SET8 prevents excessive DNA methylation by methylation-mediated degradation of UHRF1 and DNMT1.

TL;DR: It is shown that SET8, a cell-cycle-regulated protein methyltransferase, controls protein stability of both UHRF1 and DNMT1 through methylation-mediated, ubiquitin-dependent degradation and consequently prevents excessive DNA methylation.
Profiling of epigenetic DNA modifications by advanced liquid chromatography-mass spectrometry technologies
TL;DR: This work provided comprehensive review on LC-MS-based technologies and related pretreatment technologies for profiling of epigenetic DNA modifications, mainly focusing on innovative technologies developed for desalting, DNA digestion, and ionization for mass spectrometry detection.

Vertical Ultrafiltration-Facilitated DNA Digestion for Rapid and Sensitive UHPLC-MS/MS Detection of DNA Modifications.
TL;DR: Here, it is observed that inorganic cations Na+ and K+ and phosphate buffers significantly inhibited DNA digestion as catalyzed by typical set of DNase I, snake venom phosphodiesterase, and calf intestine alkaline phosphatase, leading to poor or varying performance on UHPLC-MS/MS analysis.

UHPLC-Q-TOF/MS detection of UV-induced TpT dimeric lesions in genomic DNA.
TL;DR: It is demonstrated that only are TpT dimeric lesions detectable in genomic DNA of HCT116 cells upon UVC irradiation, and the proposed UHPLC-Q-TOF/MS method is promising for accurate identification and quantitative detection of UV-induceddimeric lesions in cellular DNA.

USP7 negatively controls global DNA methylation by attenuating ubiquitinated histone-dependent DNMT1 recruitment.
Jialun Li,Ruiping Wang,Jianyu Jin,Mengmeng Han,Zhaosu Chen,Yingying Gao,Xueli Hu,Haijun Zhu,Huifang Gao,Kongbin Lu,Yanjiao Shao,Cong Lyu,Weiyi Lai,Pishun Li,Guang Hu,Jiwen Li,Dali Li,Hailin Wang,Qihan Wu,Jiemin Wong +19 more
TL;DR: Evidence is presented that UHRF1 and USP7 oppositely control ubiquitination of histones H3 and H2B in S phase of the cell cycle and that DNMT1 binds both ubiquitinated H 3 and H 2B and that USp7 suppresses DNMT 1 recruitment and DNA methylation through its deubiquitinase activity and the interaction withDNMT1.
