An efficient and reproducible in vitro propagation protocol has been established for Cadaba fruticosa (L.) Druce. Surface-sterilized nodal stem segments of mature plant were used as explants for culture establishment. Multiple shoots were optimally differentiated from the nodal stem explants through bud breaking on Murashige and Skoog (1962) medium containing 3.0 mg l−1 benzyladenine (BA). The effect of different plant growth regulators and minerals were studied on different stages of micropropagation procedure (i.e., explant establishment, shoot multiplication/growth and ex vitro rooting). Additionally, for enhancing shoot multiplication during subculture, MS medium was modified (MMS) with higher levels of magnesium, potassium and sulphate ions. Out of these, MMS3 medium containing 0.25 mg l−1 each of BA and Kin (N6-furfuryladenine), with 0.1 mg l−1 NAA (α-naphthalene acetic acid) was found the best for shoot multiplication (42.45 ± 3.82 per culture vessel). The in vitro regenerated shoots were rooted under ex vitro conditions on treating the shoot base with 500 mg l−1 of IBA (indole-3 butyric acid) for 3 min on sterile Soilrite®. The ex vitro rooted plants were hardened in the greenhouse and transferred to the field with ≈85 % survival rate. There were not any visual differences between wild and micropropagated plants in the field, although the later underwent significant changes during acclimatization. Micromorphological changes on leaf surface characters from in vitro to acclimatized plantlets were studied in terms of development of glandular trichomes, changes in vein spacing and vein structure in order to understand the nature of plant responses towards environmental conditions. The method developed and defined can be applied for commercial cultivation, which may be important for extraction of bioactive compounds and may facilitate conservation of this multipurpose endangered medicinal shrub.

 Aim: The current study aims to investigate the toxicity profile, the anti-amyloidogenic, and anti-ameliorative effects of crude ethanol extract from Cadaba indica leaves in contrast with a prominent drug. Methods: Phytochemical screening of the ethanol extract of C. indica was performed by GC–MS analysis. The cell viability of SH-SY5Y cells was assessed by a neutral red uptake assay, and neuroprotective effects were evaluated against the Aβ25–35 toxicity in SH-SY5Y cells experienced pretreatment with plant extracts. In acute and sub-chronic toxicity studies were conducted according to the OECD guidelines to ascertain the toxicity profile in Swiss Albino mice. The ameliorative effect of the extract was measured by behavioral test on drug-induced learning and memory impairment in mice. Results: Fifteen phytochemical compounds were revealed in GC–MS. Inhibitory concentration (IC50) was found to be >100 μg/mL on SH-SY5Ycells. Protection of SH-SY5Y cells from Aβ25-35-induced toxicity at the concentration of 25 μg/mL was observed. No clinical toxicity signs were observed in the plant treated group when compared to the control group in both acute and sub-chronic toxicity studies at the maximum of 2000 mg/kg dose. The oral lethal median dose (LD50) was estimated to be >2000 mg/kg. There was a reduction of the scopolamine-induced amnesia observed in the passive avoidance test at 100 mg/kg. Conclusion: The results conclude that C. indica extracts might contain potent secondary metabolites that might primarily enhance the neuroprotective effects and improve in cognitive impairments induced by cholinergic dysfunction.

In literature Review found that the Methanolic extract of Cadaba fruticosa shows highest Antimicrobial Activity as compare to Aquoes extract. Silver Nanoparticle also prepared which give higher Antimicrobial activity. Anticancer activity of plant studied against HeLa Cancer Cell Lines. Leaf juice of plant having various effect such as, used in Dysentery, Cough and Lungs problem, worm infection syphilis, Fever. Also used for general weakness, Poisoning, Deobstruent, Emmenagogue. The plant also found heavy metals, minerals and trace elements.

Cadaba fruticosa Druce: Medicinal plant

This study evaluates the efficacy of phytotherapy in treating oral cancer. This study evaluated the phytocompounds extracted from Cadaba fruticosa for anticancer potential using GC–MS analysis. A total of 10 bioactive phytocompounds were identified, of which 3,5-dichloro benzyl propyl ester emerged as a highly potent candidate. A pharmacokinetic analysis was performed using SwissADME, and the selected compound did not adhere to Lipinski’s rule of five, indicating no toxicity. A network pharmacology and molecular docking study was conducted to investigate the interaction between these compounds and the target proteins of oral cancer. The Kyoto Encyclopedia of Genes and Genomes (KEGG) database provided 10 hub genes associated with critical pathways: EGFR, MAPK3, PPARG, MTOR, KRAS, MDM2, ERBB2, IRS1, HRAS, and JAK2. Molecular docking studies confirmed strong binding interactions between 3,5-dichlorobenzyl propyl ester and the oral cancer protein SMAD, which had binding energy of -7.9 kcal/mol and interactions with the amino acids TYR280 (2.89 Å), ARG285 (3.04 Å), THR413 (3.21 Å), ARG415 (2.98 Å) and SER276 (2.09 Å), TYR280 (3.74 Å), THR289 (4.09 Å), ARG285 (4.31 Å), ARG415 (3.63 Å), ARG410 (3.54 Å), HIS441 (5.07 Å), PRO360 (4.45 Å), ARG410 (4.46 Å), ARG410 (4.93 Å) and ALA278 (4.34 Å). A molecular dynamics simulation was performed over 200 ns and showed the complex stable, with RMSD values ranging from 0.2 to 0.8 nm. Apoptotic gene expression analysis of KB-1 oral cancer cell lines showed downregulation of Bcl-2 and upregulation of pro-apoptotic genes BAD, BAX, and caspase-3 at an IC50 of 38.60 µg/ml. In a density functional theory (DFT) analysis, 3,5-dichloro benzyl propyl ester was found to have HOMO and LUMO energy levels ranging from − 0.8100 to 0.2701, highlighting its electronic properties. The RDG scattered plot topological analysis indicated significant Van der Waals interactions and steric repulsions. Accordingly, the present study concludes that 3,5-dichloro benzyl propyl ester is a promising new drug for treating oral cancer.

This communication is aimed to document the traditional medicinal uses of Cadaba fruticosa (L.) Druce and to propose conservation strategies for rare species occurring Nalgonda district, Andhra Pradesh. A rare plant species Cadaba fruticosa (L.) Druce has been locally used to treat by the indigenous people since time immemorial and needs immediate attention for conservation. Some of these are believed to promote positive health and maintain organic resistance against infection by re-establishing body equilibrium and conditioning the body tissues. The folk use of plants in medicinal value is an important part of the health care system. Cadaba fruticosa (L.) Druce has been recorded as a medicinal plant in Indian traditional system, while present investigations helpful for folklore medicinal importance, Animal health care, phytochemical screening and pharmacological activities. The present paper aims to explore the ethnomedicinal importance. Ethnomedicinal uses of Cadaba fruticosa (L.) Druce practiced by villagers of Nalgonda district and compared with the medicinal uses of valid publication.
