High-throughput PRIME-editing screens identify functional DNA variants in the human genome.
Xingjie Ren,Han Yang,Jovia L. Nierenberg,Yifan Sun,Jiawen Chen,Cooper Beaman,Thu Pham,Mai Nobuhara,Maya Asami Takagi,Vivek Narayan,Yun Li,Elad Ziv +11 more
TL;DR: A pooled prime-editing screen method that can be applied to characterize thousands of coding and non-coding variants in a single experiment with high reproducibility is developed, demonstrating that PRIME is capable of characterizing genetic variants at single-base resolution and scale.

High throughput PRIME editing screens identify functional DNA variants in the human genome
Xingjie Ren,Jovia L. Nierenberg,Yifan Sun,Jiawen Chen,Cooper Beaman,Mai Nobuhara,Maya Asami Takagi,Yun Li,Elad Ziv,Yin Shen +9 more
TL;DR: In this paper , a pooled prime editing screen method, called PRIME, was developed to characterize thousands of coding and non-coding variants in a single experiment with high reproducibility.

Functional characterization of gene regulatory elements and neuropsychiatric disease-associated risk loci in iPSCs and iPSC-derived neurons
Xiaoyu Yang,Ian Jones,Poshen B. Chen,Xingjie Ren,Lina Zheng,Bin Li,Yang Eric Li,Quan Sun,Jia Wen,Cooper Beaman,Xiekui Cui,Yun Li,Wei Wang,Min Hu,Bing Ren,Yin Shen +15 more
TL;DR: Through genome-scale CRISPR screens of 22,000 candidate CREs (cCREs) in human induced pluripotent stem cells undergoing differentiation to excitatory neurons, 2,847 and 5,540 fCREs essential for iPSC fitness and neuronal differentiation are identified.

Syk inhibition reprograms tumor-associated macrophages and overcomes gemcitabine-induced immunosuppression in pancreatic ductal adenocarcinoma.
Deepak Rohila,In H. Park,Timothy V. Pham,Jonathan Weitz,Tatiana Hurtado de Mendoza,Suresh Madheswaran,Mehreen Ishfaq,Cooper Beaman,Elisabet Navarro Tapia,Siming Sun,Jay Patel,Pablo Tamayo,Andrew M. Lowy,Shweta Joshi +13 more
TL;DR: In this article , macrophage spleen tyrosine kinase (Syk) was shown to promote PDAC growth and metastasis in orthotopic PDAC mouse models, genetic deletion of myeloid Syk reprogrammed macrophages into immunostimulatory phenotype, increased the infiltration, proliferation, and cytotoxicity of CD8+ T cells, and repressed PDAC development.
