The invention relates to the mda-7 gene, its encoded protein and fragments of the proteins. Several of these fragments of the MDA-7 protein exhibit antiproliferative activity and/or inhibited the activity of intact MDA-7. Accordingly, the invention provides, among other things, for methods and compositions that may be used in the treatment of disorders of cell proliferation, including cancer.

The present invention discloses the differential expression of the minor vault protein ADPRTL gene in specific brain regions of Alzheimer's disease patients. Based on this finding, this invention provides a method for diagnosing or prognosticating Alzheimer's disease in a subject, or for determining whether a subject is at increased risk of developing Alzheimer's disease. Furthermore, this invention provides therapeutic and prophylactic methods for treating or preventing Alzheimer's disease and related neurodegenerative disorders using a gene coding for a vault protein, in particular the gene coding for the minor vault protein ADPRTL1. A method of screening for modulating agents of neurodegenerative diseases is also disclosed.

Disclosed are nucleic acid molecules encoding a protein having poly(ADP-ribose) polymerase (PARP) activity as well as the encoded protein. Furthermore, the invention describes expression vectors, host cells, antibodies, pharmaceutical compositions and methods for treating disorders associated with aberrant regulation of cellular behaviour. Finally, screening methods are described for compounds that act as agonist or antagonist of the protein having PARP activity.
