Heparanase Enhances Syndecan-1 Shedding A NOVEL MECHANISM FOR STIMULATION OF TUMOR GROWTH AND METASTASIS
Yang Yang,Veronica MacLeod,Hua-Quan Miao,Allison Theus,Fenghuang Zhan,John D. Shaughnessy,Jeffrey R. Sawyer,Jin-Ping Li Li,Eyal Zcharia,Israel Vlodavsky,Ralph D. Sanderson +10 more
TL;DR: It is reported that tumor cell expression of heparanase regulates both the level and location of syndecan-1 within the tumor microenvironment by enhancing its synthesis and subsequent shedding from the tumor cell surface, and suggests that heParanase and syndecans act synergistically to fine tune the tumormicroenvironment and ensure robust tumor growth.

Soluble syndecan-1 promotes growth of myeloma tumors in vivo.
Yang Yang,Shmuel Yaccoby,Wei Liu,J. Kevin Langford,Carla Y. Pumphrey,Allison Theus,Joshua Epstein,Ralph D. Sanderson +7 more
TL;DR: High levels of soluble syndecan-1 present in patients with myeloma may contribute directly to the growth and dissemination of the malignant cells and thus to poor prognosis.

Enzymatic remodeling of heparan sulfate proteoglycans within the tumor microenvironment: growth regulation and the prospect of new cancer therapies.
TL;DR: This review focuses on remodeling of HSPGs by three distinct mechanisms that occur in vivo; shedding of proteoglycan extracellular domains from cell surfaces, fragmentation of heparan sulfate chains byHeparanase, and removal of sulfates from the 6‐O position of heParan sulfates chains by ext racellular sulfatases.

Heparanase promotes the spontaneous metastasis of myeloma cells to bone.
Yang Yang,Veronica MacLeod,Manali S. Bendre,Yan Huang,Allison Theus,Hua-Quan Miao,Paul Kussie,Shmuel Yaccoby,Joshua Epstein,Larry J. Suva,Thomas J. Kelly,Ralph D. Sanderson +11 more
TL;DR: It is indicated that heparanase is a critical determinant of myeloma dissemination and growth in vivo and described a novel experimental animal model for examining the spontaneous metastasis of bone-homing tumors.

Elevated levels of shed syndecan-1 correlate with tumour mass and decreased matrix metalloproteinase-9 activity in the serum of patients with multiple myeloma
Madhav V. Dhodapkar,Thomas J. Kelly,Allison Theus,Anupama B. Athota,Barthel Barlogie,Ralph D. Sanderson +5 more
TL;DR: Sera from 20 myeloma patients and 12 normal controls were analysed for the presence of syndecan‐1 and matrix metalloproteinase‐9 and provided evidence that the syndecans‐1 ectodomain is shed in vivo.
