Quantitative analysis of the EGF receptor autocrine system reveals cryptic regulation of cell response by ligand capture
TL;DR: Analysis of human mammary epithelial cells showed that a V(LT)/V(R) ratio of < 5 x 10(-4)was sufficient to evoke >20% of a maximal proliferative response, demonstrating that natural autocrine systems can be active even when no ligand appears in the extracellular medium.

Affinity Regulates Spatial Range of EGF Receptor Autocrine Ligand Binding
Ann E. Dewitt,Tomoko Iida,Ho-Yan Lam,Virginia M. Hill,H. Steven Wiley,Douglas A. Lauffenburger +5 more
TL;DR: It is shown that cells can use ligand/receptor binding affinity to regulate ligand spatial distribution when autocrine ligand production is limiting for receptor signaling, and experimental data confirm this prediction.

The effect of cellular parameters on the spatial operation of an autocrine system
Ann E. Dewitt,Douglas A. Lauffenburger,H.S. Wiley +2 more
- 13 Oct 1999
TL;DR: The probability of ligand self-capture at a given secretion rate decreases with decreasing cell surface receptor number and the effect of two parameters on the spatial operation of the EGF/EGFR pair, ligand secretion rate and cell surface receptors number is investigated.
