SLC27A5 deficiency activates NRF2/TXNRD1 pathway by increased lipid peroxidation in HCC.
Qingzhu Gao,Guiji Zhang,Yaqiu Zheng,Yi Yang,Chang Chen,Jie Xia,Li Liang,Chong Lei,Yuan Hu,Xue-Fei Cai,Wenlu Zhang,Hua Tang,Yaxi Chen,Ailong Huang,Kai Wang,Ni Tang +15 more
TL;DR: It is demonstrated that SLC27A5 acts as a novel tumor suppressor by suppressing TXNRD1 expression via the KEAP1/NRF2 pathway in HCC and combination therapy of sorafenib and NRF2/TXNRD 1 inhibitors may be a promising strategy in personalized HCC treatment.
GSTZ1 deficiency promotes hepatocellular carcinoma proliferation via activation of the KEAP1/NRF2 pathway
Jingjing Li,Jingjing Li,Qiujie Wang,Yi Yang,Chong Lei,Fan Yang,Li Liang,Chang Chen,Jie Xia,Kai Wang,Ni Tang +10 more
TL;DR: GSTZ1 serves as a tumor suppressor in HCC and the antioxidant agent N-acetylcysteine and NRF2 inhibitor brusatol effectively suppressed the growth of GstZ1-knockout HepG2 cells and HCC progression in Gstz1−/− mice.
GSTZ1-1 Deficiency Activates NRF2/IGF1R Axis in HCC via Accumulation of Oncometabolite Succinylacetone.
Fan Yang,Fan Yang,Jingjing Li,Haijun Deng,Yihao Wang,Chong Lei,Qiujie Wang,Jin Xiang,Li Liang,Jie Xia,Xuanming Pan,Xiaosong Li,Quanxin Long,Lei Chang,Ping Xu,Ailong Huang,Kai Wang,Ni Tang +17 more
TL;DR: It is revealed that GSTZ1‐1, the enzyme in phenylalanine/tyrosine catabolism, is downregulated in HCC, and its expression was negatively correlated with IGF1R, establishing succinylacetone as an oncometabolite, and GSTZ 1‐1 as an important tumor suppressor by inhibiting NRF2/IGF1R axis in H CC.

Dysregulated glucuronic acid metabolism exacerbates hepatocellular carcinoma progression and metastasis through the TGFβ signalling pathway
Qingzhu Gao,Bin Chen,Changchun Chen,Chong Lei,Xue Lin,Dan Nie,Jingjing Li,Lu-Yi Huang,Xiaosong Li,Kai Wang,Ailong Huang,Ni Tang +11 more
TL;DR: GSTZ1 deficiency and subsequent up‐regulation of the glucuronic acid metabolic pathway promotes HCC metastasis by increasing the stability of TGFβR1 mRNA and activating TGF β/Smad signalling and targeting UGDH might be a promising strategy for HCC therapy.

GSTZ1-1 downregulates Wnt/β-catenin signalling in hepatocellular carcinoma cells.
TL;DR: Results indicate that GSTZ1‐1 may play a tumour suppressor role by inactivating the Wnt/β‐catenin signalling pathway and provide a comprehensive overview of the transcriptome in GSTZ 1‐1‐overexpressing Huh7 cells.
