Photodynamic therapy for treating infected skin wounds: A systematic review and meta-analysis from randomized clinical trials.

Aminolevulinic acid (ALA) is considered one of the most critical plants growth regulators and essential precursors for chlorophyll biosynthesis; besides, its photodynamic activity can be used to exterminate larvae and microorganisms in plants and soil. Silver nanoparticles (AgNPs) have unique physicochemical properties and potent antimicrobial, antiviral, and antifungal activities, and in agriculture, their application as nanopesticides has been proposed. In this study, silver and silver–iron nanoparticles capped/stabilized with aminolevulinic acid (ALAAgNPs and ALAAgFeNPs) were synthesized by the photoreduction method and characterized by UV-vis spectroscopy, transmission electron microscopy, and zeta potential analysis. The kinetics of 1O2 generation from ALAAgFeNPs were obtained. The ALANP toxicity was evaluated on stalks of E. densa by observing cell morphology changes and measuring chlorophyll content compared with water-treated plants. Antimicrobial activity was tested against E. coli, P. aeruginosa, and Candida albicans. The results suggested that ALANPs (prepared with [AgNO3] ≤ 0.2 mM and [ALA] ≤ 0.4 mM) could be suitable for applications in the agricultural sector. The presence of ∼0.3 mmol of iron in ALAAgNPs synthesis increased cell uptake and chlorophyll synthesis.

Synthesis, characterization, antimicrobial activity, and toxicity evaluation of aminolevulinic acid–silver and silver–iron nanoparticles for potential applications in agriculture

Photodynamic therapy (PDT) is a medical treatment that utilizes photosensitizing agents, along with light, to produce reactive oxygen species that can kill nearby cells. When the photosensitizer is exposed to a specific wavelength of light, it becomes activated and generates reactive oxygen that can destroy cancer cells, bacteria, and other pathogenic micro-organisms. PDT is commonly used in dermatology for treating actinic keratosis, basal cell carcinoma, and other skin conditions. It is also being explored for applications in oncology, such as treating esophageal and lung cancers, as well as in ophthalmology for age-related macular degeneration. In this study, we provide a comprehensive review of PDT, covering its fundamental principles and mechanisms, as well as the critical components for its function. We examine key aspects of PDT, including its current clinical applications and potential future developments. Additionally, we discuss the advantages and disadvantages of PDT, addressing the various challenges associated with its implementation and optimization. This review aims to offer a thorough understanding of PDT, highlighting its transformative potential in medical treatments while acknowledging the areas requiring further research and development. 

Photodynamic Therapy Review: Past, Present, Future, Opportunities and Challenges

Infections caused by multidrug-resistant bacterial and fungal pathogens represent a significant global health concern, contributing to increased morbidity and mortality rates. Therefore, it is crucial to develop novel compounds targeting drug-resistant microbial strains. Herein, we report the synthesis of amino acid derivatives bearing an incorporated 4-hydroxyphenyl moiety with various substitutions. The resultant novel 3-((4-hydroxyphenyl)amino)propanoic acid derivatives 2–37 exhibited structure-dependent antimicrobial activity against both ESKAPE group bacteria and drug-resistant Candida species. Furthermore, these derivatives demonstrated substantial activity against Candida auris, with minimum inhibitory concentrations ranging from 0.5 to 64 µg/mL. Hydrazones 14–16, containing heterocyclic substituents, showed the most potent and broad-spectrum antimicrobial activity. This activity extended to methicillin-resistant Staphylococcus aureus (MRSA) with MIC values ranging from 1 to 8 µg/mL, vancomycin-resistant Enterococcus faecalis (0.5–2 µg/mL), Gram-negative pathogens (MIC 8–64 µg/mL), and drug-resistant Candida species (MIC 8–64 µg/mL), including Candida auris. Collectively, these findings underscore the potential utility of the novel 3-((4-hydroxyphenyl)amino)propanoic acid scaffold for further development as a foundational platform for novel antimicrobial agents targeting emerging and drug-resistant bacterial and fungal pathogens.

Synthesis of 3-((4-Hydroxyphenyl)amino)propanoic Acid Derivatives as Promising Scaffolds for the Development of Antimicrobial Candidates Targeting Multidrug-Resistant Bacterial and Fungal Pathogens

Antimicrobial photodynamic inactivation (aPDI) is a method that specifically kills target cells by combining a photosensitizer and irradiation with light at the appropriate wavelength. The natural amino acid, 5-aminolevulinic acid (5-ALA), is the precursor of endogenous porphyrins in the heme biosynthesis pathway. This review summarizes the recent progress in understanding the biosynthetic pathways and regulatory mechanisms of 5-ALA synthesis in biological hosts. The effectiveness of 5-ALA-aPDI in destroying various groups of pathogens (viruses, fungi, yeasts, parasites) was presented, but greater attention was focused on the antibacterial activity of this technique. Finally, the clinical applications of 5-ALA in therapies using 5-ALA and visible light (treatment of ulcers and disinfection of dental canals) were described.

On the Possibility of Using 5-Aminolevulinic Acid in the Light-Induced Destruction of Microorganisms

Background Antibiotic resistance is increasing day by day, thereby increase the chances of more infections by resistant bacteria. In this situation, antimicrobial photodynamic therapy (aPDT) is gaining more attraction. Objective To evaluate the antimicrobial effect of ALA derivatives using photodynamic therapy. Materials and methods In this study, we evaluated the aPDT effect of different derivatives of 5-ALA. In vivo and in vitro studies were performed to measure the antimicrobial activity. Different light doses and different concentrations of drugs were used to test anti-bacterial effect of drugs as well as to detect any physiological changes in animal model after the treatment. Results In vivo studies revealed that ALA-methyl ester, ALA-hexyl ester, and ALA-13A are potent photosensitizers. In vitro studies involved wound healing rate, body weight, and dietary intake were evaluated, and results showed that ALA, ALA-methyl ester, ALA-hexyl ester, and ALA-13A had good anti-bacterial effects, fast healing rate, and no effect on other physical parameters. Conclusion Photodynamic therapy is increasingly used to treat different types of skin infections caused by bacterial strains. Our studies revealed that ALA-methyl ester, ALA-hexyl ester, and ALA-13A are promising photosensitizers for photodynamic therapy to inhibit the growth of resistant bacterial strains.

The invention relates to a 6'-subsituted aminoyl benzimidazole-5-substituted methylindole derivative and preparation and application thereof. The structure is shown in the general formula I. In preparation, reactants are subjected to substituting reaction; then, alkaline liquor is added for hydrolysis, and the derivative is obtained. The obtained compound is an angiotensin II receptor (AT1 receptor) antagonist and is applied in preparation of medicines for preventing or treating hypertension, coronary heart diseases, heart, brain and kidney vessel diseases, migraine, pulmonary arterial hypertension and other diseases.
