In recent years, cellulose has attracted much attention because of its excellent properties, such as its hydrophilicity, mechanical properties, biodegradability, biocompatibility, low cost and low toxicity. In addition, cellulose and its derivatives contain abundant hydrophilic functional groups (such as hydroxyl, carboxyl and aldehyde groups), which are good raw materials for synthesizing biocompatible hydrogels. In this paper, the application prospects of cellulose and its derivatives-based hydrogels in biomedical tissue engineering are summarized and discussed through the analysis of recent research. Firstly, we discuss the structure and properties of cellulose, nano celluloses (NC) from different sources (including cellulose nanocrystals (CNC), cellulose nanofibrils (CNF) and bacterial nano celluloses (BNC)) and cellulose derivatives (including cellulose ethers and cellulose esters) obtained by different modification methods. Then, the properties and preparation methods of physical and chemical cellulose hydrogels are described, respectively. The application of cellulose-based hydrogels as a tissue engineering scaffold (skin, bone and cartilage) in the biomedical field is introduced. Finally, the challenges and prospects of cellulose-based hydrogels in tissue engineering are summarized.

Chronic ulcers are among the main causes of morbidity and mortality due to the high probability of infection and sepsis and therefore exert a significant impact on public health resources. Numerous types of dressings are used for the treatment of skin ulcers-each with different advantages and disadvantages. Bacterial cellulose (BC) has received enormous interest in the cosmetic, pharmaceutical, and medical fields due to its biological, physical, and mechanical characteristics, which enable the creation of polymer composites and blends with broad applications. In the medical field, BC was at first used in wound dressings, tissue regeneration, and artificial blood vessels. This material is suitable for treating various skin diseases due its considerable fluid retention and medication loading properties. BC membranes are used as a temporary dressing for skin treatments due to their excellent fit to the body, reduction in pain, and acceleration of epithelial regeneration. BC-based composites and blends have been evaluated and synthesized both in vitro and in vivo to create an ideal microenvironment for wound healing. This review describes different methods of producing and handling BC for use in the medical field and highlights the qualities of BC in detail with emphasis on biomedical reports that demonstrate its utility. Moreover, it gives an account of biomedical applications, especially for tissue engineering and wound dressing materials reported until date. This review also includes patents of BC applied as a wound dressing material.

Today, numerous studies have focused on the design of novel scaffolds for tissue engineering and regenerative medicine applications; however, several challenges still exist in terms of biocompatibility/cytocompatibility, degradability, cell attachment/proliferation, nutrient diffusion, large-scale production, and clinical translation studies. Greener and safer technologies can help to produce scaffolds with the benefits of cost-effectiveness, high biocompatibility, and biorenewability/sustainability, reducing their toxicity and possible side effects. However, some challenges persist regarding their degradability, purity, having enough porosity, and possible immunogenicity. In this context, naturally derived cellulose-based scaffolds with high biocompatibility, ease of production, availability, sustainability/renewability, and environmentally benign attributes can be applied for designing scaffolds. These cellulose-based scaffolds have shown unique mechanical properties, improved cell attachment/proliferation, multifunctionality, and enhanced biocompatibility/cytocompatibility, which make them promising candidates for tissue engineering applications. Herein, the salient developments pertaining to cellulose-based scaffolds for neural, bone, cardiovascular, and skin tissue engineering are deliberated, focusing on the challenges and opportunities.

Cellulose-Based Composites as Scaffolds for Tissue Engineering: Recent Advances

The escalating demand for sustainable materials has propelled cellulose into the spotlight as a promising alternative to petroleum-based products. As the most abundant organic polymer on Earth, cellulose is ubiquitous, found in plants, bacteria, and even a unique marine animal-the tunicate. Cellulose polymers naturally give rise to microscale semi-crystalline fibers and nanoscale crystalline regions known as cellulose nanocrystals (CNCs). Exhibiting rod-like structures with widths spanning 3 to 50 nm and lengths ranging from 50 nm to several microns, CNC characteristics vary based on the cellulose source. The degree of crystallinity, crucial for CNC properties, fluctuates between 49 and 95 % depending on the source and synthesis method. CNCs, with their exceptional properties such as high aspect ratio, relatively low density (≈1.6 g cm-3), high axial elastic modulus (≈150 GPa), significant tensile strength, and birefringence, emerge as ideal candidates for biodegradable fillers in nanocomposites and functional materials. The percolation threshold, a mathematical concept defining long-range connectivity between filler and polymer, governs the effectiveness of reinforcement in nanocomposites. This threshold is intricately influenced by the aspect ratio and molecular interaction strength, impacting CNC performance in polymeric and pure nanocomposite materials. This comprehensive review explores diverse aspects of CNCs, encompassing their derivation from various sources, methods of modification (both physical and chemical), and hybridization with heterogeneous fillers. Special attention is devoted to the hybridization of CNCs derived from tunicates (TCNC) with those from wood (WCNC), leveraging the distinct advantages of each. The overarching objective is to demonstrate how this hybridization strategy mitigates the limitations of WCNC in composite materials, offering improved interaction and enhanced percolation. This, in turn, is anticipated to elevate the reinforcing effects and pave the way for the development of nanocomposites with tunable viscoelastic, physicochemical, and mechanical properties. 

Cellulose nanocrystals in the development of biodegradable materials: A review on CNC resources, modification, and their hybridization.

Carbon nanomaterials (CNMs) mainly include fullerene, carbon nanotubes, graphene, carbon quantum dots, nanodiamonds, and their derivatives. As a new type of material in the field of nanomaterials, it has outstanding physical and chemical properties, such as minor size effects, substantial specific surface area, extremely high reaction activity, biocompatibility, and chemical stability, which have attracted widespread attention in the medical community in the past decade. However, the single use of carbon nanomaterials has problems such as self-aggregation and poor water solubility. Researchers have recently combined them with bacterial cellulose to form a new intelligent composite material to improve the defects of carbon nanomaterials. This composite material has been widely synthesized and used in targeted drug delivery, biosensors, antibacterial dressings, tissue engineering scaffolds, and other nanomedicine fields. This paper mainly reviews the research progress of carbon nanomaterials based on bacterial cellulose in nanomedicine. In addition, the potential cytotoxicity of these composite materials and their components in vitro and in vivo was discussed, as well as the challenges and gaps that need to be addressed in future clinical applications. 

A review of carbon nanomaterials/bacterial cellulose composites for nanomedicine applications.

As a renewable, biodegradable, and non-toxic material with moderate mechanical and thermal properties, nanocellulose-based hydrogels are receiving immense consideration for various biomedical applications. With the unique properties of excellent skeletal structure (hydrophilic functional groups) and micro-nano size (small size effect), nanocellulose can maintain the three-dimensional structure of the hydrogel to a large extent, providing mechanical strength while ensuring the moisture content. Owing to its unique features, nanocellulose-based hydrogels have made excellent progress in research and development on tissue engineering, drug carriers, wound dressings, development of synthetic organs, 3D printing, and biosensing. This review provides an overview of the synthesis of different types of nanocellulose, including cellulose nanocrystals, cellulose nanofibers, and bacterial nanocellulose, and describes their unique features. It further provides an updated knowledge of the development of nanocellulose-based functional biomaterials for various biomedical applications. Finally, it discusses the future perspective of nanocellulose-based research for its advanced biomedical applications.
