: 1. The brine shrimp lethality test (BST) (a general bioassay), 2. The inhibition of crown gall tumors on discs of potato tubers (an antitumor bioassay), 3. The inhibition of frond proliferation in duckweed (a bioassay for herbicides and plant growth stimulants), and 4. The yellow fever mosquito larvae lethality test (a bioassay for pesticides). The materials and procedures for those bioassays will be briefly described. The authors’ results in applying these simple methods in the discovery of Annonaceous acetogenins will be presented to illustrate their successful use. The BST is especially suggested as an inexpensive, simple, and rapid means of standardization of bioactivity in heterogeneous botanical products.

Structure and Biosynthesis of the Jamaicamides, New Mixed Polyketide-Peptide Neurotoxins from the Marine Cyanobacterium Lyngbya majuscula

Marine-derived fungi have been shown in recent years to produce a plethora of new bioactive secondary metabolites, some of them featuring new carbon frameworks hitherto unprecedented in nature. These compounds are of interest as new lead structures for medicine as well as for plant protection. The aim of this protocol is to give a detailed description of methods useful for the isolation and cultivation of fungi associated with various marine organisms (sponges, algae and mangrove plants) for the extraction, characterization and structure elucidation of biologically active secondary metabolites produced by these marine-derived endophytic fungi, and for the preliminary evaluation of their pharmacological properties based on rapid 'in house' screening systems. Some results exemplifying the positive outcomes of the protocol are given at the end. From sampling in marine environment to completion of the structure elucidation and bioactivity screening, a period of at least 3 months has to be scheduled.

Methods for isolation of marine-derived endophytic fungi and their bioactive secondary products

Medicinal plants constitute an important component of flora and are widely distrib- uted in India. The pharmacological evaluation of substances from plants is an established method for the identification of lead compounds which can leads to the development of novel and safe medicinal agents. Based on the ethnopharmacological literature, several species of medicinal plants used in traditional medicine in India were collected. In the present study, aqueous extracts of these medicinal plants were screened for their cytotoxicity using brine shrimp lethality test. Out of the 120 plants tested, Pistacia lentiscus exhibited potent brine shrimp lethality with LC50 2.5μg . Aristolochia indica (Aristolochiaceae), Boswellia serrata (Burseraceae), Ginkgo biloba (Ginkgoaceae), Garcinia cambogia (Clusiaceae), and Semecarpus anacardium (Anacardiaceae) have also showed significant cytotoxicity with LC50 13, 18, 21, 22, and 29.5μg respectively. The present study supports that brine shrimp bioassay is simple reliable and convenient method for assessment of bioactivity of medicinal plants and lends support for their use in traditional medi- cine.

Assessment of Bioactivity of Indian Medicinal Plants Using Brine Shrimp (Artemia salina) Lethality Assay

A method, utilizing brine shrimp (Artemia salina Leach), is proposed as a simple bioassay for natural product research. The procedure determines LC (50) values in microg/ml of active compounds and extracts in the brine medium. Activities of a broad range of known active compounds are manifested as toxicity to the shrimp. Screening results with seed extracts of 41 species of Euphorbiaceae were compared with 9KB and 9PS cytotoxicities. The method is rapid, reliable, inexpensive, and convenient as an in-house general bioassay tool.
