Ascorbic acid plays an essential role in human health and its deficiency severely impacts physical well-being. Fruits are a good dietary source of ascorbic acid and thereby, its composition and strategies to increase total vitamin C contents in fruits have been reviewed previously. Nevertheless, ascorbic acid is also a fundamental antioxidant that regulates fruit ripening by its efficiency to eliminate reactive oxygen species and modulate the cellular redox state. The present review summarizes current knowledge on ascorbic acid from a plant physiological perspective both during preharvest and postharvest to dissect key regulatory points and offers an updated discussion on its capacity to regulate fruit ripening. At the same time, ascorbic acid capacity to modulate ethylene and abscisic acid signaling is discussed, together with its relevance to control the cellular redox state thereby fine-tuning fruit ripening onset and postharvest shelf-life. Finally, we emphasize the relevance of ascorbic acid to regulate fruit physiological disorders during postharvest and its relationship with current technologies of postharvest management. 

Ascorbic acid as a master redox regulator of fruit ripening

The color changes brought on by the enzymatic interactions of phenolic compounds with released endogenous polyphenol oxidase and the penetration of oxygen into the tissue has a significant impact on the commercialization of fresh-cut fruit, such as apples. This process causes a loss of quality in fresh-cut apples, resulting in browning of the fruit surface. By acting as a semipermeable barrier to gases and water vapor and thus lowering respiration, enzymatic browning, and water loss, edible coatings can provide a chance to increase the shelf life of fresh-cut produce. In this study, the effect of edible coatings composed of carboxymethylcellulose (CMC, 1%), sodium alginate (SA, 1%), citric acid (CA, 1%), and oxalic acid (OA, 0.5%) on fresh-cut ‘Annurca Rossa del Sud’ apple was studied. Four formulations of edible coatings, A. SA+CMC, B. SA+CMC+CA, C. SA+CMC+OA, and D. SA+CMC+CA+OA, were tested. Fresh-cut apples were dipped into different solutions and then stored at 4 °C, and physicochemical and biochemical analyses were performed at 0, 4, 8, and 12 days of storage. Results demonstrated that all four combinations improved the shelf-life of fresh-cut apple by slowing down the qualitative postharvest decay, total soluble solid, and titratable acidity. The browning index was highest in the control samples (82%), followed by CMC+SA (53%), CMC+SA+CA (32%), CMC+SA+OA (22%), and finally CMC+SA+CA+OA (7%) after 12 days of cold storage. Furthermore, coating application increased the bioactive compound content and antioxidant enzyme activities. Furthermore, the synergistic activity of SA+CMC+CA+OA reduces enzymatic browning, prolonging the postharvest life of minimally processed ‘Annurca Rossa del Sud’ apples.

Comparative analysis of husk microstructure, fruit quality and concentrations of bioactive compounds of different pomegranate cultivars during low temperature storage

The aim of this experiment was to assess the effect of different storage temperatures on the texture quality, phenolic profile, and antioxidant capacity of a grape. Fresh grapes were stored at 4 and 25 °C for nine days and sampled on alternate days. The hardness, total phenolics, total flavanones, total flavanols, total anthocyanin content, antioxidant activity, differential metabolite screening, and key gene expression were evaluated. In addition, four phenolic compounds were screened out as differential metabolites in response to storage temperature by OPLS-DA analysis. The results showed that the fruit firmness was better maintained in low-temperature storage and the storage life was longer than that at 25 °C. During the whole storage process, the contents of phenolics, flavanones, flavanols, and anthocyanins all showed an increasing trend first and then decreased regardless of what temperature. Since the antioxidant capacity of a grape was positively correlated with the contents of total phenols and total flavonoids, the same trend was also shown. However, the grape's phenolic compound content and antioxidant activity were higher at 25 °C than at 4 °C. Furthermore, through qualitative and quantitative analysis of 16 monomeric phenols, this study selected catechin, 1-O-vanilloyl-β-d-glucose, p-coumaric acid 4-glucoside, and resveratrol-3-O-glucoside as the main differentially expressed metabolites at the two temperatures. In conclusion, for a short shelf life or immediate consumption, keeping grapes at room temperature is more beneficial to obtain high antioxidants. However, if the goal is to prolong the storage period of the fruit, keeping the fruit at 4 °C is recommended.

Changes in Polyphenolic Compounds of Hutai No. 8 Grapes during Low-Temperature Storage and Their Shelf-Life Prediction by Identifying Biomarkers.

The reduction in shelf-life and nutritional value of cauliflower florets are the most vital problems during cold storage. This research was performed to explore the impact of the pre-harvest foliar implementation of proline (PR), methionine (MT), and melatonin (ML), at a rate of 25 mg.L−1, on shelf-life and active composites of cauliflower florets (cv. ‘Arasya’) stored at 10 °C for 14 d. The obtained results exhibited that florets from treated plants with PR, MT, and ML were lower in water loss, electrolyte leakage, browning index, and titratable acidity (TA) compared with untreated plants, at the end of storage. Furthermore, these pre-harvest treatments significantly slow down the loss of total sugar content, reducing sugar, total soluble solid (TSS), glucosinolates concentration (Gly), total phenols (TP), vitamin C (VC), and antioxidant activity (DPPH) than control samples. After 14 days of storage, the application of PR, MT, and ML significantly improved the activity levels of phenylalanine ammonia-lyase (PAL), and catalase (CAT) as well as declined the polyphenol oxidase (PPO), peroxidase (POD) activates in the florets during the cold storage. Therefore, the pre-harvest application of PR, MT, and ML might be promising substances to keep the nutritional quality of cauliflower forests and reduce the browning index during cold storage.

Pericarp browning occurs widely in postharvest pomegranate fruit, but little is known about its mechanism. In this study, ‘Baiyushizi’, a browning-sensitive cultivar, was used to investigate the physiological and genetic changes during pericarp browning. The pericarp browning index (BI) increased greatly after 3 d storage at room temperature, and, consequently, the fruit appearance became unattractive after 6 d. The increasing concentration of malondialdehyde (MDA) and electrolyte leakage were detected in browning pericarp. Polyphenol oxidase (PPO) activity increased significantly after 3 d, while ascorbate peroxidase (APX) and catalase (CAT) activity decreased steadily during storage. Total phenolics content decreased gradually during storage, while ascorbic acid (AsA) and glutathione (GSH) contents increased firstly, then declined. Differentially expressed genes (DEGs) of pericarp under different browning degrees were identified in the metabolic processes of phenolic compounds, lipids, ascorbic acid, glutathione, sugar, starch, energy-related, ethylene biosynthesis and signal transduction pathways. Accordingly, the potential metabolic pathways and related processes involved in pericarp browning were described. The results not only provided insights on the genetic mechanism of pericarp browning in pomegranate but also guidance on controlling postharvest fruit browning in general.

Potential Metabolic Pathways and Related Processes Involved in Pericarp Browning for Postharvest Pomegranate Fruits

The ETHYLENE INSENSITIVE3-LIKE (EIL) family is one of the most important transcription factor (TF) families in plants and is involved in diverse plant physiological and biochemical processes. In this study, ten EIL transcription factors (CsEILs) in sweet orange were systematically characterized via whole-genome analysis. The CsEIL genes were unevenly distributed across the four sweet orange chromosomes. Putative cis-acting regulatory elements (CREs) associated with CsEIL were found to be involved in plant development, as well as responses to biotic and abiotic stress. Notably, quantitative reverse transcription polymerase chain reaction (qRT-PCR) revealed that CsEIL genes were widely expressed in different organs of sweet orange and responded to both high and low temperature, NaCl treatment, and to ethylene-dependent induction of transcription, while eight additionally responded to Xanthomonas citri pv. Citri (Xcc) infection, which causes citrus canker. Among these, CsEIL2, CsEIL5 and CsEIL10 showed pronounced upregulation. Moreover, nine genes exhibited differential expression in response to Candidatus Liberibacter asiaticus (CLas) infection, which causes Citrus Huanglongbing (HLB). The genome-wide characterization and expression profile analysis of CsEIL genes provide insights into the potential functions of the CsEIL family in disease resistance.
