Gold nanoparticles (GNPs) have generated keen interest among researchers in recent years due to their excellent physicochemical properties. In general, GNPs are biocompatible, amenable to desired functionalization, non-corroding, and exhibit size and shape dependent optical and electronic properties. These excellent properties of GNPs exhibit their tremendous potential for use in diverse biomedical applications. Herein, we have evaluated the recent advancements of GNPs to highlight their exceptional potential in the biomedical field. Special focus has been given to emerging biomedical applications including bio-imaging, site specific drug/gene delivery, nano-sensing, diagnostics, photon induced therapeutics, and theranostics. We have also elaborated on the basics, presented a historical preview, and discussed the synthesis strategies, functionalization methods, stabilization techniques, and key properties of GNPs. Lastly, we have concluded this article with key findings and unaddressed challenges. Overall, this review is a complete package to understand the importance and achievements of GNPs in the biomedical field.

Role of gold nanoparticles in advanced biomedical applications

Biomass-assisted fabrication of rGO-AuNPs as surface-enhanced Raman scattering substrates for in-situ monitoring methylene blue degradation.

Background: Homocysteine (Hcy) has been found to be closely related to the occurrence of diabetes mellitus (DM) and is considered as one of the risk factors of DM. However, Hcy alone is not enough as a factor to predict DM, and our study analyzed and determined the relationship between the main metabolites involved in the Hcy metabolic pathway and DM. Methods: A total of 48 clinical samples were collected, including 18 health control samples and 30 DM samples. All standards and samples were detected by LC-QTOF-MS. Multivariate statistical analysis and k-means cluster analysis were performed to screen and confirm the metabolites significantly correlated with DM. Results: A total of 13 metabolites of the Hcy metabolic pathway were detected in the samples. The content of Hcy, cysteine, taurine, pyridoxamine, methionine, and choline were significantly increased in the DM group (p < 0.05). Hcy, choline, cystathionine, methionine, and taurine contributed significantly to the probabilistic principal component analysis (PPCA) model. The odds ratios (OR) of Hcy, cysteine, taurine, methionine, and choline were all greater than one. K-means cluster analysis showed that the Hcy, taurine, methionine, and choline were significantly correlated with the distribution of glucose values (divided into four levels: 10.5–11.7 mmol/L, 7.7–9.7 mmol/L, 6.0–6.9 mmol/L, and 5.0–5.9 mmol/L, respectively). Conclusion: Hcy, taurine, methionine, and choline can be used as risk factors for diabetes diagnosis and are expected to be used for the assessment of diabetes severity.

Profiling of Homocysteine Metabolic Pathway Related Metabolites in Plasma of Diabetic Mellitus Based on LC-QTOF-MS

Simultaneous determination of total homocysteine, methionine, methylmalonic acid and 2-methylcitric acid in dried blood spots by ultra-performance liquid chromatography–tandem mass spectrometry

Immunochromatographic test strips (ICTS) are widely used for the immediate and rapid detection of toxins; however, they are prone to false-positive results and insufficient sensitivity when applied to complex sample matrices. To overcome these limitations, herein, a multifunctional signal probe-based ICTS was developed for the rapid detection of aflatoxin B1 (AFB1) in complex matrix samples by conjugating biotinylated nanobodies (G8-Bio) with streptavidin-modified magnetic beads (MB-SA) as the probes. The constructed ICTS achieved a limit of detection (LOD) of 0.063 ng/mL for AFB1. In corn spiked experiments, the recovery rates ranged from 98 % to 102 %, with coefficients of variation (CV) between 1.4 % and 6.86 %, confirming the reliability of this method. Furthermore, the IC10 value in soy sauce samples was 0.069 ng/mL, indicating its suitability for detecting AFB1 in dark-colored matrices. This approach provides a novel strategy for the rapid and accurate detection of trace toxins in complex matrix samples. 

Multifunctional signal probe-based immunochromatographic test strip for rapid detection of aflatoxin B1 in complex matrix samples

A simple, fast, and eco-friendly method for the biosynthesis of gold nanoparticles (AuNPs) using Lilium casa blanca petals as a reducing and protecting agent was reported. The heating time had been shortened to 30 min for biosynthesis of AuNPs, which only took 15 mg Lilium casa blanca petals powder. The biosynthetic AuNPs, which had a uniform particle size distribution, were in spherical with the average size of 5.933 ± 1.158 nm. In addition, the conversion rate of Au3+ to Au0 was quite high. Through the discussion on the nutrients' concentrations before and after the synthesis, it was found that sugars, alkaloids, flavonoids, and proteins were the reducing and stabilising agents. The biosynthetic AuNPs had good catalytic activity, taking the hydroboration of nitrophenol and methylene blue as examples. Catalytic reduction followed pseudo-first-order kinetics. Moreover, the apparent rate constant was a linear correlation with the concentrations of AuNPs. The normalised rate constant of the proposed AuNPs was higher than other reported AuNPs, which indicated the excellent catalytic activity. All the results foreshowed the wide range of applications of the biosynthetic AuNPs by Lilium casa blanca petals.

Homocysteine (Hcy) is an amino acid found in the human body and has the potential to predict cardiovascular risk where timely detection and treatment are very important. In this study, we established a method to combine enzymatic hydrolysis with a colloidal gold lateral flow immunoassay (LFIA) for the detection of total Hcy (tHcy) based on the monoclonal antibody (mAb) 3B3 against S-adenosyl-L-homocysteine (SAH). The 50% inhibitory concentration (IC50) for this mAb against SAH was 2.94 nM with a linear range of 0.73-11.75 nM. The developed LFIA could detect SAH with a vLOD and cLOD of 10 nM and 2.07 nM, respectively. For tHcy detection in serum samples, the LFIA strip was able to detect tHcy after enzymatic hydrolysis, and the results correlated well with that of an enzymatic cycling test kit.
