Abstract With the outbreak and pandemic of coronavirus disease‐2019 (COVID‐19), a huge number of people died of it. Apart from lung injuries, multiple organs have been confirmed to be impaired. In COVID‐19 time, primary wound healing processes always prolong, however, its possible underlying mechanisms are still unclear. Therefore, to overcome this clinical problem, clarifying its underlying mechanisms clearly is necessary and urgently needed. In this review, we summarized that COVID‐19 can prolong primary wound healing by inducing excessive inflammation and oxidative stress, disturbing immune system and haematological system, as well as influencing the functions and viability of epidermal stem cells (ESCs). Otherwise, we summarized that the strict control measures of blocking up COVID‐19 pandemic can also have side effects on primary wound healing process.

The coronavirus outbreak, which initially started in Wuhan China, has rapidly led to numerous morbidities and mortalities worldwide. Although potential antiviral and antiinflammatory medicines are available, several individuals are dying daily. In order to thoroughly tackle this deadly virus, the knowledge of genomics, metagenomics, and pan genomics is required, not only to devise new treatment regimens but also to improve the present approaches. Understanding the genomic organization, diversity and structural complexity of this virus can help to figure out several previously unanswered questions. SARS-CoV-2 corona virus comprises of four major structural proteins, namely, the spike surface glycoprotein, tiny envelope protein, matrix protein, and nucleocapsid protein along with accessory proteins that contribute to pathogenesis and persistence of the virus in one way or the other. This chapter covers genomics, metagenomics, and pan-genomics-based strategies that can facilitate to figure the possible mutational recombination and trace the phylogenetic background of the species. Sequencing has been performed and the hence derived viral sequences have been deposited into exclusive repositories to speed up research and explore targeted treatment options. Moreover, immunoinformatics approaches and reverse vaccinology have been applied to speed up the process of formulating reliable, safe, and specific therapeutic options, rapidly. 

Genomics, metagenomics, and pan-genomics approaches in COVID-19

COVID-19, caused by SARS-CoV-2, has emerged as the most destructive emerging infectious disease of the 21st century. Vaccination is an effective method to combat viral diseases. However, due to the constant mutation of the virus, new variants may weaken the efficacy of vaccines. In the current field of new coronavirus research, viral protease inhibitors have emerged as a highly regarded therapeutic strategy. Nevertheless, existing viral protease inhibitors do not fully meet the therapeutic needs. Therefore, this paper turned to traditional Chinese medicine to explore new active compounds. This study focused on 24 isolated compounds from Acorus calamus L. and identified 8 active components that exhibited significant inhibitory effects on SARS-CoV-2 PLpro. Among these, the compound 1R,5R,7S-guaiane-4R,10R-diol-6-one demonstrated the best inhibitory activity with IC50 values of 0.386 ± 0.118 μM. Additionally, menecubebane B and neo-acorane A exhibited inhibitory activity against both Mpro and PLpro proteases, indicating their potential as dual-target inhibitors. The molecular docking results confirmed the stable conformations of these compounds with the key targets and their good activity. ADMET and Lipinski's rule analyses revealed that all the small molecule ligands possessed excellent oral absorption properties. This study provides an experimental foundation for the discovery of promising antiviral lead compounds. 

Screening for Active Compounds of Acorus calamus against SARS-CoV-2 Viral Protease and Mechanism Prediction

SARS-CoV-2 belongs to the coronavirus family. Comparing genomic features of viral genomes of coronavirus family can improve our understanding about SARS-CoV-2. Here we present the first pan-genome analysis of 3,932 whole genomes of 101 species out of 4 genera from the coronavirus family. We found that a total of 181 genes in the pan-genome of coronavirus family, among which only 3 genes, the S gene, M gene and N gene, are highly conserved. We also constructed a pan-genome from 23,539 whole genomes of SARS-CoV-2. There are 13 genes in total in the SARS-CoV-2 pan-genome. All of the 13 genes are core genes for SARS-CoV-2. The pan-genome of coronaviruses shows a lower level of diversity than the pan-genomes of other RNA viruses, which contain no core gene. The three highly conserved genes in coronavirus family, which are also core genes in SARS-CoV-2 pan-genome, could be potential targets in developing nucleic acid diagnostic reagents with a decreased possibility of cross-reaction with other coronavirus species.
