Interest in individual differences in animal behavioural plasticities has surged in recent years, but research in this area has been hampered by semantic confusion as different investigators use the same terms (e.g. plasticity, flexibility, responsiveness) to refer to different phenomena. The first goal of this review is to suggest a framework for categorizing the many different types of behavioural plasticities, describe examples of each, and indicate why using reversibility as a criterion for categorizing behavioural plasticities is problematic. This framework is then used to address a number of timely questions about individual differences in behavioural plasticities. One set of questions concerns the experimental designs that can be used to study individual differences in various types of behavioural plasticities. Although within-individual designs are the default option for empirical studies of many types of behavioural plasticities, in some situations (e.g. when experience at an early age affects the behaviour expressed at subsequent ages), ‘replicate individual’ designs can provide useful insights into individual differences in behavioural plasticities. To date, researchers using within-individual and replicate individual designs have documented individual differences in all of the major categories of behavioural plasticities described herein. Another important question is whether and how different types of behavioural plasticities are related to one another. Currently there is empirical evidence that many behavioural plasticities [e.g. contextual plasticity, learning rates, IIV (intra-individual variability), endogenous plasticities, ontogenetic plasticities) can themselves vary as a function of experiences earlier in life, that is, many types of behavioural plasticity are themselves developmentally plastic. These findings support the assumption that differences among individuals in prior experiences may contribute to individual differences in behavioural plasticities observed at a given age. Several authors have predicted correlations across individuals between different types of behavioural plasticities, i.e. that some individuals will be generally more plastic than others. However, empirical support for most of these predictions, including indirect evidence from studies of relationships between personality traits and plasticities, is currently sparse and equivocal. The final section of this review suggests how an appreciation of the similarities and differences between different types of behavioural plasticities may help theoreticians formulate testable models to explain the evolution of individual differences in behavioural plasticities and the evolutionary and ecological consequences of individual differences in behavioural plasticities.

Individual differences in behavioural plasticities

When making decisions, animals, including humans, do not always choose the same option. One reason for this is that their “internal state” changes the value of different options. This is particularly evident when deciding what type of food to eat. Depending on which nutrients the animal needs, it will choose to eat different foods. Amino acids are key nutrients that affect health, lifespan and reproduction. Female fruit flies that have recently mated, for example, eat more amino acids in order to obtain the raw materials required to produce eggs. Despite the importance of amino acids, little was known about how animal behavior changes in response to a lack of this nutrient. Corrales-Carvajal et al. used a video tracking system to measure the time that fruit flies – some of which had a need for amino acids – spent feeding on patches of yeast (which are rich in amino acids) versus patches of sucrose. Recently mated females – and virgins that had been fed a diet lacking in amino acids – consumed more yeast than sucrose, whereas virgin females that were not amino acid deficient showed the opposite pattern. To bias the fly toward eating the right food for their needs, several aspects of the fly’s behavior changed, including the number and length of individual feeding bouts. These different behaviors did not all change at the same time. The pattern of exploration taken by the flies also depended on their need for amino acids. Amino acid deficient flies spent most of their time near known yeast patches. By contrast, fully fed flies adopted a riskier foraging strategy, moving away from known sources of food to explore their environment more widely. In common with humans, the flies relied upon their sense of smell to efficiently identify different types of food. Overall, the results presented by Corrales-Carvajal et al. provide us with a detailed understanding about how changes to the internal state of the fly affect its behavior. The next step will be to use the powerful genetic tools available for studying fruit flies to reveal the neural circuits and molecular mechanisms that help animals find the types of food that they need.

Internal states drive nutrient homeostasis by modulating exploration-exploitation trade-off

Activity patterns and sleep-wake cycles are among the physiological processes that change most prominently as animals age, and are often good indicators of healthspan. In this study, we used the video-based high-resolution Behavioral Monitoring System (BMS) to monitor the daily activity cycle of tephritid fruit flies Anastrepha ludens over their lifetime. Surprisingly, there was no dramatic change in activity profile with respect to age if flies were consistently fed with a nutritionally balanced diet. However, if flies were fed with sugar-only diet, their activity profile decreased in amplitude at old age, suggesting that suboptimal diet affected activity patterns, and its detrimental effect may not manifest itself until the animal ages. Moreover, by simulating different modes of behavior monitoring with a range of resolution and comparing the resulting conclusions, we confirmed the superior performance of video-based monitoring using high-resolution BMS in accurately representing activity patterns in an insect model.

CORRIGENDUM: Deleterious effect of suboptimal diet on rest-activity cycle in Anastrepha ludens manifests itself with age

Functional data analysis (FDA) is concerned with inherently infinite-dimensional data objects and therefore can be viewed as part of the methodology for big data. The size of functional data may vary from terabytes as encountered in functional magnetic resonance imaging (fMRI) and other applications in brain imaging to just a few kilobytes in longitudinal data with small or modest sample sizes. In this contribution, we highlight some applications of FDA methodology through various data illustrations. We briefly review some basic computational tools that can be used to accelerate implementations of FDA methodology. The analyses presented in this paper illustrate the principal analysis by conditional expectation (PACE) package for FDA, where our applications include both relatively simple and more complex functional data from the biomedical sciences. The data we discuss range from functional data that result from daily movement profile tracking and that are modeled as repeatedly observed functions per subject, to medfly longitudinal behavior profiles, where the goal is to predict remaining lifetime of individual flies. We also discuss the quantification of connectivity of fMRI signals that is of interest in brain imaging and the prediction of continuous traits from high-dimensional SNPs in genomics. The methods of FDA that we demonstrate for these analyses include functional principal component analysis, functional regression and correlation, the modeling of dependent functional data, and the stringing of high-dimensional data into functional data and can be implemented with the PACE package.

Quantifying Infinite-Dimensional Data: Functional Data Analysis in Action

BACKGROUND

The Sterile Insect Technique (SIT) requires vast numbers of consistently high quality insects to be produced over long periods. Quality control (QC) procedures are critical to effective SIT, both providing quality assurance and warning of operational deficiencies. We here present a potential new QC assay for mass rearing of Queensland fruit flies (Bactrocera tryoni Froggatt) for SIT; locomotor activity monitoring. We investigated whether automated locomotor activity monitors (LAMs) that simply detect how often a fly passes an infrared sensor in a glass tube might provide similar insights but with much greater economy.



RESULTS

Activity levels were generally lower for females than for males, and declined over five days in the monitor for both sexes. Female activity levels were not affected by irradiation, but males irradiated at 60 or 70 Gy had reduced activity levels compared with unirradiated controls. We also found some evidence that mild heat shock of pupae results in adults with reduced activity.



CONCLUSION

LAM offers a convenient, effective and economical assay to probe such changes. © 2013 Society of Chemical Industry

Automated locomotor activity monitoring as a quality control assay for mass‐reared tephritid flies

Characterization of lifetime behavioral changes is essential for understanding aging and aging-related diseases. However, such studies are scarce partly due to the lack of efficient tools. Here we describe and provide proof of concept for a stereo vision system that classifies and sequentially records at an extremely fine scale six different behaviors (resting, micro-movement, walking, flying, feeding and drinking) and the within-cage (3D) location of individual tephritid fruit flies by time-of-day throughout their lives. Using flies fed on two different diets, full sugar-yeast and sugar-only diets, we report for the first time their behavioral changes throughout their lives at a high resolution. We have found that the daily activity peaks at the age of 15–20 days and then gradually declines with age for flies on both diets. However, the overall daily activity is higher for flies on sugar-only diet than those on the full diet. Flies on sugar-only diet show a stronger diurnal localization pattern with higher preference to staying on the top of the cage during the period of light-off when compared to flies on the full diet. Clustering analyses of age-specific behavior patterns reveal three distinct young, middle-aged and old clusters for flies on each of the two diets. The middle-aged groups for flies on sugar-only diet consist of much younger age groups when compared to flies on full diet. This technology provides research opportunities for using a behavioral informatics approach for understanding different ways in which behavior, movement, and aging in model organisms are mutually affecting.

Hypotheses exploring the influence of dietary conditions on the life-history trade-off between survival and reproductive success are extensively tested in female insects but only rarely explored in males. The present study examines the impact of dietary quality and female access on age-specific reproduction and survival of the male Mexican fruit fly Anastrepha ludens Loew (Diptera: Tephritidae). There is a clear cost of female access for males with access to dietary protein, measurable as a decrease in life expectancy, which is further influenced by the age when females are introduced. A protein deficient diet reduces the lifespan benefit of virginity and masks the detrimental effect of female access on male life expectancy. Dietary protein is not necessary for reproductive success, although access to protein at eclosion improves the lifetime reproductive success of males compared to when it is delayed. Overall, reproductive success diminishes as the male flies age, regardless of the dietary conditions, providing evidence for reproductive senescence in males. Delaying the males' access to a protein source fails to influence the negative effect of age on reproductive ability. Because age-specific reproductive rates decline with age, regardless of diet, male fitness does not benefit from lifespan extension. Therefore, males can be expected to allocate available resources towards reproductive effort in favour of an extended lifespan, regardless of mate and protein availability.
