Abstract Biodegradation is one of the most economical methods and usually no production of toxic by‐products. In general, this approach is favored because it offers reduced expenses and the potential for complete mineralization. In order to enhance the viability and longevity of the bioremediation agents within polluted areas, it becomes necessary to immobilize the cells. Cell immobilization refers to the procedure of confining intact cells to specific areas within a device or material, without compromising their essential biological functions. A wide variety of carriers and approaches have been used for the restriction of various cells. Immobilization techniques, such as microencapsulation, have opened up new possibilities in biotechnology by facilitating the development of artificial organs, cell therapies, and drug delivery systems. Researchers have found promising outcomes in various applications through the immobilization of microorganisms. This approach enhances stability, reusability, and catalytic efficiency, making immobilization a valuable strategy for biocatalysis, bioremediation, and other biotechnological processes. Notably, the use of immobilized microorganisms has led to significant improvements in the removal of pollutants, with some studies achieving 100% efficiency. When comparing the degradation of pollutants between free and immobilized microorganisms over the same time period, the results demonstrated that immobilized microorganisms achieved a removal efficiency of more than 21% than the free microbial consortium. The primary objective of this review is to give an overview of the key scientific aspects related to bioremediation of various pollutants using immobilized cell, with a particular focus on the techniques to entrap the cells. Immobilization: the promising technique to protect and increase the efficiency of microorganisms to remove contaminants

Biotechnological processes are essential for developing economies that aim to stand out in future markets. The use of bioreactors is one of the most important unit operations of biotechnological processes, and real-time monitoring of bioreactors is essential to ensure precise bioprocess control. This review presents different types of bioreactors, sensors, and applications in other sectors. Bioreactors, controlled systems for cultivating microorganisms and cells, are essential tools in various fields, from scientific research to industrial production. The use of a variety of sensors is critical for accurate, real-time monitoring, early problem detection, reproducibility, cost reduction, and increased efficiency. These benefits are being realized in numerous applications, including biofuel production, bioremediation and leaching processes, tissue engineering, and drug manufacturing. Innovations in bioreactor technology are expanding opportunities for a more sustainable and healthier future. By developing new types of bioreactors, integrating advanced sensors, and exploring promising applications, bioreactors are playing a key role in addressing global challenges and sustainably advancing science and technology. 

Bioreactors: Applications and Innovations for a Sustainable and Healthy Future—A Critical Review

The necessity to improve in vitro cell screening assays is becoming ever more important. Pharmaceutical companies, research laboratories and hospitals require technologies that help to speed up conventional screening and therapeutic procedures to produce more data in a short time in a realistic and reliable manner. The design of new solutions for test biomaterials and active molecules is one of the urgent problems of preclinical screening and the limited correlation between in vitro and in vivo data remains one of the major issues. The establishment of the most suitable in vitro model provides reduction in times, costs and, last but not least, in the number of animal experiments as recommended by the 3Rs (replace, reduce, refine) ethical guiding principles for testing involving animals. Although two-dimensional (2D) traditional cell screening assays are generally cheap and practical to manage, they have strong limitations, as cells, within the transition from the three-dimensional (3D) in vivo to the 2D in vitro growth conditions, do not properly mimic the real morphologies and physiology of their native tissues. In the study of human pathologies, especially, animal experiments provide data closer to what happens in the target organ or apparatus, but they imply slow and costly procedures and they generally do not fully accomplish the 3Rs recommendations, i.e., the amount of laboratory animals and the stress that they undergo must be minimized. Microfluidic devices seem to offer different advantages in relation to the mentioned issues. This review aims to describe the critical issues connected with the conventional cells culture and screening procedures, showing what happens in the in vivo physiological micro and nano environment also from a physical point of view. During the discussion, some microfluidic tools and their components are described to explain how these devices can circumvent the actual limitations described in the introduction.

Repairing and regenerating damaged tissues or organs and restoring their functioning has been the ultimate aim of medical innovations. Reviving healthcare blends tissue engineering with alternative techniques, particularly hydrogels, which have emerged as vital tools in modern medicine. Additive manufacturing (AM) is a practical manufacturing revolution that uses building strategies like molding as a viable solution for precise hydrogel manufacturing. Recent advances in this technology have led to the successful manufacturing of hydrogels with enhanced reproducibility, accuracy, precision, and ease of fabrication. Hydrogels continue to metamorphose as the vital compatible bio-ink matrix for AM. AM hydrogels have paved the way for complex 3D/4D hydrogels that can be loaded with drugs or cells. Bio-mimicking 3D cell cultures designed via hydrogel-based AM is a groundbreaking in-vivo assessment tool in biomedical trials. This brief review focuses on preparations and applications of additively manufactured hydrogels in the biomedical spectrum, such as targeted drug delivery, 3D-cell culture, numerous regenerative strategies, biosensing, bioprinting, and cancer therapies. Prevalent AM techniques like extrusion, inkjet, digital light processing, and stereo-lithography have been explored with their setup and methodology to yield functional hydrogels. The perspectives of limitations and the possible prospects of AM hydrogels have been critically examined in this study. 

Innovative technologies on the fabrication of 3D/4D smart hydrogels and its biomedical applications - A compendious review

In this second part of our systematic review on the research area of 3D cell culture in micro-bioreactors we give a detailed description of the published work with regard to the existing micro-bioreactor types and their applications, and highlight important results gathered with the respective systems. As an interesting detail, we found that micro-bioreactors have already been used in SARS-CoV research prior to the SARS-CoV2 pandemic. As our literature research revealed a variety of 3D cell culture configurations in the examined bioreactor systems, we defined in review part one “complexity levels” by means of the corresponding 3D cell culture techniques applied in the systems. The definition of the complexity is thereby based on the knowledge that the spatial distribution of cell-extracellular matrix interactions and the spatial distribution of homologous and heterologous cell–cell contacts play an important role in modulating cell functions. Because at least one of these parameters can be assigned to the 3D cell culture techniques discussed in the present review, we structured the studies according to the complexity levels applied in the MBR systems.

Advanced 3d cell culture techniques in micro-bioreactors, part ii: Systems and applications

Bioreactors have proven useful for a vast amount of applications. Besides classical large-scale bioreactors and fermenters for prokaryotic and eukaryotic organisms, micro-bioreactors, as specialized bioreactor systems, have become an invaluable tool for mammalian 3D cell cultures. In this systematic review we analyze the literature in the field of eukaryotic 3D cell culture in micro-bioreactors within the last 20 years. For this, we define complexity levels with regard to the cellular 3D microenvironment concerning cell–matrix-contact, cell–cell-contact and the number of different cell types present at the same time. Moreover, we examine the data with regard to the micro-bioreactor design including mode of cell stimulation/nutrient supply and materials used for the micro-bioreactors, the corresponding 3D cell culture techniques and the related cellular microenvironment, the cell types and in vitro models used. As a data source we used the National Library of Medicine and analyzed the studies published from 2000 to 2020.
