SSR analysis of genetic diversity and structure of the germplasm of faba bean (Vicia faba L.).

Faba bean (Vicia faba L.) is one of the most important legume crops in Egypt. However, production of faba bean is affected by several diseases including fungal diseases. Fusarium wilt incited by Fusarium oxysporum Schlecht. was shown to be the most common wilt disease of faba bean in Assiut Governorate. Evaluation of 16 faba bean genotypes for the resistance to Fusarium wilt was carried out under greenhouse conditions. Three molecular marker systems (inter-simple sequence repeat [ISSR], sequence related amplified polymorphism [SRAP], and simple sequence repeat [SSR]) and a biochemical marker (protein profiles) were used to study the genetic diversity and detect molecular and biochemical markers associated with Fusarium wilt resistance in the tested genotypes. The results showed that certain genotypes of faba bean were resistant to Fusarium wilt, while most of the genotypes were highly susceptible. The percentage of disease severity ranged from 32.83% in Assiut-215 to 64.17% in Misr-3. The genotypes Assiut-215, Roomy-3, Marut-2, and Giza-2 were the most resistant, and the genotypes Misr-3, Misr-1, Assiut-143, Giza-40, and Roomy-80 performed as highly susceptible. The genotypes Assiut-215 and Roomy-3 were considered as promising sources of the resistance to Fusarium wilt. SRAP markers showed higher polymorphism (82.53%) compared with SSR (76.85%), ISSR markers (62.24%), and protein profile (31.82%). Specific molecular and biochemical markers associated with Fusarium wilt resistance were identified. The dendrogram based on combined data of molecular and biochemical markers grouped the 16 faba bean genotypes into three clusters. Cluster I included resistant genotypes, cluster II comprised all moderate genotypes and cluster III contained highly susceptible genotypes.

Genetic Diversity Studies and Identification of Molecular and Biochemical Markers Associated with Fusarium Wilt Resistance in Cultivated Faba Bean (Vicia faba).

Breeding programs need to work with magnitudes of genetic variation in order to achieve the best results. In this study, we have estimated the genetic diversity and evaluated the field performance of 18 faba bean genotypes from different origins, during the 2014–2015 and 2015–2016 growing seasons. The field evaluations revealed that the faba bean genotypes showed enough variation for their agronomic thoughtful parameters. ALB was noted as being the genotype that was the tallest (126.25 cm), but the superlative genotypes were L251 and L252 for a high number of seeds per plant (72.25) and the maximum seed yield per plant (56.40 g). The estimations of the genetic diversity between all of the faba bean genotypes were done using eight sequence-related amplified polymorphism (SRAP)-combined primers. The number of different alleles amplified for each polymorphic marker, ranged from 2 to 9, was observed by SRAP genotyping, with polymorphic information contents (PIC) between 0.445and 0.896, and discrimination power (DP) with an average of 0.125 per allele. Across both years, the best grain yields were gathered from the L25, L252, and L182 genotypes, in descending order, whereas the ALB genotype had the lowest grain yield in comparison with the other genotypes. The cluster analysis of faba bean genotypes was generally based on not only the origin but also the genetic background.

Evaluation of the genetic variability of faba bean ( Vicia faba L.) genotypes using agronomic traits and molecular markers

The Multi-Trait selection index (MTSI) has been used to select highly productive and stable faba bean landraces across various traits. Fifteen productive, morphological, and phenological traits with desired and undesired selection gains (SGs) were evaluated in fifteen faba bean landraces under Mediterranean field conditions between 2019–2021. According to our results, most of the faba bean traits are influenced by the genotype × environment interaction (GEI) effects. Banner length (BL), seed yield (SY), diameter (D), number of flowers per inflorescence (NFI), branch number (BRN), and 100 seed weight (HSW) were found to be highly influenced by the environment, the proportions of variance explained by environmental factors were arranged between 83.21% and 74.08%, and only seed number per pod (SNP) was the highest genotypic effect (54.02%). According to the MTSI index, only three landraces (Gaada Mascara (G4), Oued zentai (G13), and Khmakhem (G15)) were considered to be highly yield-productive and to have a broad adaptability across Mediterranean environments. When numerous traits were evaluated, the MTSI index provided a strong and powerful tool for developing improved selection techniques for Faba bean studies.

Evaluation of Adaptability of Different Faba Bean Landraces under Mediterranean Field Conditions of Central-Northern Algeria

The success of the breeding programmes depends on the dimension of genetic variability. Simple sequence repeats (SSR) have been extensively used in the studies including genetic characterization of many plant species. The objective of this study was to investigate the genetic variation in 22 faba bean genotypes, 18 of which were originated from International Center for Agricultural Research in the Dry Areas (ICARDA) and 4 of which were cultivated genotypes in Turkey, using SSR markers. Of 41 SSR markers used, 25 produced bands. As a result of SSR amplification, a total of 39 bands, 25 of which were polymorphic and 14 of which were monomorphic, were obtained. The mean gene diversity and polymorphism information content values were 0.27 and 0.24, respectively. The faba bean genotypes cultivated in Turkey had greater genetic diversity than those that originated from ICARDA.
