DNA base composition was determined by reversed-phase high-performance liquid chromatography (HPLC). DNA was hydrolysed into nucleosides with nuclease P1 and bacterial alkaline phosphatase. The mixture of nucleosides was applied to HPLC without any further purification. One determination by chromatography needed 2 μg of hydrolysed nucleosides and took only 8 min. The relative standard error of nucleoside analysis was less than 1%. The system described here gives a direct and precise method for determining DNA base composition.

In this review paper, several aspects of fruit juice microbiology, from past to future perspectives, are considered. An overview of the most relevant outbreaks involving foodborne pathogens and spoilage microorganisms associated with fruit juices is provided. One of the sections provides data on the sources of fruit juice contamination, followed by perspectives on preservation methods. Furthermore, considerations on the role of international guidelines about exotic fruit juices in respect to public health, and of the microbiological status of fruit juices used as food/beverage ingredients are discussed. Issues and challenges highlight how the microbiology of fruit juices has evolved over the years, when aspects of stability or microbiological safety are under consideration.

A novel mesophilic, acidophilic, endospore-forming bacterium, designated strain ZJ-6T, was isolated from Zi-Jin copper mine in Inner Mongolia, China. Cells of strain ZJ-6T were rod-shaped, stained Gram-positive or were Gram-variable, and grew aerobically at 25–35 °C (optimum, 30 °C) and pH 2.0–6.0 (optimum, pH 3.5). 16S rRNA gene sequence analysis showed that strain ZJ-6T was related phylogenetically to members of the genus Alicyclobacillus, with 16S rRNA gene sequence similarities of 89.5–94.2 %. Cells contained MK-7 as the major quinone and the DNA G+C content was 51.2 mol%. Strain ZJ-6T possessed a number of phenotypic characteristics that differentiated it from recognized Alicyclobacillus species, including its growth temperature, assimilation of various carbon sources, production of acids from a range of compounds, and the ability to grow chemoautotrophically using ferrous iron, elemental sulfur and tetrathionate as electron donors. The predominant cellular fatty acids of strain ZJ-6T were anteiso-C15 : 0 (67.1 %), iso-C16 : 0 (7.7 %) and anteiso-C17 : 0 (7.4 %); ω-alicyclic fatty acids were not found. On the basis of these results, it is concluded that strain ZJ-6T represents a novel species within the genus Alicyclobacillus, for which the name Alicyclobacillus aeris sp. nov. is proposed; the type strain is ZJ-6T (=CGMCC 1.7072T=NBRC 104953T).

Alicyclobacillus aeris sp. nov., a novel ferrous- and sulfur-oxidizing bacterium isolated from a copper mine

In the course of a microbiological survey of various beverages and their raw materials, 180 moderately thermophilic, acidophilic, spore-forming bacteria were isolated from several samples. Seven representative strains, selected from 180 isolates based on morphology and the hyper-variable region of the 16S rRNA gene, were identified as Alicyclobacillus acidocaldarius, A. acidoterrestris and Alicyclobacillus genomic species 1, according to their phenotypic, chemotaxonomic, genotypic and phylogenetic features. Cellular fatty acid composition, quinone species, and DNA base composition of the seven strains were similar to those of the type/reference strains. However, physiological and biochemical characteristics were variable within each species. Moreover, sequence analyses of the 16S rRNA and partial gyrB genes revealed that, in a few strains, the phylogeny derived from both molecules was not consistent. Results of a ribotype analysis also supported this genetic heterogeneity. These results indicate a high level of diversity within species in the genus Alicyclobacillus with regards to both phenotype and genotype. In addition, an emended description of A. acidocaldarius is given on the basis of obtained polyphasic taxonomic data.
