There is great need for stand-alone luminescence-based chemosensors that exemplify selectivity, sensitivity, and applicability and that overcome the challenges that arise from complex, real-world media. Discussed herein are recent developments toward these goals in the field of supramolecular luminescent chemosensors, including macrocycles, polymers, and nanomaterials. Specific focus is placed on the development of new macrocycle hosts since 2010, coupled with considerations of the underlying principles of supramolecular chemistry as well as analytes of interest and common luminophores. State-of-the-art developments in the fields of polymer and nanomaterial sensors are also examined, and some remaining unsolved challenges in the area of chemosensors are discussed.

Supramolecular Luminescent Sensors

Calix[n]arenes (n = 4, 5, 6, 8) are "chalicelike" phenol-based macrocycles that are among the most fascinating and highly studied scaffolds in supramolecular chemistry. This stems from the functional and tunable diversity at both their upper and lower rims, their preorganized nonpolar cavities and preorganized ion-binding sites, and their well-defined conformations. Conjugation of calixarene scaffolds with various fluorogenic groups has led to the development of smart fluorescent probes that have been utilized as molecular sensors, in bioimaging, for drug and gene delivery, in self-assembly/aggregation, and as smart materials. The fine-tuning and incorporation of different ligating sites in the calix[4]arene scaffold have produced numerous molecular sensors for cations, anions, and biomolecules. Moreover, the aqueous solubility of p-sulfonatocalix[4]arenes has engendered their potential use in drug/gene delivery and enzymatic assays. In addition, because of their strong optical properties, fluorescent calix[4]arenes have been used to develop smart materials, including gels as well as nonlinear optical, organic light-emitting diode, and multiphoton materials. Finally, significant developments in the utility of fluorescent higher calixarenes have been made for bioapplications. This review critically summarizes the recent advances made in all of these different areas.

Revisiting Fluorescent Calixarenes: From Molecular Sensors to Smart Materials

Ion and Molecular Recognition by Lower Rim 1,3-Di-conjugates of Calix[4]arene as Receptors

A simple and easily synthesized fluorescent and colorimetric chemosensor 1 , based on juloidine-imidazole moieties as a binding and signaling unit, has been synthesized in a one-step procedure. Receptor 1 showed immediate responses toward Zn 2+ and Al 3+ ions through selective fluorescence enhancement in buffer-acetonitrile and dimethylformamide (DMF) solution, respectively. Moreover, receptor 1 sensed the two states (Fe(II) and Fe(III)) of iron by “naked eye” with a different color. Upon the addition of Fe 2+ and Fe 3+ into each solution of 1 , the color of the solutions instantly changed from colorless to orange for Fe 2+ and to purple for Fe 3+ . Thus, this sensor provides a novel approach for selectively recognizing the biologically important three elements in the human body simultaneously, for Zn 2+ and Al 3+ by emission spectra and Fe 2+ and Fe 3+ by the naked eye.

A single schiff base molecule for recognizing multiple metal ions: A fluorescence sensor for Zn(II) and Al(III) and colorimetric sensor for Fe(II) and Fe(III)

Considerable amount of research has been carried out on designing and improving metal-recognition methodologies in environmental and biological media. The development of fluorescent-based techniques has proven to be an important milestone for non-invasive metal detection and quantification in a multichannel environment. Metals as natural components of the Earth's crust are generally present in trace concentrations in environmental samples, wherein humic substances have a complexation affinity toward them. Iron, zinc and copper are the 1st, 2nd and 3rd most abundant elements that are indispensable to the human body in trace amounts as they play crucial roles in many biological processes. However, unregulated amounts either an excess or deficiency may exacerbate deterioration of the vital organs and trigger the progression of complications. In addition to these three essential elements, mercury is widely considered to be one of the most hazardous pollutants and highly dangerous elements due to its recognized accumulative and toxic effects in the environment and in biological media. In the present study, we attempted to summarize all the recently developed fluorescent signaling materials for the detection of Cu2+, Fe2+/Fe3+, Zn2+ and Hg2+. The spectral shifts in the molecules on metal chelation, the mode of complexation and the stoichiometries of the resulting adducts have been discussed in detail. Furthermore, we highlight molecules that have been reported as an intracellular metal detector via bioimaging, which can be useful for the future design and development of cell-viable and membrane-permeable molecular probes.
