Increased antitumor activity, intratumor paclitaxel concentrations, and endothelial cell transport of cremophor-free, albumin-bound paclitaxel, ABI-007, compared with cremophor-based paclitaxel.
Neil Desai,Vuong Trieu,Zhiwen Yao,Leslie Louie,Sherry Ci,Andrew Yang,Chunlin Tao,Tapas De,Bridget Beals,Donald J. Dykes,Patricia E. Noker,Rosie Yao,Elizabeth Labao,Michael M. Hawkins,Patrick Soon-Shiong +14 more
TL;DR: Endothelial binding and transcytosis of paclitaxel were markedly higher for ABI-007 versus Cremophor-based pac litaxel, and this difference was abrogated by a known inhibitor of endothelial gp60 receptor/caveolar transport.

Comparative Preclinical and Clinical Pharmacokinetics of a Cremophor-Free, Nanoparticle Albumin-Bound Paclitaxel (ABI-007) and Paclitaxel Formulated in Cremophor (Taxol)
Alex Sparreboom,Charity D. Scripture,Vuong Trieu,Paul J. Williams,Tapas De,Andrew Yang,Bridget Beals,William D. Figg,Michael J. Hawkins,Neil Desai +9 more
TL;DR: The volume of distribution at steady state and clearance for paclitaxel formulated as Cremophor-free nanoparticle ABI-007 were significantly greater than those for paclitexel formulated with Cremophileor (Taxol) in rats, consistent with the absence of pac litaxel-sequestering Cremphor micelles after administration of A BI-007.

Molecular mass distribution of sodium alginate by high-performance size-exclusion chromatography.
Sherry X Ci,Tanya H Huynh,Leslie Louie,Andrew Yang,Bridget Beals,Nilesh Ron,Wen-Ghih Tsang,Patrick Soon-Shiong,Neil P. Desai +8 more
TL;DR: A sensitive high-performance size-exclusion chromatography method with simple UV detection was developed for the molecular mass analysis of sodium alginate and revealed the effect of chemical composition of a polysaccharide and its effect on apparent molecular mass distribution.

Nanoparticle albumin-bound (nab) rapamycin as an anticancer agent
Tapas De,Vuong Trieu,Zachary Yim,Jon Cordia,Andrew Yang,Bridget Beals,Sherry Ci,Leslie Louie,Neil Desai +8 more
TL;DR: Nab-Rapamycin was well tolerated, showed linear PK, and was highly effective against a number of tumor models in vivo, which is typical for nab technology, e.g. nab-paclitaxel and nab-docetaxel.

Pharmacokinetic and ADME study of nanoparticle albumin-bound 17AAG (nab-17AAG) in mice
TL;DR: Application of nab technology to 17AAG resulted in a formulation of 17AAGs with the expected characteristics of albumin-bound nanoparticles, inclusive of a linear PK profile with respect to dose and large volume of distribution.
