Chalcone dye-based fluorescent probe for selective and specific detection of cysteine in lysosomes of living cells

There has been rapid progress in the development of fast, sensitive, cheap and low-cytotoxicity micro-molecule fluorescent probes for application in various fields, including disease diagnosis, food safety and environmental safety. As an analytical tool, dual-function fluorescent probes with dual-emission responses have attracted considerable attention due to their cost-effectiveness and efficiency over single-function sensors. This review primarily describes research progress on multifunctional probes in terms of the reaction type and coordination type, as well as the general design principles of probes. The analytes include reactive oxygen species (ROS), reactive sulfur species (RSS), harmful cations and anions, etc. Multifunctional probes for food, medical and environmental applications are listed for future research. To improve the development of rapid detection methods, trends and strategies in the development of multifunctional fluorescent probes are also discussed.

Research Progress on Multifunctional Fluorescent Probes for Biological Imaging, Food and Environmental Detection.

Owing to the important physiological sits of biothiols (Cys, Hcy, and GSH), developing accurate detection methods capable of qualitative and quantitative analysis of biothiols in living systems is needed for understanding the biological profile of biothiols. In this work, we have designed and synthesized a 4′-hydroxy-[1,1′-biphenyl]-4-carbonitrile modified with NBD group-based fluorescent probe, BPN-NBD, for sensitive detection of Cys/Hcy and GSH by dual emission signals via a single-wavelength excitation. BPN-NBD exhibited an obvious blue fluorescence (λmaxem = 475 nm) upon the treatment with GSH and reacted with Cys/Hcy to give a mixed blue-green fluorescence (λmaxem = 475 and 545 nm). Meanwhile, BPN-NDB performed sufficient selectivity, rapid detection (150 s), high sensitivity (0.011 µM for Cys, 0.015 µM for Hcy, and 0.003 µM for GSH) and could work via a single-wavelength excitation to analytes and had the ability to image Cys/Hcy from GSH in living MCF-7 cells, tumor tissues, and zebrafish by exhibiting different fluorescence signals. Overall, this work provided a powerful tool for thiols visualization in biological and medical applications.

The eco–environmental toxicity of the rare metal palladium (Pd) has received extensive attention, thereby requiring simple and practical methods for detecting Pd. Herein, a new probe (HPD) based on dicyanoisophorone dye was designed and synthesized for the detection of Pd. HPD provides several advantages for tracking Pd, including near–infrared (NIR) emission (λex = 670 nm, λem = 700 nm), naked–eye recognition, and comprehensive detection of 0/2/4 valence Pd with good selectivity and anti–interference. The reaction–sensing mechanism of HPD for Pd was further studied using density functional theory calculations. HPD was demonstrated to be a promising tool for the detection of Pd in real water samples, as well as the imaging of Pd in living cells and animals.
