Lipophilic bisphosphonates as dual farnesyl/geranylgeranyl diphosphate synthase inhibitors: an X-ray and NMR investigation
Yonghui Zhang,Rong Cao,Fenglin Yin,Michael P. Hudock,Rey-Ting Guo,Kilannin Krysiak,Sujoy Mukherjee,Yi Gui Gao,Howard Robinson,Yongcheng Song,Joo Hwan No,Kyle Bergan,Annette Leon,Lauren M. Cass,Amanda Goddard,Ting Kai Chang,Fu Yang Lin,Ermond van Beek,Socrates E. Papapoulos,Andrew H.-J. Wang,Tadahiko Kubo,Mitsuo Ochi,Dushyant Mukkamala,Eric Oldfield +23 more
TL;DR: How these compounds inhibit cell growth and how cell activity can be predicted based on enzyme inhibition data is explored, and using X-ray diffraction, solid state NMR, and isothermal titration calorimetry, it is shown how these compounds bind to FPPS and/or GGPPS.
Isoprenoid Biosynthesis as a Drug Target: Bisphosphonate Inhibition of Escherichia coli K12 Growth and Synergistic Effects of Fosmidomycin
Annette Leon,Lei Liu,Yan Yang,Michael P. Hudock,Patrick Hall,Fenglin Yin,Danielle Studer,Kia Joo Puan,Craig T. Morita,Eric Oldfield +9 more
TL;DR: A library of 117 bisphosphonates was screened for antibacterial activity against Escherichia coli, and the activity of the most potent compound, N-[methyl(4-phenylbutyl)]-3-aminopropyl-1-hydroxy-1,1-bisph phosphonate (13), was strongly potentiated by the drug fosmidomycin.

Activity of nitrogen-containing and non-nitrogen-containing bisphosphonates on tumor cell lines.
Yonghui Zhang,Annette Leon,Yongcheng Song,Danielle Studer,Christa Haase,Lukasz A. Koscielski,Eric Oldfield +6 more
TL;DR: It is shown that bisphosphonate esters can have potent activity against a variety of tumor cell lines, with the most active terphenyl- and halophenyl-containing species having IC(50) values approximately 10-40x lower than the most potent commercially available bisph phosphonates.

Synthesis and anticancer activity of new class of bisphosphonates/phosphanamidates.
Y.B. Kiran,C. Devendranath Reddy,D. Gunasekar,C. Suresh Reddy,Annette Leon,Luiz C. A. Barbosa +5 more
TL;DR: Their anticancer activity studies in vitro, on three human tumor cell lines NCI-H460, MCF-7, and SF-268, showed that bis-[3-(3-chloro-4-fluorophenyl)-2-oxo-3,4-dihydro-2H-2lambda(5)-benzo[e][1,3,2]oxazaphosphinin-2-yl]arylmethanes and [(4
