Nutrient requirements of dairy cattle

The experiment was conducted to compare the growth performance, rumen fermentation, nutrient digestibility, and ruminal and fecal bacterial community between yaks and cattle-yaks. Ten male yaks (36-month-old) were used as the yak (YAK) group and 10 male cattle-yaks with similar age were selected as the cattle-yak (CAY) group. All the animals were fed same ration and the experiment lasted for 60 days. The results showed that the average daily gain and dry matter intake of CAY group were higher (P < 0.05) than those of YAK group. The ruminal concentrations of total volatile fatty acids, acetate, and butyrate were higher (P < 0.05) in CAY group than those in YAK group. However, the neutral detergent fiber and acid detergent fiber digestibility exhibited an opposite between two groups. In the rumen, the relative abundances of Prevotella 1 and Prevotellaceae UCG-001 were higher (P < 0.05) and Succiniclasticum and Butyrivibrio 2 were lower (P < 0.05) in YAK group compared to CAY group. In the feces, the unclassified Lachnospiraceae, Lachnospiraceae NK4A136 group, and Lachnospiraceae AC2044 group were significantly enriched (P < 0.05) in YAK group, whereas the Ruminococcaceae UCG-010, Ruminococcaceae UCG-013, and Succiniclasticum were significantly enriched (P < 0.05) in CAY group. Overall, under the same diet, the yaks have higher fiber utilization and cattle-yaks have higher energy utilization.

Comparative study of growth performance, nutrient digestibility, and ruminal and fecal bacterial community between yaks and cattle-yaks raised by stall-feeding

BACKGROUND
Considering the high energy demand of lactation and the potential of guanidinoacetic acid (GAA) addition on the increase in creatine supply for cows, the study investigated the effects of 0, 0.3, 0.6 and 0.9 g kg-1 DM of GAA supplementation on lactation performance, nutrient digestion and ruminal fermentation in dairy cows. The study used fourty mid-lactation multiparous Holstein cows and lasted for 100 days.


RESULTS
Dry matter intake was not affected but milk and milk components yields and feed efficiency increased linearly with increasing GAA addition. The total-tract digestibility of dry matter, organic matter, neutral detergent fibre, acid detergent fibre and non-fibre carbohydrates increased linearly and that of crude protein increased quadratically with increasing GAA addition. When the addition level of GAA increased, ruminal pH, molar percentages of butyrate, isobutyrate and isovalerate and acetate-to-propionate ratio decreased linearly, total volatile fatty acids concentration and propionate molar percentage increased linearly, but acetate molar percentage and ammonia-N concentration were unaltered. The activities of fibrolytic enzymes, α-amylase and protease increased linearly. The populations of total bacteria, fungi, Ruminococcus albus, Fibrobacter succinogenes, R. flavefaciens, Ruminobacter amylophilus and Prevotella ruminicola increased linearly, but protozoa and methanogens decreased linearly with increasing GAA addition. As for the blood metabolites, concentrations of glucose, urea nitrogen and methionine were unchanged, total protein, albumin, creatine and homocysteine increased linearly but folate decreased linearly with increasing GAA supply.


CONCLUSION
The current results indicated that supplementation of GAA improved milk performance and rumen fermentation in lactating dairy cows. This article is protected by copyright. All rights reserved.

Effects of guanidinoacetic acid supplementation on lactation performance, nutrient digestion and rumen fermentation in Holstein dairy cows.

Guanidine acetic acid (GAA) is increasingly considered as a nutritional growth promoter in monogastric animals. Whether or not such response would exist in rapid-growing lambs is unclear yet. The objective of this study was to investigate whether dietary supplementation with uncoated GAA (UGAA) and coated GAA (CGAA) could alter growth performance, nutrient digestion, serum metabolites, and antioxidant capacity in lambs. Seventy-two small-tailed Han lambs initially weighed 12 ± 1.6 kg were randomly allocated into six groups in a 2 × 3 factorial experimental design including two forage-type rations [Oaten hay (OH) vs. its combination with wheat silage (OHWS)] and three GAA treatment per ration: no GAA, 1 g UGAA, and 1 g CGAA per kg dry matter. The whole experiment was completed in two consecutive growing stages (stage 1, 13–30 kg; stage 2, 30–50 kg). Under high-concentrate feeding pattern (Stage 1, 25: 75; Stage 2, 20: 80), UGAA or CGAA supplementation in young lambs presented greater dry matter intake (DMI) in stage 1 and average daily gain (ADG) in the whole experimental period; lambs in OH group had higher ADG and DMI than that in OHWS group in stage 1 and whole experimental period, but this phenomenon was not observed in stage 2. Both UCGA and CGAA addition increased dietary DM, organic matter (OM), neutral detergent fiber (NDF), and acid detergent fiber (ADF) digestion in both stages. In blood metabolism, UCGA and CGAA addition resulted in a greater total protein (TP) and insulin-like growth factor 1(IGF-1) levels, as well as antioxidant capacity; at the same time, UCGA and CGAA addition increased GAA metabolism-creatine kinase and decreased guanidinoacetate N-methyltransferase (GAMT) and L-Arginine glycine amidine transferase catalyzes (AGAT) activity. In a brief, the results obtained in the present study suggested that GAA (UGAA and CGAA; 1 g/kg DM) could be applied to improve growth performance in younger (13–30 kg) instead of older (30–50 kg) lambs in high-concentrate feedlotting practice.

Objective
The aim of this research was to explore the effects of dietary substitution of alfalfa hay by amaranth hay on production performance, rumen fermentation, nutrient digestibility, serum biochemical parameters and antioxidant ability in dairy cows.


Methods
A total of 45 healthy Holstein cows with same parity and similar milk yield and body weight were randomly divided into 3 groups: control diet without amaranth hay (CON) or 50% and 100% alfalfa hay replaced by an equal amount of amaranth hay (dry matter basis, AH1 and AH2, respectively). All the cows were fed regularly 3 times a day at 06:30, 14:30 and 22:30 and had free access to water. The experiment lasted for 60 d.


Results
The dry matter intake of CON and AH1 groups was higher (p<0.05) than that of AH2 group. Compared with AH1 group, the milk yield of AH2 group was reduced (p<0.05). Moreover, dietary substitution of alfalfa hay by amaranth hay increased (p<0.05) milk fat, ammonia nitrogen and acetate concentrations. However, the crude protein digestibility of AH2 group was lower (p<0.05) than that of CON group, while an opposite tendency of serum urea nitrogen was found between two groups. The neutral detergent fiber digestibility of AH1 group was increased (p<0.05) when compared to AH2 group. Amaranth hay treatment increased (p<0.05) the serum concentration of glutathione peroxidase in dairy cows. Compared with CON group, the malonaldehyde activity of AH1 group was decreased (p<0.05).


Conclusion
Dietary replacing alfalfa hay with amaranth hay (50% ratio) of dairy cows did not affect production performance but improved their antioxidant ability.

Guanidinoacetic acid (GAA) is the only precursor for the creatine synthesis of vertebrates. Creatine (Cr) and phosphocreatine (PCr) are able to provide energy for the rapid growth and development of the muscle tissue. This study evaluated the effects of dietary different levels GAA on growth performance, GAA absorption and creatine metabolism of lambs. Twenty-four 3-month-old healthy Kazakh male lambs (body weight = 27.35± 0.58 kg) were randomly divided into four groups with 6 lambs in each group. The lambs were fed with the basal diets supplemented with 0 (0 mg/kg group), 500 (500 mg/kg group), 1000 (1000 mg/kg group) and 1500 mg (1500 mg/kg group) GAA per kg diet (DM basis), respectively. The results showed that, as the GAA content of the diet increased, there was a quadratic change in DMI, with the lowest in the 500 mg/kg group and the highest in the 0 mg/kg group. The CK enzyme activity and ATP content in quadriceps muscle increased linearly with increasing levels of diary GAA in the diet. PCr levels and ADP levels in the longest dorsal muscle increased linearly with increasing levels of GAA in the diet. The relative expression of SLC6A6 and SLC6A8 mRNA in the jejunum and ileum mucosa showed a quadratic change as the dietary GAA level increased, with the lowest relative expression in both the 1500 mg/kg group. With the increase of dietary GAA level, both Cr concentration in hepatic vein plasma and the portal plasma GAA concentration shows a quadratic change, with the highest concentration in the 500 mg/kg group and the lowest concentration in the 0 mg/kg group. Therefore, dietary supplementation with 500~1000 mg/kg DM GAA is recommended for lambs.

Effects of dietary guanidinoacetic acid on growth performance, guanidinoacetic acid absorption and creatine metabolism of lambs

Context L-citrulline (L-Cit) does not degrade in the rumen and has the ability to form peptide bonds in the body; however, it does not participate in protein synthesis.   Aims This study aimed to evaluate the effects of L-Cit on the reproductive performance of Hu ewes.   Methods In total, 30 ewes were randomly categorised into five groups. The control group was fed with a basic diet, whereas the Experimental Groups I, II, III, and IV were provided feed supplemented with 5, 10, 15, and 20g/day of L-Cit respectively. Blood samples of ewes were collected 4h after feeding on Day 21 of the experiment and before feeding on Day 30. The optimal supplementary feeding dose was selected on the basis of blood biochemical indexes. Overall, ninety 2-year-old ewes were classified into two groups. The control group was fed with a basic diet and the experimental group was fed with a diet supplemented with 10g/day of L-Cit. After 30days of supplementary feeding, reproductive performance of ewes was determined.   Key results The plasma concentrations of Cit, ornithine, and arginine in ewes increased linearly with an increase in the level of L-Cit supplementation. The plasma concentrations of gonadotropin-releasing hormone, luteinising hormone, and follicle-stimulating hormone in the experimental group increased significantly compared with those in the control group. The plasma total antioxidant capacity and catalase, superoxide dismutase, and glutathione peroxidase in the experimental group were significantly higher than those in the control group, whereas the concentrations of malondialdehyde in all experimental groups were significantly lower than those in the control group. The conception, lambing, and double lambing rates of the experimental group were increased by 28.76%, 15.90%, and 40.21% respectively.   Conclusions Supplementation with different doses of L-Cit can improve the concentrations of some plasma amino acids and reproductive hormones as well as antioxidant capacity of ewes. Supplementary feeding with 10g/day of L-Cit could increase the lambing and double lambing rates of ewes.   Implication L-Cit can improve the reproductive performance of ewes.

Effects of supplementation with different concentrations of L-citrulline on the plasma amino acid concentration, reproductive hormone concentrations, antioxidant capacity, and reproductive performance of Hu ewes

OBJECTIVE The purpose of present study was to investigate the effects of condensed molasses fermentation solubles (CMS) on lactation performance, rumen fermentation, nutrient digestibility, and serum parameters of dairy cows. METHODS A total of 75 healthy Holstein cows with the same parity (milk production = 35±2.5 kg, body weight = 570±28 kg) were randomly selected and divided into 5 groups. One group served as control group (CON; no CMS), whereas the other 4 groups were CMS1 (accounted for 1% of the diet), CMS2 (2%), CMS3 (3%), and CMS4 (4%). All cows were fed regularly three times each day at 0800, 1600, and 2400 h. Cows received diet and water ad libitum. The experiment lasted for 60 days. RESULTS Results showed that the dry matter intake, milk yield, and protein of CMS2 were maximum and higher (p<0.05) than CMS4. The ruminal pH was observed less than 6 in CMS3 and CMS4 groups. No noticeable difference of microbial protein was found between CON and CMS2 groups, while the microbial protein in these groups was higher (p<0.05) than CMS3 and CMS4 groups. The apparent digestibility of dry matter, organic matter, and crude protein in CMS2 group was higher (p<0.05) than CMS3 and CMS4 groups. Compared to CMS3 and CMS4 groups, the CMS2 group increased (p<0.05) the serum concentrations of immunoglobulin G and immunoglobulin M on d 60. CONCLUSION Therefore, it is practicable that CMS substitutes for a part of concentrates in lactating cows' diets, but higher addition of CMS (more than 3% of the diet) could decrease production performance of dairy cows as seen in the present study.

Use of condensed molasses fermentation solubles as an alternative source of concentrates in dairy cows.

Gossypol, a yellow polyphenolic compound found in the Gossypium genus, is toxic to animals that ingest cotton-derived feed materials. However, ruminants display a notable tolerance to gossypol, attributed to the pivotal role of ruminal microorganisms in its degradation. The mechanisms of how rumen microorganisms degrade and tolerate gossypol remain unclear. Therefore, in this study, Enterobacter sp. GD5 was isolated from rumen fluid, and the effects of gossypol on its metabolism and gene expression were investigated using liquid chromatography-mass spectrometry (LC-MS) and RNA analyses. The LC-MS results revealed that gossypol significantly altered the metabolic profiles of 15 metabolites (eight upregulated and seven downregulated). The Kyoto Encyclopedia of Genes and Genomes analysis results showed that significantly different metabolites were associated with glutathione metabolism in both positive and negative ion modes, where gossypol significantly affected the biosynthesis of amino acids in the negative ion mode. Transcriptomic analysis indicated that gossypol significantly affected 132 genes (104 upregulated and 28 downregulated), with significant changes observed in the expression of catalase peroxidase, glutaredoxin-1, glutathione reductase, thioredoxin 2, thioredoxin reductase, and alkyl hydroperoxide reductase subunit F, which are related to antioxidative stress. Furthermore, Gene Ontology analysis revealed significant changes in homeostatic processes following gossypol supplementation. Overall, these results indicate that gossypol induces oxidative stress, resulting in the increased expression of antioxidative stress-related genes in Enterobacter sp. GD5, which may partially explain its tolerance to gossypol.
