A rapid method for cross‐species quantitation of apolipoproteins A1, B48 and B100 in plasma by ultra‐performance liquid chromatography/tandem mass spectrometry
Michael E. Lassman,Theresa McLaughlin,Elizabeth Polizzi Somers,Alice C. Stefanni,Zhu Chen,Beth Ann Murphy,Kathleen K. Bierilo,Amy M. Flattery,Kenneth K. Wong,Jose Castro-Perez,Brian K. Hubbard,Thomas P. Roddy +11 more
TL;DR: This assay is capable of supporting cardiovascular research by providing a single assay to measure critical biomarkers across multiple species without the need for antibodies, and does so in a high-throughput manner.

AAV8-mediated long-term expression of human LCAT significantly improves lipid profiles in hCETP;Ldlr(+/-) mice.
Zhu Chen,Donald Chu,Jose Castro-Perez,Weihua Ni,Aiwu Zhang,Mihajlo L. Krsmanovic,Dan Xie,Vinit Shah,Steven J. Stout,David G. McLaren,Alice C. Stefanni,Sang Ho Lee,Thomas P. Roddy,Andrew S. Plump,Brian K. Hubbard,Thomas F. Vogt,Heather Zhou +16 more
TL;DR: In this human-like model for lipoprotein metabolism, AAV8-mediated overexpression of human LCAT resulted in profound changes in plasma lipid profiles, suggesting that LCAT's beneficial effect on lipid metabolism includes not only enhanced HDL cholesterol esterification but also improved metabolism of apoB-containing particles and triglycerides.

Validation of Human ApoB and ApoAI Immunoturbidity Assays for Non-human Primate Dyslipidemia and Atherosclerosis Research
Zhu Chen,Alison M. Strack,Alice C. Stefanni,Ying Chen,Weizhen Wu,Yi Pan,Olga Urosevic-Price,Li Wang,Theresa McLaughlin,Neil S. Geoghagen,Michael E. Lassman,Thomas P. Roddy,Kenny K. Wong,Brian K. Hubbard,Amy M. Flattery +14 more
TL;DR: Analysis of purified lipoproteins demonstrated that the immunoturbidity assays detect NHP apoAI and apoB, with good dilution linearity and spike recovery from NHP plasma, and demonstrate that these assays provide a high-throughput, fully automated analytical platform for NHP samples.

ApoB siRNA-induced Liver Steatosis is Resistant to Clearance by the Loss of Fatty Acid Transport Protein 5 (Fatp5)
Brandon Ason,Brandon Ason,Jose Castro-Perez,Jose Castro-Perez,Samnang Tep,Alice C. Stefanni,Marija Tadin-Strapps,Thomas P. Roddy,Thomas Hankemeier,Brian K. Hubbard,Alan B. Sachs,W. Michael Flanagan,Nelly A. Kuklin,Lyndon J. Mitnaul +13 more
TL;DR: It was determined that, although Fatp5 siRNA treatment was sufficient to increase the proportion of unconjugated bile acids 100-fold, consistent with FATP5's role in bile acid reconjugation,Fatp5 knockdown failed to influence the degree, zonal distribution, or composition of the hepatic triglycerides that accumulated following ApoB si RNA treatment.
Depot-specific regulation of the conversion of cortisone to cortisol in human adipose tissue.
Mi-Jeong Lee,Susan K. Fried,Susan K. Fried,Susan K. Fried,Steven S. Mundt,Yanxin Wang,Sean Sullivan,Alice C. Stefanni,Bruce L. Daugherty,Anne Hermanowski-Vosatka +9 more
TL;DR: The main objective was to compare the regulation of cortisol production within omental (Om) and abdominal subcutaneous (Abd sc) human adipose tissue.
