Bovine milk represents an essential source of nutrients for lactating calves and a key raw material for human food preparations. A wealth of data are present in the literature dealing with massive proteomic analyses of milk fractions and independent targeted studies on specific groups of proteins, such as caseins, globulins, hormones and cytokines. In this study, we merged data from previous investigations to compile an exhaustive list of 573 non-redundant annotated protein entries. This inventory was exploited for integrated in silico studies, including functional GO term enrichment (FatiGO/Babelomics), multiple pathway and network analyses. As expected, most of the milk proteins were grouped under pathways/networks/ontologies referring to nutrient transport, lipid metabolism and objectification of the immune system response. Notably enough, another functional family was observed as the most statistically significant one, which included proteins involved in the induction of cellular proliferation processes as well as in anatomical and haematological system development. Although the latter function for bovine milk proteins has long been postulated, studies reported so far mainly focused on a handful of molecules and missed the whole overview resulting from an integrated holistic analysis. A preliminary map of the bovine milk proteins interactome was also built up, which will be refined in future as result of the widespread use of quantitative methods in protein interaction studies and consequent reduction of false-positives within associated databases.

The aim of this study was to monitor the metabolic profile and oxidative status in goats during the peripartum period. A blood sample was taken from 10 Red Syrian goats on days −21, −3, +1, +14, and +28 from delivery. Samples were assayed for glutathione peroxidase and superoxide dismutase activities, and reactive oxygen metabolites, insulin, insulin-like growth factor-I, free triiodothyronine (fT3), free thyroxine (fT4), leptin, urea, non-esterified fatty acids, triglycerides, cholesterol, total protein, albumin, globulin, calcium and inorganic phosphorus concentrations. Blood glutathione peroxidase activity decreased during the postpartum period and its values were significantly (P < 0.05) lower on days 14 and 28 postpartum. Albumin levels were significantly (P < 0.05) lower on days −3, 1 and 28 from delivery compared with day −21. Plasma urea levels significantly (P < 0.001) decreased starting from day −3 from delivery. No effect of time from delivery was noted on reactive oxygen metabolites, non-esterified fatty acids, triglycerides, cholesterol, total protein, calcium and inorganic phosphorus, insulin and leptin concentrations and superoxide dismutase activity. Plasma concentrations of fT3 were significantly (P < 0.01) higher on days 14 and 28 compared with days −21, −3 and 1 from delivery. Plasma levels of fT4 were significantly (P < 0.01) lower on days −3 and 1 from delivery. Plasma insulin-like growth factor-I values slowly decreased during the postpartum period and its values were significantly (P < 0.05) lower on day 28. This study indicates that goats experienced moderate oxidative stress during the peripartum period.

The essential role of mammary fat pads in mammary growth and morphogenesis was the first indication that biologically active molecules, secreted from adipocytes or other stromal cells, could regulate endocrine cues for growth and function of the mammary gland. The presence of leptin and adiponectin receptors in mammary tissues suggested that locally produced or circulating adipokines could affect mammary growth and function. Herein, we present the current knowledge on the role of adipokines in mammary cell proliferation and differentiation and in lactogenesis and galactopoiesis in farm animals. We also address the role of milk adipokines in the neonate. Accumulating evidence suggests that adipokines could act as metabolic sensors, regulating mammary growth and function in periods of metabolic adaptations such as late pregnancy and early lactation. Indeed, different experiments reported that adiponectin and leptin expression varies according to physiological stages and nutritional status of the animal. The current review also demonstrates that adipokines, such as leptin and adiponectin, are important regulators of the action of lactogenic hormones in the mammary gland. Findings also suggest important roles for adipokines in growth and intestinal maturation of the neonate.

Leptin gene is expressed in the mammary tissue and the expression of both leptin and its receptor changes significantly during pregnancy and lactation, with high levels during the first half of pregnancy and a decrease at delivery. The aim of this work was to investigate into leptin concentration in plasma and in milk during pregnancy and the first week after parturition in dairy cow and to analyze the correlation between leptin and beta-lactoglobulin (beta-LG) concentrations in plasma and in milk. The trial was conducted on six Holstein dairy cows, reared in the same environmental conditions and evaluated with similar body condition score, during the complete reproductive cycle from insemination to the delivery. Blood from the jugular vein and milk samples were collected at weekly intervals. Plasma leptin concentration showed a lower level (p < 0.05) at the beginning of pregnancy. Milk leptin concentration showed a higher level (p < 0.01) than plasma level from week 23 to week 29 of pregnancy. Plasma beta-LG concentrations were higher (p < 0.01) compared to plasma concentrations during the first part of pregnancy, then milk levels rise and become higher than plasma levels during the last weeks before dry period. A positive correlation (p < 0.01) was observed between leptin and beta-LG both in plasma and in milk profiles.
