In this study, we investigated the antifungal properties of an acetone extract derived from the leaves of Swietenia mahagoni (SMAL) against two isolated fungi, Fusarium equiseti (OQ820153) and Rhizoctonia solani (OQ820152), from rice sheath. The extract was effective in inhibiting the growth of both fungi at the highest concentration tested, 3000 μg·mL−1. Laboratory tests on the LC20 of SMAL extract (49.86 mg·L−1) versus pyriproxyfen 10% EC (1.96 mg·L−1) were accomplished on Aphis gossypii Glover. The extract potently reduced the survival of the nymphs (49.58%) more than the other treatments. The longevity of nymphs treated with the extract had the highest prolongation at 9.67 days. The olfactory choice test exhibited the lowest aphid attraction percentage (23.33%). The HPLC of SMAL extract contained various phenolic compounds, and the most abundant found were catechin (752.64 µg·g−1), gallic acid, and chlorogenic acid, as well as flavonoids such as rutin (585.24 µg·g−1) and naringenin. A GC–MS analysis revealed n-hexadecanoic acid (37.1%) as the major compound, followed by oleic acid. These results suggest that SMAL extract has the potential to help plants fight against fungal and insect infections, making it a promising natural and renewable solution for long-term plant pest regulation.

Swietenia mahagoni Leaves Extract: Antifungal, Insecticidal, and Phytochemical Analysis

Seventy-eight common bean (Phaseolus vulgaris L.) seed samples were collected from twenty-one field locations across six governorates (Ismailia, Sharkia, Port Said, South Sinai, North Sinai, and Suez) in the East Delta region of Egypt between 2021 and 2022. Thirty-five Rhizoctonia solani Kühn were isolated. Sharkia had the highest percentage of R. solani isolates (RS) from the collected samples (59%), followed by Ismailia, South Sinai, Suez, North Sinai, and Port Said (49.7%, 36.7%, 33%, 29.2%, and 27.8%, respectively). Pathogenicity tests proved that the seven highest-virulence strains of R. solani had a pre-emergence damping off ranging from 17% to 30% and a post-emergence damping off ranging from 11% to 20%. The seven most pathogenic isolates were identified using the Internal Transcribed Spacer (ITS) with code numbers as follows: R. solani RSA1, R. solani RIT2, R. solani RIF3, R. solani RSoR4, R. solani RNB5, R. solani RSzA6, and R. solani RPS11. Principle component analysis (PCA), which depends on the pathogenicity result and soil variables, revealed that soil texture and soil water holding capacity (MWHC) were the most effective factors in grouping and clustering the studied R. solani isolates from the different locations into two groups. From this study, it can be established that the biodiversity of R. solani in the East Delta area of Egypt depends on changes in the environmental conditions and soil variables. The soil type and pH variables have the greatest impact on R. solani diversity, followed by the humidity-holding ability and soil porosity.

Biodiversity of Rhizoctonia solani in Phaseolus vulgaris Seeds in East Delta of Egypt

Dermatophytes are notorious pathogens capable of infecting various mammals skin, posing serious threats to human health and overall life quality worldwide. Artemisia argyi has been recorded and applied for over a thousand years to treat skin itching. Although it has the potential to be developed as a plant-based antifungal agent, it's antifungal activity and action mechanism of active ingredients are still unclear. The aim of this study was to investigate the chemical composition, antifungal activity against skin fungi, and potential mechanisms of Artemisia argyi essential oil (AEO). The chemical composition of AEO was analyzed by gas chromatography-mass spectrometry (GC-MS) firstly. Flat growth restraint and double half dilution tests was performed to evaluate AEO antifungal activity against Microsporum gypseum, Trichophyton mentagrophytes, and Trichophyton rubrum. And then, the physiological mechanism of AEO inhibiting dermatophytes was systematically explored through scanning electron microscopy, relative conductivity, membrane leakage, ROS content, and antioxidant enzyme activity. Finally, the main pathways were screened through transcriptome sequencing, while the related genes expression levels and enzyme activity were validated. Monoterpenes and sesquiterpenoids were the most highly representative class of AEO. AEO had powerful antifungal activity against M. gypseum, T. mentagrophytes, and T. rubrum, with minimum inhibitory concentration (MIC) values of 0.6, 1.2, and 1.2 μL/mL, respectively. Moreover, AEO can also damage the cell membrane integrity of T. mentagrophytes, resulting in cellular extravasation of intracellular substances. Transcriptome analysis revealed that the main target of AEO is to inhibit electron transfer and oxidative phosphorylation during respiration, ultimately leading to obstruction of normal ATP synthesis and energy metabolism in mitochondria. And a large amount of ROS will generate due to the incompletely catalysis of oxygen under mitochondrial complexes. Coupled with the decrease of antioxidant enzyme (SOD, POD) activity, excessive accumulation of ROS will cause serious oxidative damage to cells and eventually exhibiting antifungal activity against dermatophytes. The present study demonstrated that Artemisia argyi was a valuable source of active compounds with antifungal activity. These findings support AEO as a potential agent to inhibit dermatophytes and prevent related dermatophytoses.

The emergence of microplastics (MPs) as pollutants in agricultural soils is increasingly alarming, presenting significant threats to soil ecosystems. Given the widespread contamination of ecosystems by various types of MPs, including polystyrene (PS), polyvinyl chloride (PVC), and polyethylene (PE), it is crucial to understand their effects on agricultural productivity. The present study was conducted to investigate the effects of different types of MPs (PS, PVC, and PE) on various aspects of sunflower's (Helianthus annuus L.) growth with the addition of rice straw biochar (RSB). This study aimed to examine plant growth and biomass, photosynthetic pigments and gas exchange characteristics, oxidative stress indicators, and the response of various antioxidants (enzymatic and non-enzymatic) and their specific gene expression, proline metabolism, the AsA–GSH cycle, cellular fractionation in the plants and post-harvest soil properties. The research outcomes indicated that elevated levels of different types of MPs in the soil notably reduced plant growth and biomass, photosynthetic pigments, and gas exchange attributes. Different types of MPs also induced oxidative stress, which caused an increase in various enzymatic and non-enzymatic antioxidant compounds, gene expression and sugar content; pertinently, a significant increase in proline metabolism, AsA–GSH cycle, and pigmentation of cellular components was also observed. Favorably, the addition of RSB significantly increased plant growth and biomass, gas exchange characteristics, enzymatic and non-enzymatic compounds, and relevant genes' expression with a decrease in oxidative stress. In addition, RSB amendment decreased proline metabolism and AsA-GSH cycle in H. annuus plants, resultantly enhancing cellular fractionation with an improvement in post-harvest soil properties. These results open new vistas for sustainable agriculture practices and show great potential for resolving the urgent issues caused by microplastic contamination in agricultural soils.

Evaluation of Some Nanoparticles Against Sclerotium rolfsii the Cause of Root and Crown Rots in Common Beans

This study was conducted to evaluate eco-friendly control agents (carvone, cuminaldehyde, and linalool) against Rhizoctonia solani, which causes root rot disease either by induction of defense response or direct antifungal activity. The induction of resistance was examined by detecting the transcription of defense genes and the effect of the tested control agents on the growth and the yield of common bean plants. The growth of R. solani was significantly inhibited after treatment with the tested compounds compared to the untreated control under laboratory conditions. The disease severity of root rot was decreased in common bean plants treated with the tested compounds compared to untreated control plants under greenhouse conditions. Common bean plants treated with the tested control agents expressed defense genes (Phenylalanine ammonia lyase and β-1,3-Glucanase) involved in jasmonic acid (JA) and salicylic acid (SA) signaling pathways with 2–5 fold higher than the control. Treatment of common beans with the tested control agents and fungicide significantly improved the growth and yield characteristics of common bean. Therefore, the use of monoterpenes could be a novel strategy to control this pathogen and consider the first report.
