Digital holography is an emerging field of new paradigm in general imaging applications. We present a review of a subset of the research and development activities in digital holography, with emphasis on microscopy techniques and applications. First, the basic results from the general theory of holography, based on the scalar diffraction theory, are summarized, and a general description of the digital holographic microscopy process is given, including quantitative phase microscopy. Several numerical diffraction methods are described and compared, and a number of representative configurations used in digital holography are described, including off-axis Fresnel, Fourier, image plane, in-line, Gabor, and phase-shifting digital holographies. Then we survey numerical techniques that give rise to unique capabilities of digital holography, including suppression of dc and twin image terms, pixel resolution control, optical phase unwrapping, aberration compensation, and others. A survey is also given of representative application areas, including biomedical microscopy, particle field holography, micrometrology, and holographic tomography, as well as some of the special techniques, such as holography of total internal reflection, optical scanning holography, digital interference holography, and heterodyne holography. The review is intended for students and new researchers interested in developing new techniques and exploring new applications of digital holography.

Principles and techniques of digital holographic microscopy

A novel method is proposed for simulating free-space propagation. This method is an improvement of the angular spectrum method (AS). The AS does not include any approximation of the propagation distance, because the formula thereof is derived directly from the Rayleigh-Sommerfeld equation. However, the AS is not an all-round method, because it produces severe numerical errors due to a sampling problem of the transfer function even in Fresnel regions. The proposed method resolves this problem by limiting the bandwidth of the propagation field and also expands the region in which exact fields can be calculated by the AS. A discussion on the validity of limiting the bandwidth is also presented.

The numerical calculation of the Rayleigh-Sommerfeld diffraction integral is investigated. The implementation of a fast-Fourier-transform (FFT) based direct integration (FFT-DI) method is presented, and Simpson's rule is used to improve the calculation accuracy. The sampling interval, the size of the computation window, and their influence on numerical accuracy and on computational complexity are discussed for the FFT-DI and the FFT-based angular spectrum (FFT-AS) methods. The performance of the FFT-DI method is verified by numerical simulation and compared with that of the FFT-AS method.

Fast-Fourier-transform based numerical integration method for the Rayleigh-Sommerfeld diffraction formula

Optical examination of microscale features in pathology slides is one of the gold standards to diagnose disease. However, the use of conventional light microscopes is partially limited owing to their relatively high cost, bulkiness of lens-based optics, small field of view (FOV), and requirements for lateral scanning and three-dimensional (3D) focus adjustment. We illustrate the performance of a computational lens-free, holographic on-chip microscope that uses the transport-of-intensity equation, multi-height iterative phase retrieval, and rotational field transformations to perform wide-FOV imaging of pathology samples with comparable image quality to a traditional transmission lens-based microscope. The holographically reconstructed image can be digitally focused at any depth within the object FOV (after image capture) without the need for mechanical focus adjustment and is also digitally corrected for artifacts arising from uncontrolled tilting and height variations between the sample and sensor planes. Using this lens-free on-chip microscope, we successfully imaged invasive carcinoma cells within human breast sections, Papanicolaou smears revealing a high-grade squamous intraepithelial lesion, and sickle cell anemia blood smears over a FOV of 20.5 mm(2). The resulting wide-field lens-free images had sufficient image resolution and contrast for clinical evaluation, as demonstrated by a pathologist's blinded diagnosis of breast cancer tissue samples, achieving an overall accuracy of ~99%. By providing high-resolution images of large-area pathology samples with 3D digital focus adjustment, lens-free on-chip microscopy can be useful in resource-limited and point-of-care settings.

A fast and accurate numerical method for free-space beam propagation between arbitrarily oriented planes is developed. The only approximation made in the development of the method was that the vector nature of light was ignored. The method is based on evaluating the Rayleigh–Sommerfeld diffraction integral by use of the fast Fourier transform with a special transformation to handle tilts and offsets of planes. The fundamental aspects of a software package based on the developed method are presented. A numerical example realized with the software package is presented to establish the validity of the method.
