Electrochemical non-enzymatic glucose sensors: recent progress and perspectives

In recent decades, as a subclass of biomaterials, biologically sensitive nanoparticles have attracted increased scientific interest. Many of the demands for physiologically responsive nanomaterials in applications involving the human body cannot be met by conventional technologies. Due to the field’s importance, considerable effort has been expended, and biologically responsive nanomaterials have achieved remarkable success thus far. This review summarizes the recent advancements in biologically responsive nanomaterials and their applications in biosensing and molecular imaging. The nanomaterials change their structure or increase the chemical reaction ratio in response to specific bio-relevant stimuli (such as pH, redox potentials, enzyme kinds, and concentrations) in order to improve the signal for biologically responsive diagnosis. We use various case studies to illustrate the existing issues and provide a clear sense of direction in this area. Furthermore, the limitations and prospects of these nanomaterials for diagnosis are also discussed.

Recent Advance in Biological Responsive Nanomaterials for Biosensing and Molecular Imaging Application

Hydrogen peroxide (H2O2) is a major metabolite of oxygen known as reactive oxygen species (ROS) in living organisms. Monitoring H2O2 released from living cells is of great significance as its concentration level is related to several diseases, which imposes stricter requirements on the sensitivity of detection technology. In this work, we have found that carbon black (CB) could preserve bimetallic PdCu alloy’s nanostructure, meanwhile improve the dispersion of nanoparticles, provided high conductivity and large specific surface area, thus the PdCu/CB nanohybrid exhibited enhanced electroanalytical performance of H2O2. Under the optimal conditions, the PdCu/CB nanohybrid modified electrode showed a low detection limit of 54 nM with a wide detection range from 0.4 μM to 5.0 mM. Finally, the application of PdCu/CB/GCE in real-time monitoring H2O2 released from Raw 264.7 cells was achieved, suggesting the PdCu/CB nanohybrid was the promising nanomaterial in monitoring physiological processes.

Self-assembly exploits noncovalent interaction to offer an effective method for the fabrication of materials. For Na9 [EuW10 O36 ] ⋅ 32H2 O (EuW10 ), the negative charges and abundant oxygen atoms on its surface provide a handle for static self-assembly. New properties are envisioned for EuW10 aggregates which are able to display such kinetics and time-programming characteristics, in order to satisfy more complex and intelligent application scenarios, such as DNA binding and information encryption. In this work, EuW10 coupling with stimuli-responsive dodecyl dimethylamine oxide (C12 DMAO) can generate versatile aggregates with pH-responsive properties. We demonstrated the temporal programming of the assembly and disassembly of EuW10 nanospheres using a pH clock reaction of acid/urease hydrolysis. The pH clock reaction endows EuW10 assemblies with dynamical properties, in which the charges and fluorescence changes are coded in this system. These fluorescent assemblies provide new application in time-programmed DNA capture and information encryption.

Nanoporous (NP) PdCu alloy is easily fabricated by dealloying PdCuAl ternary alloy in dilute sulfuric acid. Selectively dissolving Al from PdCuAl alloy generates the three-dimensional uniform nanosponge architecture with narrow ligament size distribution. Benefitting from the unique nanoporous architecture and the alloying effect, the as-made NP-PdCu exhibits outstanding sensing performance towards the detection of hydrogen peroxide (H2O2) and glucose. Compared with NP-Pd and commercial Pd/C catalysts, the NP-PdCu alloy presents high sensitivity, wide linear range of 0.1–2.0 mM, low detection limit of 2.1 μM, and long-term stability toward H2O2 detection. In addition, the NP-PdCu can efficiently detect glucose in a wide concentration range (1–30 mM) with the low detection limit of 1.9 μM. Moreover, the NP-PdCu exhibits good anti-interference toward ascorbic acid, uric acid, and dopamine. Characterized by easy preparation, unique electrocatalytic activity, and high structure stability, the NP-PdCu alloy possesses great application prospect to construct platform for electrochemical sensing.
