Ubiquitin proteasome system in immune regulation and therapeutics.
Sameer Ahmed Bhat,Zahra Vasi,Ritika Adhikari,Anish Gudur,Asceal Ali,Liping Jiang,Rachel Ferguson,David Liang,Shafi Kuchay +8 more
TL;DR: The ubiquitin proteasome system (UPS) is a proteolytic machinery for the degradation of protein substrates that are post-translationally conjugated with ubiquin polymers through the enzymatic action of E3-ligases, in a process termed ubiquitylation as mentioned in this paper .

Geranylgeranylated-SCFFBXO10 Regulates Selective Outer Mitochondrial Membrane Proteostasis and Function
Sameer Ahmed Bhat,Zahra Vasi,Liping Jiang,Shruthi Selvaraj,Rachel Ferguson,Anish Gudur,Hagar Ismail,Ritika Adhikari,Avantika Dhabaria,Beatrix M. Ueberheide,Shafi Kuchay +10 more
TL;DR: Examination of the role of this pathway in a physiological context revealed that the loss of FBXO10 or expression of prenylation-deficient-FBXO10(C953S) inhibited PGAM5 degradation, disrupted mitochondrial homeostasis, and impaired myogenic differentiation of human iPSCs and murine myoblasts.
Palmitoylation and PDE6δ regulate membrane-compartment-specific substrate ubiquitylation and degradation
David Liang,L. Jiang,Sameer Ahmed Bhat,Sonia Missiroli,Maria Rita Perrone,Angela Lauriola,Ritika Adhikari,Anish Gudur,Zahra Vasi,Ian M. Ahearn,Daniele Guardavaccaro,Carlotta Giorgi,Mark R. Philips,Shafi Kuchay +13 more
TL;DR: In this paper , the interplay of two lipid modifications and PDE6δ regulates compartmental substrate targeting via the SCFFBXL2, which is palmitoylated in a prenylation-dependent manner on cysteines 417 and 419 juxtaposed to the CaaX motif.
