Overcoming Acquired Resistance to AZD9291, A Third-Generation EGFR Inhibitor, through Modulation of MEK/ERK-Dependent Bim and Mcl-1 Degradation.
Puyu Shi,Puyu Shi,You-Take Oh,Liang Deng,Guojing Zhang,Guoqing Qian,Shuo Zhang,Shuo Zhang,Hui Ren,Grant Wu,Benjamin Legendre,Emily Anderson,Suresh S. Ramalingam,Taofeek K. Owonikoko,Mingwei Chen,Shi-Yong Sun +15 more
TL;DR: Modulation of MEK/ERK-dependent Bim and Mcl-1 degradation critically mediates sensitivity and resistance of EGFR-mutant NSCLC cells to AZD9291 and hence is an effective strategy to overcome acquired resistance to AZd9291.

Kit and method for sequencing a target dna in a mixed population
Katherine Richardson,Benjamin Legendre,Yanggu Shi +2 more
- 28 Feb 2012
TL;DR: In this paper, the authors present methods and kits for sequencing a target DNA sequence in a sample having a related reference sequence, and methods for detecting mitochondrial mutations and for differentiating between closely related viral strains.

Abstract 5438: Multiplexed ICE COLD-PCR coupled to NGS and ddPCR enables enhanced detection of low-level DNA mutations in tissues and liquid biopsies
Katherine Richardson,Sarah Statt,Grant Wu,Karissa Scott,Erin Montagne,Sheena Jensen,Courtney Cubrich,Phil Krzycki,Jason Stoddard,Amy Kruempel,Emily McCutchen,Stephanie Veys,Kylee Baughman,Sarah Cherubin,Vicki Rosendale,Jaclyn Pope,Paula Bartlett,Phil Eastlake,Stephanie Peterson,Benjamin Legendre +19 more
TL;DR: This work compared detection of low-level of mutations in limiting amounts of DNA, with or without the use of MX-ICP prior to NGS and ddPCR, using digitally verified chromosomal DNA mixtures from Horizon Diagnostics.

Characterization and identification of specific EGFR mutations in circulating tumor cells (CTCs) isolated from non-small cell lung cancer patients using antibody independent method, ApoStream.
Hai T. Tran,Vladislava O. Melnikova,Anne S. Tsao,Frank V. Fossella,Faye M. Johnson,Vassiliki A. Papadimitrakopoulou,Miguel Garza,Christopher L. Neal,Dave Hasegawa,Amy Kruempel,Grant Wu,Katherine Richardson,Marcia Lewis,Benjamin Legendre,Kenna Anderes,Darren W. Davis,John V. Heymach +16 more
TL;DR: The use of ApoStream is reported here the use of a novel dielectrophoresis field-flow-assisted, antibody-free method to isolate CTCs from blood to determine specific EGFR mutations from captured C TCs.

High sensitivity detection and robust enrichment for PIK3CA somatic mutations using ice COLD-PCR technology.
TL;DR: Improved and complete enrichment CO-amplification at lower denaturation temperature PCR (Ice COLD-PCR) technology is applied for high sensitivity detection and robust enrichment of any mutations present in a DNA sample, allowing for the enrichment of mutations any of the key mutations in the PIK3CA gene.
