Nitrogen fixation is mediated by a variety of autotrophic and heterotrophic bacteria. Cyanobacteria appear responsible for most planktonic fixation in aquatic ecosystems, and rates of fixation are high only when thcsc organisms make up a major percentage of the planktonic biomass, Planktonic nitrogen fixation tends to be low in oligotrophic and mesotrophic lakes (generally 20% of the nitrogen input to the Asko region of the Baltic Sea and 17% of the nitrogen input to the PeelHarvey estuary in Australia. Fixation in sediments of estuaries and eutrophic and mesotrophic lakes usually constitutes a small percentage of the nitrogen inputs to these systems. However, benthic fixation appears to be a major source of nitrogen for many oligotrophic tropical lagoons and for some oligotrophic lakes, even though fixation rates are moderate because other nitrogen inputs tend to be low. Nitrogen fixation probably is a fairly minor input of nitrogen to marine wetlands, which are generally open to other inputs, but contributes roughly half the total nitrogen input to some freshwater wetlands (bogs, cypress domes), where other inputs are more limited. Nitrogen fixation appears important in making up deficits in nitrogen availability relative to phosphorus availability in many lakes, contributing to the phosphorus-limited status of these systems. That many estuaries and coastal seas are nitrogen limited is due in part to the generally low rates of nitrogen fixation found in these systems.

w Traditionally, nitrogen control is generally considered an important component of reducing lake eutrophication and cyanobacteria blooms. However, this viewpoint is refuted recently by researchers in China and North America. In the present paper, the traditional viewpoint of nitrogen control is pointed out to lack a scientific basis: the N/P hypothesis is just a subjective assumption; bottle bioassay experiments fail to simulate the natural process of nitrogen fixation. Our multi-year comparative research in more than 40 Yangtze lakes indicates that phosphorus is the key factor determining phytoplankton growth regardless of nitrogen concentrations and that total phytoplankton biomass is determined by total phosphorus and not by total nitrogen concentrations. These results imply that, in the field, nitrogen control will not decrease phytoplankton biomass. This finding is supported by a long-term whole-lake experiment from North America. These outcomes can be generalized in terms that a reduction in nitrogen loading may not decrease the biomass of total phytoplankton as it can stimulate blooms of nitrogen-fixing cyanobacteria. To mitigate eutrophication, it is not nitrogen but phosphorus that should be reduced, unless nitrogen concentrations are too high to induce direct toxic impacts on human beings or other organisms. Mitigation of lake eutrophication: Loosen nitrogen control and focus on phosphorus abatement

The effect of soil water content [60%–100% water-holding capacity (WHC)] on N2O production during autotrophic nitrification and denitrification in a loam soil was studied in a laboratory experiment by selectively inhibiting nitrification with a low C2H2 concentration (2.1 Pa). Nitrifiers usually produced more N2O than denitrifiers. During an initial experimental period of 0–6 days the nitrifiers produced more N2O than the denitrifiers by a factor ranging from 1.4 to 16.5, depending on the water content and length of incubation. The highest N2O production rate by nitrifiers was observed at 90% WHC, when the soil had become partly anaerobic, as indicated by the high denitrification rate. At 100% WHC there were large gaseous losses from denitrification, while nitrification losses were smaller except for the first period of measurement, when there was still some O2 remaining in the soil. The use of 10 kPa C2H2 to inhibit reduction of N2O to N2 stimulated the denitrification process during prolonged incubation over several days; thus the method is unsuitable for long-term studies.

Relationships between soil moisture content and nitrous oxide production during nitrification and denitrification

Nitrapyrin and C2H2 were evaluated as nitrification inhibitors in soil to determine the relative contributions of denitrification and nitrification to total N2O production. In laboratory experiments nitrapyrin, or its solvent xylene, stimulated denitrification directly or indirectly and was therefore considered unsuitable. Low partial pressures of C2H2 (2.5–5.0 Pa) inhibited nitrification and had only a small effect on denitrification, which made it possible to estimate the contribution of denitrification. The contribution of nitrification was estimated by subtracting the denitrification value from total N2O production (samples without C2H2). The critical C2H2 concentrations needed to achieve inhibition of nitrification, without affecting the N2O reductase in denitrifiers, must be individually determined for each set of experimental conditions.

A method of selective inhibition to distinguish between nitrification and denitrification as sources of nitrous oxide in soil

Nitrogen (N) fixation is fueling planktonic production in a multitude of aquatic environments. In meso- and poly-haline estuaries, however, the contribution of N by pelagic N2 fixation is believed to be insignificant due to the high input of N from land and the presumed absence of active N2-fixing organisms. Here we report N2 fixation rates, nifH gene composition and nifH gene transcript abundance for key diazotrophic groups over 1 year in two contrasting, temperate, estuarine systems: Roskilde Fjord (RF) and the Great Belt (GB) strait. Annual pelagic N2 fixation rates averaged 17 and 61 mmol N m−2 per year at the two sites, respectively. In RF, N2 fixation was mainly accompanied by transcripts related to heterotrophic (for example, Pseudomonas sp.) and photoheterotrophic bacteria (for example, unicellular diazotrophic cyanobacteria group A). In the GB, the first of two N2 fixation peaks coincided with a similar nifH-expressing community as in RF, whereas the second peak was synchronous with increased nifH expression by an array of diazotrophs, including heterotrophic organisms as well as the heterocystous cyanobacterium Anabaena. Thus, we show for the first time that significant planktonic N2 fixation takes place in mesohaline, temperate estuaries and that the importance of heterotrophic, photoheterotrophic and photosynthetic diazotrophs is clearly variable in space and time.

Significant N 2 fixation by heterotrophs, photoheterotrophs and heterocystous cyanobacteria in two temperate estuaries

In situ fixation of molecular nitrogen was mcasurcd in 19170 in the pelagial of Lake Erkcn by the acetylene reduction technique every 2 weeks during a period of 5 months. The diurnal cycles and vertical distribution of algae were also investigated. Fixation was correlated with the presence of heterocystous blue-green algae, especially Aphanixomenon, in the phytoplankton, and was light dependent, though appreciable dark fixation also occurred, owing to endogenous utilization of photosynthetic products formed during previous light periods. The annual contribution of nitrogen fixation in the pelagial was of the order 0.5 g N/m’. The highest values were obtained before mass development of Aphanixomenon and maximal primary production. The algae showed highest nitrogenase activity in the first period of algal development. Lake Erken is moderately eutrophic and unpolluted. The contribution of easily available combined nitrogen by pelagic nitrogen fixation increases the annual loading of combined nitrogen by 40% and must therefore be considered quite important.
