The critical role of dysregulated epigenetic pathways in cancer genesis, development, and therapy has typically been established as a result of scientific and technical innovations in next generation sequencing. RNA interference, histone modification, DNA methylation and chromatin remodelling are epigenetic processes that control gene expression without causing mutations in the DNA. Although epigenetic abnormalities are thought to be a symptom of cell tumorigenesis and malignant events that impact tumor growth and drug resistance, physicians believe that related processes might be a key therapeutic target for cancer treatment and prevention due to the reversible nature of these processes. A plethora of novel strategies for addressing epigenetics in cancer therapy for immuno-oncological complications are currently available - ranging from basic treatment to epigenetic editing. - and they will be the subject of this comprehensive review. In this review, we cover most of the advancements made in the field of targeting epigenetics with special emphasis on microbiology, plasma science, biophysics, pharmacology, molecular biology, phytochemistry, and nanoscience. 

Novel epigenetic therapeutic strategies and targets in cancer.

Background: Although previous studies have identified the cervical cancer biomarkers by utilizing gene expression omnibus (GEO) expression datasets, they are limited in their application because of the heterogeneity-specific natures. Therefore, the present study was initiated to discover the GEO-based cervical cancer biomarkers that could employ over the heterogeneity barrier. Methods: Initially, the already reported cervical cancer-associated hub genes were mined and extracted through the literature search. Then, a protein-protein interaction network was developed and analyzed to discover six more closely cervical cancer-linked hub genes (real hub genes). Later, a comprehensive bioinformatics approach was applied to determine the diagnostic and prognostic potential of the real hub genes in cervical cancer patients of different clinicopathological features. Results: From a pool of 110 collected hub genes, in total 6 genes (CDK1, CCNB1, topoisomerase 2 alpha, checkpoint kinase 1, AURKA, and CDC6) were regarded as the real hub genes. Expression analysis revealed the significant upregulation of the real hub genes in cervical cancer patients of different races, cancer stages, body weights, and age groups. Further, correlational analyses have shown different interesting correlations between the expression and various other parameters of the real hub genes including promoter methylation status, genetic alteration, overall survival duration, tumor purity, and CD8+ T immune cells infiltration. Finally, related to hub genes, we have also identified potential miRNAs and chemotherapeutic drugs showing a great therapeutic potential. Conclusion: We have discovered a set of six real hub genes that might be utilized as new biomarkers for cervical cancer patients of different clinicopathological characteristics, overcoming the heterogeneity barrier.

In dogs, mammary cancer is the most common tumor type, especially in unspayed females. As in humans, this type of cancer has spontaneous development and is influenced by several risk factors, such as age and hormonal exposure in addition to genetic and epigenetic factors. Epigenetic mechanisms are responsible for gene expression modulation without alterations in the DNA sequence and include but are not limited to DNA methylation, histone modifications, and noncoding RNAs. Epigenetic patterns are known to influence a variety of biological mechanisms, such as cellular differentiation and development, and dysregulations of those patterns may result in several diseases, such as cancer. In this respect, this review summarizes the main findings concerning epigenetic alterations in canine mammary cancer, their relationship with the carcinogenic process, and their use as diagnostic and prognostic markers.

Cancer is a major global health challenge for our health system, despite the important pharmacological and therapeutic discoveries we have seen since past 5 decades. The increasing prevalence and mortality of cancer may be closely related to smoking, exposure to environmental pollution, dietary and genetic factors. Despite significant promising discoveries and developments such as cell and biotechnological therapies a new breakthrough in the medical field is needed to develop specific and effective drugs for cancer treatment. On the development of cell therapies, anti-tumor vaccines, and new biotechnological drugs that have already shown promising effects in preclinical studies. With the continuous enrichment and development of chromatin immunoprecipitation sequencing (ChIP-seq) and its derivative technologies, epigenetic modification has gradually become a research hotspot. As key ingredients of epigenetic modification, Writers, Readers, Erasers have been gradually unveiled. Cancer has been associated with epigenetic modification especially methylation and therefore different epigenetic drugs have been developed and some of those are already undergoing clinical phase I or phase II trials, and it is believed that these drugs will certainly assist the treatment in the near future. With respect to this, an overview of anti-tumor drugs targeting modified enzymes and de-modified enzymes will be performed in order to contribute to future research.
