Fungal phytases are histidine acid phosphatases, a subclass of acid phosphatases, which catalyse the hydrolysis of phytic acid resulting in the release of phosphate moieties and thus mitigate its antinutritional properties. The supplementation of feed with phytases increases the bioavailability of phosphorus and minerals in non-ruminant animals and reduces the phosphorus pollution due to phosphorus excretion in the areas of intensive livestock production. Although phytases are reported in plants, animals and micro-organisms, fungal sources are used extensively for the production of phytases on a commercial scale. Phytases have been produced by fungi in both solid-state fermentation (SSF) and submerged fermentation (SmF). The fungal phytases are high molecular weight proteins ranging from 35 to 500 kDa. They are optimally active within pH and temperature ranges between 4.5 and 6.0, and 45 and 70 °C respectively. Phytate degradation leads to amelioration in the nutritional status of foods and feeds by improving the availability of minerals, phosphorus and proteins in non-ruminant animals and human beings and thus mitigates the environmental phosphorus pollution. Our article focuses on the role of fungal phytases in improving nutritional value of foods and feeds with concomitant increase in growth of non-ruminant animals and mitigating environmental phosphorus pollution.

Fungal phytases: characteristics and amelioration of nutritional quality and growth of non-ruminants.

Phytates have been considered as a threat in human diet due to its antinutrients behaviour which known as strong chelators of divalent minerals such as Ca²+, Mg²+, Zn²+ and Fe²+. Phytic acid has a potential for binding positively charged proteins, amino acids, and/or multivalent cations or minerals in foods. The resulting complexes are insoluble, difficult for humans to hydrolyze during digestion, and thus, typically are nutritionally less available for absorption. The reduction of this phytates can be achieved through both enzymatic and non-enzymatic removal. Enzymatic degradation includes addition of either isolated form of wild-type or recombinant exogenous phytate-degrading enzymes microorganisms in the food matrix. Non-enzymatic hydrolysis of phytate occurred in the final food during food processing or physical separation of phytate-rich parts of the plants seed. The application of phytase with respect to breadmaking process, probiotics, animal feed supplement and transgenic crops are emphasised in this paper.

Phytase: application in food industry

The marine environment is one of the most important sources of bioactive compounds in the world. Marine microbial communities from marine environments secrete variety enzymes based on their habitat and their ecological roles. They are considered important ecological components in marine environments due to their performance in biogeochemical processes. Marine microbial enzymes including protease, lipase, collagenase, agarase, cellulose and Tother enzymes are important in different industrial due to their properties and applications. The biochemical diversity of marine microbial makes them reasonable sources of a wide variety of enzymes for use in different industrial including food, medicine and other biotechnological systems. Therefore, marine microbial enzymes preparations due to a special applications and functions in the biotechnology projects, also these can suggestion novel catalysts of biological and biochemical reactions with unique properties and functions. In this review, we focus on microbial enzymes from marine derived and their biochemical significance and their increasing importance in biotechnology.

Purification and biochemical characterization of an Aspergillus niger phytase produced by solid-state fermentation using triticale residues as substrate.

Phytases have gathered considerable attention in recent years due to their applications in human and animal nutrition, environmental protection and biotechnology. Phytases were isolated and characterized from a variety of bacteria and fungi but rarely from actinomycetes. In the present study, a Nocardia sp. MB 36, isolated from rice mill waste, was found to be a producer of a thermostable and acid/alkalistable phytase. Optimization of phytase production by Plackett–Burman designed experiments indicated that wheat bran, medium pH, FeSO4 and CaCl2 are the most effective fermentation variables that influence the phytase yield under submerged fermentation. Further optimization of these four variables by using Closed-Composite-Design of response surface methodology resulted in enhancement of phytase production by 64.14%. This newly isolated phytase from Nocardia sp. has potentially desirable features for animal feed applications.

Studies on the dephosphorylation of phytic acid in livestock feed using phytase from Aspergillus niger van Teighem

LC-MS technique described here is a new way for the separation and direct determination of UV–Vis insensitive inositol phosphates (InsP2-InsP6). This circumvents the need of radioisotopic labeling and post-column derivatization techniques. The method involves separation of various enzymatically dephosphorylated derivatives of InsP6 on C18-column using MeOH/H2O (30:70 v/v) and their identification using electron spray ionization MS in positive ion mode (+pESI-MS). The LC-MS studies revealed that the purified phytase from Aspergillus niger van Teighem hydrolyzes InsP6 in a sequential manner leading to InsP2 (InsP2·2Na, tR 4.4–4.54 min, base peak m/z 382.9) as the end product.
