Les antivitamines K (AVK), produits de synthèse actifs par voie orale, sont à l’heure actuelle les médicaments les plus efficaces pour réaliser au long cours cette prévention, du moins dans la plupart des indications. Ils inhibent la carboxylation des facteurs II, VII, IX et X et des protéines C et S. La diminution des facteurs de la coagulation vitamine K-dépendants est fonction de leur demi-vie d’élimination, les facteurs à demi-vie la plus courte disparaissent les premiers (notamment les facteurs VII et IX).
