Diabetes-specific microvascular disease is a leading cause of blindness, renal failure and nerve damage, and diabetes-accelerated atherosclerosis leads to increased risk of myocardial infarction, stroke and limb amputation. Four main molecular mechanisms have been implicated in glucose-mediated vascular damage. All seem to reflect a single hyperglycaemia-induced process of overproduction of superoxide by the mitochondrial electron-transport chain. This integrating paradigm provides a new conceptual framework for future research and drug discovery.

Biochemistry and molecular cell biology of diabetic complications

▪ Abstract The innate immune system is a universal and ancient form of host defense against infection. Innate immune recognition relies on a limited number of germline-encoded receptors. These receptors evolved to recognize conserved products of microbial metabolism produced by microbial pathogens, but not by the host. Recognition of these molecular structures allows the immune system to distinguish infectious nonself from noninfectious self. Toll-like receptors play a major role in pathogen recognition and initiation of inflammatory and immune responses. Stimulation of Toll-like receptors by microbial products leads to the activation of signaling pathways that result in the induction of antimicrobial genes and inflammatory cytokines. In addition, stimulation of Toll-like receptors triggers dendritic cell maturation and results in the induction of costimulatory molecules and increased antigen-presenting capacity. Summary Two Salmonella S-forms and two R-mutants were cultivated in complex medium supplemented with different amounts (0.5-3%) glucose. Cultivation was performed batchwise in a fermentor under aerobic conditions. With all strains investigated, the yield of bacterial mass increased with increasing concentration of glucose. In the case of three strains, the LPS content of bacteria also increased, thus achieving the aim of this investigation. The synthesis of bacterial mass and LPS did not proceed in parallel and differed from strain to strain. At optimal glucose concentration, the yield of LPS could be increased up to 250%. The chemical composition of the LPS was independent of the glucose concentration. The individual strains exhibited an identical composition with regard to lipid A and polysaccharide when cultured at different glucose concentrations. The uniformity of the molecular distribution of LPS could also be confirmed by SDS-polyacrylamide gel electrophoresis. In the S-form LPSs, also the proportion of the unsubstituted R-form LPS was not affected by the glucose concentration in the culture medium. The present results demonstrate that optimisation of the cultivation conditions with respect to the glucose concentration of the medium would be of advantage especially for Salmonella strains that are cultivated frequently.

Influence of the glucose concentration on the yield of biomass and lipopolysaccharide in Salmonella cultures.

The object of the present investigation was to extend fundamental understanding of the composition, function and origin of bronchoalveolar lavage (BAL) cells, and to reduce the considerable gaps in our existing knowledge. BAL cells from the B10.D2 mouse were identified by morphological, enzyme cytochemical and immunocytochemical methods and a further functional characterisation of macrophages was undertaken in vitro using latex phagocytosis. The possible bone marrow origin of BAL cells was investigated with a radiation chimera. The bone marrow from H2k-positive F1 mice (B10.BR×B10.D2) was transplanted into the irradiated H2k-negative parent animals. At various time intervals after transplantation, BAL cells were tested immunocytochemically for the presence of H2k-positive cells. The principal finding was that BAL cells are functionally heterogeneous. It was possible by morphological, enzyme cytochemical and immunocytochemical techniques to subdivide the total BAL cell count into 89% macrophages, 8.5% lymphocytes and 2.5% granulocytes. Only 14.3% of the macrophages expressed the corresponding epitope, and this small population could be further subdivided immunocytochemically into mature and immature (7∶1) macrophages. The ability to phagocytose, which is typical of active macrophages, could be induced in vitro in only one-third of all BAL macrophages. Three-quarters of the macrophages carried the H2d antigen and 9.5% of the BAL cells were natural killer cells showing a macrophage phenotype. Subdivision of the lymphocytes showed a significant majority of T-cells (75%) to B-cells (20%), and 5% Asialo GM1-positive cells. The CD4/CD8 ratio amounted to 1.3∶1 and 38% of the lymphocytes expressed on the surface the H2d antigen. Throughout the experiment with the chimeras (6 to 52 weeks), H2k-positive BAL cells could be detected. These observations verify the participation of the bone marrow in the replacement of available intraalveolar cells. These findings show that the lung, as indicated by BAL cells, has at its disposal a highly developed mobile cellular defence mechanism, of which the numerically dominant component is the macrophage population. These cells should be regarded as possessing various as yet largely unknown functions.

Sera of mice sensitized with bacteria and subsequently challenged with lipopolysaccha- ride promote hemorrhagic necrosis of tumors in vivo and display cytotoxic activity against tumor cells in vitro, which has been attributed to the induction of tumor necrosis factor (TNF). Here, we describe the induction of a previously unrecog- nized antitumor activity in such sera, which is dis- tinct from TNF but displays tumor-specific cyto- cidal activity in vitro as well as potent tumor-re- gressing activity in vivo. Biochemical analysis of this activity yielded a molecular mass of 150 kDa, closely resembling a novel tumoricidal factor of murine macrophages (M) termed MTC 170 (M tumor cytotoxin, approximate molecular mass 170 kDa), which we have previously pro- posed to constitute a major effector pathway for the destruction of tumor cells by activated M.
