The current trajectory for crop yields is insufficient to nourish the world’s population by 20501. Greater and more consistent crop production must be achieved against a backdrop of climatic stress that limits yields, owing to shifts in pests and pathogens, precipitation, heat-waves and other weather extremes. Here we consider the potential of plant sciences to address post-Green Revolution challenges in agriculture and explore emerging strategies for enhancing sustainable crop production and resilience in a changing climate. Accelerated crop improvement must leverage naturally evolved traits and transformative engineering driven by mechanistic understanding, to yield the resilient production systems that are needed to ensure future harvests. Genetic strategies for improving the yield and sustainability of agricultural crops, and the resilience of crops in the face of biotic and abiotic stresses contingent on projected climate change, are evaluated.

Genetic strategies for improving crop yields

Water use efficiency (WUE) is defined as the amount of carbon assimilated as biomass or grain produced per unit of water used by the crop. One of the primary questions being asked is how plants will respond to a changing climate with changes in temperature, precipitation, and carbon dioxide (CO2) that affect their WUE At the leaf level, increasing CO2 increases WUE until the leaf is exposed to temperatures exceeded the optimum for growth (i.e., heat stress) and then WUE begins to decline. Leaves subjected to water deficits (i.e., drought stress) show varying responses in WUE. The response of WUE at the leaf level is directly related to the physiological processes controlling the gradients of CO2 and H2O, e.g., leaf:air vapor pressure deficits, between the leaf and air surrounding the leaf. There a variety of methods available to screen genetic material for enhanced WUE under scenarios of climate change. When we extend from the leaf to the canopy, then the dynamics of crop water use and biomass accumulation have to consider soil water evaporation rate, transpiration from the leaves, and the growth pattern of the crop. Enhancing WUE at the canopy level can be achieved by adopting practices that reduce the soil water evaporation component and divert more water into transpiration which can be through crop residue management, mulching, row spacing, and irrigation. Climate change will affect plant growth, but we have opportunities to enhance WUE through crop selection and cultural practices to offset the impact of a changing climate.

Water-Use Efficiency: Advances and Challenges in a Changing Climate

With the global climate anomalies and the destruction of ecological balance, the water shortage has become a serious ecological problem facing all mankind, and drought has become a key factor restricting the development of agricultural production. Therefore, it is essential to study the drought tolerance of crops. Based on previous studies, we reviewed the effects of drought stress on plant morphology and physiology, including the changes of external morphology and internal structure of root, stem, and leaf, the effects of drought stress on osmotic regulation substances, drought-induced proteins, and active oxygen metabolism of plants. In this paper, the main drought stress signals and signal transduction pathways in plants are described, and the functional genes and regulatory genes related to drought stress are listed, respectively. We summarize the above aspects to provide valuable background knowledge and theoretical basis for future agriculture, forestry breeding, and cultivation.

Response Mechanism of Plants to Drought Stress

Global warming and associated precipitation changes will negatively impact on many agricultural ecosystems. Major food production areas are expected to experience reduced water availability and increased frequency of drought over the coming decades. In affected areas, this is expected to reduce the production of important food crops including wheat, rice, and maize. The development of crop varieties able to sustain or improve yields with less water input is, therefore, a priority for crop research. Almost all water used for plant growth is lost to the atmosphere by transpiration through stomatal pores on the leaf epidermis. By altering stomatal pore apertures, plants are able to optimize their CO2 uptake for photosynthesis while minimizing water loss. Over longer periods, stomatal development may also be adjusted, with stomatal size and density being adapted to suit the prevailing conditions. Several approaches to improve drought tolerance and water-use efficiency through the modification of stomatal traits have been tested in the model plant Arabidopsis thaliana. However, there is surprisingly little known about the stomata of crop species. Here, we review the current understanding of how stomatal number and morphology are involved in regulating water-use efficiency. Moreover, we discuss the potential and limitations of manipulating stomatal development to increase drought tolerance and to reduce water loss in crops as the climate changes.

Impact of Stomatal Density and Morphology on Water-Use Efficiency in a Changing World.

Much of humanity relies on rice (Oryza sativa) as a food source, but cultivation is water intensive and the crop is vulnerable to drought and high temperatures. Under climate change, periods of reduced water availability and high temperature are expected to become more frequent, leading to detrimental effects on rice yields.
We engineered the high-yielding rice cultivar ‘IR64’ to produce fewer stomata by manipulating the level of a developmental signal. We overexpressed the rice epidermal patterning factor OsEPF1, creating plants with substantially reduced stomatal density and correspondingly low stomatal conductance.
Low stomatal density rice lines were more able to conserve water, using c. 60% of the normal amount between weeks 4 and 5 post germination. When grown at elevated atmospheric CO2, rice plants with low stomatal density were able to maintain their stomatal conductance and survive drought and high temperature (40°C) for longer than control plants. Low stomatal density rice gave equivalent or even improved yields, despite a reduced rate of photosynthesis in some conditions.
Rice plants with fewer stomata are drought tolerant and more conservative in their water use, and they should perform better in the future when climate change is expected to threaten food security.

The epidermal patterning factor (EPF) family of secreted signaling peptides regulate the frequency of stomatal development in model dicot and basal land plant species. Here, we identify and manipulate the expression of a barley (Hordeum vulgare) ortholog and demonstrate that when overexpressed HvEPF1 limits entry to, and progression through, the stomatal development pathway. Despite substantial reductions in leaf gas exchange, barley plants with significantly reduced stomatal density show no reductions in grain yield. In addition, HvEPF1OE barley lines exhibit significantly enhanced water use efficiency, drought tolerance, and soil water conservation properties. Our results demonstrate the potential of manipulating stomatal frequency for the protection and optimization of cereal crop yields under future drier environments.

Reducing stomatal density in barley improves drought tolerance without impacting on yield.

Many plant pathogens gain entry to their host via stomata. On sensing attack, plants close these pores to restrict pathogen entry. Here, we show that plants exhibit a second longer term stomatal response to pathogens. Following infection, the subsequent development of leaves is altered via a systemic signal. This reduces the density of stomata formed, thus providing fewer entry points for pathogens on new leaves. Arabidopsis thaliana leaves produced after infection by a bacterial pathogen that infects through the stomata (Pseudomonas syringae) developed larger epidermal pavement cells and stomata and consequently had up to 20% reductions in stomatal density. The bacterial peptide flg22 or the phytohormone salicylic acid induced similar systemic reductions in stomatal density suggesting that they might mediate this effect. In addition, flagellin receptors, salicylic acid accumulation, and the lipid transfer protein AZI1 were all required for this developmental response. Furthermore, manipulation of stomatal density affected the level of bacterial colonization, and plants with reduced stomatal density showed slower disease progression. We propose that following infection, development of new leaves is altered by a signalling pathway with some commonalities to systemic acquired resistance. This acts to reduce the potential for future infection by providing fewer stomatal openings.
