As polyphagous, holometabolous insects, tephritid fruit flies (Diptera: Tephritidae) provide a unique habitat for endosymbiotic bacteria, especially those microbes associated with the digestive system. Here we examine the endosymbiont of the olive fly [Bactrocera oleae (Rossi) (Diptera: Tephritidae)], a tephritid of great economic importance. "Candidatus Erwinia dacicola" was found in the digestive systems of all life stages of wild olive flies from the southwestern United States. PCR and microscopy demonstrated that "Ca. Erwinia dacicola" resided intracellularly in the gastric ceca of the larval midgut but extracellularly in the lumen of the foregut and ovipositor diverticulum of adult flies. "Ca. Erwinia dacicola" is one of the few nonpathogenic endosymbionts that transitions between intracellular and extracellular lifestyles during specific stages of the host's life cycle. Another unique feature of the olive fly endosymbiont is that unlike obligate endosymbionts of monophagous insects, "Ca. Erwinia dacicola" has a G+C nucleotide composition similar to those of closely related plant-pathogenic and free-living bacteria. These two characteristics of "Ca. Erwinia dacicola," the ability to transition between intracellular and extracellular lifestyles and a G+C nucleotide composition similar to those of free-living relatives, may facilitate survival in a changing environment during the development of a polyphagous, holometabolous host. We propose that insect-bacterial symbioses should be classified based on the environment that the host provides to the endosymbiont (the endosymbiont environment).

The olive fly (OLF), Bactrocera oleae (Rossi), is an invasive tephritid fruit fly that causes extensive damage to olive crops around the world (especially in the Mediterranean basin and North America). Previous attempts to use the Sterile Insect Technique (SIT) for the OLF were not successful because of the inability to rear high quality OLF in the laboratory on an artificial diet. New improvements in rearing methods and additional understanding of the basic biology of the OLF have led to a renewal of interest in using SIT for OLF. This review discusses the history, difficulties, improvements and future directions of OLF mass-rearing. Issues include: the design of cages and oviposition substrates, cost and quality of artificial diets, maintenance of endosymbiotic microbiota, control of pathogenic microbes, collection of pupae, the fitness of adults, and the competitiveness of sterilized laboratory males released to the field.

A basis for the renewal of sterile insect technique for the olive fly, Bactrocera oleae (Rossi)

The oriental fruit fly Bactrocera dorsalis (Hendel) is a destructive insect pest of a wide range of fruits and vegetables. This pest is an invasive species and is currently distributed in some provinces of China. To recover the symbiotic bacteria of B. dorsalis from different invasion regions in China, we researched the bacterial diversity of this fruit fly among one laboratory colony (Guangdong, China) and 15 wild populations (14 sites in China and one site in Thailand) using DNA-based approaches. The construction of 16S rRNA gene libraries allowed the identification of 24 operational taxonomic units of associated bacteria at the 3% distance level, and these were affiliated with 3 phyla, 5 families, and 13 genera. The higher bacterial diversity was recovered in wild populations compared with the laboratory colony and in samples from early term invasion regions compared with samples from late term invasion regions. Moreover, Klebsiella pneumoniae and Providencia sp. were two of the most frequently recovered bacteria, present in flies collected from three different regions in China where B. dorsalis is invasive. This study for the first time provides a systemic investigation of the symbiotic bacteria of B. dorsalis from different invasion regions in China.

Bacterial communities associated with invasive populations of Bactrocera dorsalis (Diptera: Tephritidae) in China.

The olive fruit fly, Bactrocera oleae (Rossi) (Diptera: Tephritidae), is the major insect pest in commercial olive (Olea europaea L., Oleaceae) production worldwide. Its population management is largely based on the use of insecticides. However, concerns about the impact of insecticides on the environment and human health along with increasing resistance development calls for novel and environment-friendly approaches for population management. Integrated pest management programmes with a sterile insect technique (SIT) component and parasitoids are currently considered for the control of B. oleae. A major challenge for the development of such tools is mass rearing of both host and parasitoids. In this review, we consider the role of endogenous microbiota and its potential exploitation for improving the efficacy, quality, and cost effectiveness of mass rearing B. oleae as well as their parasitoids.

Review of the role of gut microbiota in mass rearing of the olive fruit fly, Bactrocera oleae, and its parasitoids

The applied importance of symbiosis has been gaining recognition. The relevance of symbiosis has been increasing in agriculture, in developing sustainable practices, including pest management. Insect symbiotic microorganisms’ taxonomical and functional diversity is high, and so is the potential of manipulation of these microbial partners in suppressing pest populations. These strategies, which rely on functional organisms inhabiting the insect, are intrinsically less susceptible to external environmental variations and hence likely to overcome some of the challenges posed by climate change. Rates of climate change in the Mediterranean Basin are expected to exceed global trends for most variables, and this warming will also affect olive production and impact the interactions of olives and their main pest, the obligate olive fruit fly (Bactrocera oleae). This work summarizes the current knowledge on olive fly symbiotic bacteria towards the potential development of symbiosis-based strategies for olive fruit fly control. Particular emphasis is given to Candidatus Erwinia dacicola, an obligate, vertically transmitted endosymbiont that allows the insect to cope with the olive-plant produced defensive compound oleuropein, as a most promising target for a symbiosis disruption approach.

Symbiosis in Sustainable Agriculture: Can Olive Fruit Fly Bacterial Microbiome Be Useful in Pest Management?

Microflora species and volatiles emitted from artificial diets were examined from the larvae of three homozygous alcohol dehydrogenase (Adh) strains of the olive fruit fly, Bactrocera (Dacus) oleae (Gmelin), reared under identical conditions. Differences in volatile composition were detected when Adh-I homozygous larvae developed in a diet lacking the preservative p-hydroxy-benzoic acid methyl ester (nipagin). Larval development of the Adh-I strain in the preservative-free diet was reduced by 50%, whereas pupal emergence was completely inhibited. The larval development and pupal emergence of Adh-F and Adh-S strains were not affected. Unique microorganisms with characteristic volatile profiles were isolated from the preservative-free diet of the Adh-I strain that were different from those, isolated from Adh-S, Adh-F, laboratory colony, and wild insect populations. Our results indicated that the variations in volatile composition of the artificial diets, and the inhibition of larval development and pupal emergence in Adh-I strain were related to changes in the microflora that developed in the diets of the Adh-I strain.
