Entrectinib, a Pan-TRK, ROS1, and ALK Inhibitor with Activity in Multiple Molecularly Defined Cancer Indications.
Elena Ardini,Maria Menichincheri,Patrizia Banfi,Roberta Bosotti,Cristina De Ponti,Romana Pulci,Dario Ballinari,Marina Ciomei,Gemma Texido,Anna Degrassi,Nilla Avanzi,Nadia Amboldi,Maria Beatrice Saccardo,Daniele Casero,Paolo Orsini,Tiziano Bandiera,Luca Mologni,David F. Anderson,Ge Wei,Jason Harris,Jean-Michel Vernier,Gang Li,Eduard R. Felder,Daniele Donati,Antonella Isacchi,Enrico Pesenti,Paola Magnaghi,Arturo Galvani +27 more
TL;DR: Entrectinib is a novel, potent inhibitor of ALK, ROS1, and TRK family kinases, which shows promise for therapy of tumors bearing oncogenic forms of these proteins, and is potentially suited to the therapy of several molecularly defined cancer settings, especially that of TRK-dependent tumors, for which no approved drugs are currently available.

Early antiangiogenic activity of SU11248 evaluated in vivo by dynamic contrast-enhanced magnetic resonance imaging in an experimental model of colon carcinoma.
Pasquina Marzola,Anna Degrassi,Laura Calderan,Paolo Farace,Elena Nicolato,Caterina Crescimanno,Marco Sandri,Anna Maria Giusti,Enrico Pesenti,Andrea Terron,Andrea Sbarbati,Francesco Osculati +11 more
TL;DR: The early antiangiogenic activity of SU11248 was detected in vivo by DCE-MRI techniques using either macromolecular or low molecular weight contrast agents, which could be relevant for the design of clinical trials based on new paradigms.

Efficacy of PHA-848125, a cyclin-dependent kinase inhibitor, on the K-Ras(G12D)LA2 lung adenocarcinoma transgenic mouse model: evaluation by multimodality imaging.
Anna Degrassi,M. Russo,Cristina Nanni,Veronica Patton,Rachele Alzani,A. M. Giusti,Stefano Fanti,Marina Ciomei,Enrico Pesenti,Gemma Texido +9 more
TL;DR: Multiodality imaging was successfully used for the preclinical assessment of PHA-848125 therapeutic efficacy on a lung adenocarcinoma mouse model and could represent a valid therapeutic approach for NSCLC patients with mutant K-ras.

NMS-E973, a Novel Synthetic Inhibitor of Hsp90 with Activity against Multiple Models of Drug Resistance to Targeted Agents, Including Intracranial Metastases
Gianpaolo Fogliatto,Laura Gianellini,Maria Gabriella Brasca,Elena Casale,Dario Ballinari,Marina Ciomei,Anna Degrassi,Anna De Ponti,Massimiliano Germani,Marco Guanci,Mauro Paolucci,Paolo Polucci,M. Russo,Francesco Sola,Barbara Valsasina,Carlo Visco,Fabio Zuccotto,Daniele Donati,Eduard R. Felder,Enrico Pesenti,Arturo Galvani,Sergio Mantegani,Antonella Isacchi +22 more
TL;DR: The efficacy profile of NMS-E973 suggests a potential for development in different clinical settings, including tumors that have become resistant to molecular targeted agents, particularly in cases of tumors which reside beyond the BBB.
