Summary Theeffect ofthedopamine agonist, bromocriptine, andthedopamine antagonist, metoclopramide, on glucose tolerance wasexamined inmaturity onset diabetics andnormalsubjects. Afterbromocriptine therewasa lowered fasting bloodglucose and improved glucose tolerance inthediabetics. The controls showedaninitial improvement inglucose tolerance. A reduction ininsulin levels andamarked fall inplasma prolactin wasobserved inallsubjects. Growthhormoneconcentrations werelowandunaffected inthediabetics, andthere wasaninconsistent rise inthenormals. After metoclopramide there was someimpairment inglucose tolerance inthenormal subjects, butthis wasnotsignificant inthediabetics. Therewasalsoa markedprolactin release inall subjects, butnosignificant effect oninsulin orgrowth hormone. Itisconcluded thatbromocriptine lowers thebloodglucose andimproves glucose tolerance in maturity onset diabetics. This action maybetheresult ofthelowered prolactin whichoccurred without aconcomitant rise ingrowth hormone.
