Publisher Summary This chapter describes the structures of alkaloids and their biological activities, isolated from various plants of the genus Buxus during the period 1992–2008. Additionally, the semisynthesis and structural modifications of these alkaloids are also discussed. The genus Buxus is composed of various species including Buxussempervirens, B. papillosa, B. microphylla, B. hildebrandtii, and B. hyrcana. The crude extracts of these plants have been reported to have applications in traditional medicine to cure fatigue, rheumatism, malaria, depression, and skin infections. The ethanolic extract of B. sempervirens has been reported to exhibit anti-HIV activity, and it has also exhibited delayed progression in HIV-infected asymptomatic patients. Phytochemical studies on various plants of the genus Buxus have resulted in the isolation of over 200 new steroidal alkaloids. A few of these alkaloids have shown interesting biological activities, including antibacterial, antimyco-bacterial, antimalarial, and acetylcholinesterase (AChE) inhibitory activities. B. longifolia, commonly known as ‘‘English Box,’’ is widely distributed in Europe and Turkey. The crude extract of this plant has shown inhibition against serum cholinesterase of human and horse blood. Studies on structure-activity relationships of moderately bioactive compounds can be worthwhile in the discovery of new pharmaceuticals.
