Plant tissue culture involves the culture of all types of plant cells, tissues and organs under aseptic conditions. This definition also extends to the culture of excised embryos and to protoplast culture. An overview of tissue culture techniques and their applications in plant propagation and genetic improvement of plants is presented. The areas under review include: (1) embyro culture, (2) meristem culture, (3) micropropagation, (4) somatic embryogenesis, (5) somaclonal variation, (6) in vitro selection, (7) anther culture and (8) protoplast culture. Problems and limitations of each of the techniques are also discussed. Examples are given of work that has been undertaken or that is currently in progress on the application of these techniques to the improvement of Queensland's subtropical horticultural industries. Key examples are: (1) embryo culture to facilitate incorporation of genes conferring disease-resistance from wild Cucurbita species into cultivated varieties, (2) meristem culture for virus elimination in strawberries (Fragaria × ananassa) and sweet potato (Ipomoea batatas), (3) micropropagation for rapid increase in new varieties of ginger (Zingiber officinale) and pineapple (Ananas comosus) to enable more rapid field evaluation and early release, (4) micropropagation of disease-free, genetically uniform planting material of superior female papaya (Carica papaya) selections and banana (Musa spp.) selections and (5) the use of somaclonal variation and gamma-irradiation for the genetic improvement of banana. Finally, future opportunities for the utilisation of tissue culture in plant propagation and improvement in Queensland's horticultural industries are summarised.

Though there is increasing recognition of the cultural dimensions that shape climate change adaptation, our experience from working with actors engaged in adaptation policy and practice suggests that the role of culture still tends to be conceived in overly narrow and fixed terms. This is exemplified in portrayals of conservative cultural norms as stifling positive change. A growing body of research across the world indicates that the reality is seldom as simple as this – culture works in complex and variable ways, and, most importantly, is inherently dynamic. Drawing especially from research work on vulnerability and adaptation conducted in semi-arid regions, we illustrate this argument by briefly exploring three themes - multiple knowledge systems for farming in Botswana, the dynamics of pastoralist values and livelihoods in Kenya, and the interplay of caste and livelihood choices in India. Understanding how different facets of culture such as these operate in context helps move away from viewing culture statically as a barrier or enabler, and toward a more plural and dynamic appreciation of the role of culture in adaptation. This includes recognising the potential for factors that may be construed as barriers to become enablers. Critical, balanced engagement with cultural dimensions in both research and practice, understanding and working with these dynamic social structures, is essential if adaptation is to create meaningful and lasting change for those who need it most.

Culture as a mediator of climate change adaptation: Neither static nor unidirectional

Smallholder agriculture in Northwestern Ghana continues to suffer the increasing threats of climate change and variability. The extant literature has argued that climate-smart agriculture is the way forward for smallholder farmers to reduce the threats of climate change and variability in agriculture production. However, smallholder farmers continue to rely on indigenous knowledge and practices in their day-to-day agricultural activities. Few studies have explored the rationale and factors that explain smallholder farmers choice of local agriculture practices. This study explored the rationale and factors that explain smallholder farmers' choice of indigenous knowledge and agriculture practices. The mixed research method approach involving both quantitative and qualitative methods were employed for data collection and analysis. A survey, involving 305 household heads, 31 in-depth interviews and 18 focus group discussions were held with key participants for the data. The results showed that smallholder farmers' decisions to adopt indigenous practices for climate change adaptation were influenced by socio-demographic characteristics, access to farm capital, landscape and distance to farms, accessibility and reliability of practices, accessibility and cost of inputs, land tenure, access to extension services, and socio-cultural beliefs. These variables were statistically significant at 5 %. The paper concludes that these factors will continue to limit farmers' ability to adopt climate-smart and other improved agricultural practices. This will aggravate smallholder households' vulnerability to food insecurity and poverty. It is, therefore, recommended that climate-smart agriculture practices should be framed within the context of the aforementioned factors influencing farmers choice of indigenous farming practices in mainstreaming them into climate-smart agriculture.
