Glucocorticoids Suppress Bone Formation by Attenuating Osteoblast Differentiation via the Monomeric Glucocorticoid Receptor
Alexander Rauch,Sebastian Seitz,Ulrike Baschant,Arndt F. Schilling,Anett Illing,Brenda D. Stride,Milen Kirilov,Vice Mandic,Andrea Takacz,Ruth Schmidt-Ullrich,Susanne Ostermay,Thorsten Schinke,Rainer Spanbroek,Mario M. Zaiss,Peter Angel,Ulf H. Lerner,Ulf H. Lerner,Jean-Pierre David,Holger M. Reichardt,Michael Amling,Günther Schütz,Jan Tuckermann +21 more
TL;DR: It is demonstrated that GCs are unable to repress bone formation in the absence of glucocorticoid receptor (GR) expression in osteoblasts as they become refractory to hormone-induced apoptosis, inhibition of proliferation, and differentiation.

Obesity-Associated Hypermetabolism and Accelerated Senescence of Bone Marrow Stromal Stem Cells Suggest a Potential Mechanism for Bone Fragility.
Michaela Tencerova,Michaela Tencerova,Morten Frost,Morten Frost,Florence Figeac,Tina Nielsen,Dalia Ali,Jens Jacob Lindegaard Lauterlein,Thomas Levin Andersen,Thomas Levin Andersen,Anders K. Haakonsson,Anders K. Haakonsson,Alexander Rauch,Jonna Skov Madsen,Charlotte Ejersted,Kurt Højlund,Kurt Højlund,Moustapha Kassem,Moustapha Kassem +18 more
TL;DR: The data provide a plausible explanation for the bone fragility in obesity caused by enhanced insulin signaling leading to accelerated metabolic senescence of BM-MSCs.

DNA binding - dependent glucocorticoid receptor activity promotes adipogenesis via krüppel-like factor 15 gene expression
Maki Asada,Alexander Rauch,Hirohito Shimizu,Hiromi Maruyama,Shigeru Miyaki,Masafumi Shibamori,Hideki Kawasome,Hironobu Ishiyama,Jan Tuckermann,Hiroshi Asahara,Hiroshi Asahara +10 more
TL;DR: It is shown that inhibition of GR activity using the GR antagonist RU486 prevents human mesenchymal stem cell and mouse embryonic fibroblast differentiation into adipocytes and that GR promotes adipogenesis via KLF15 gene expression as a transcriptional direct target.

Transcriptional networks controlling stromal cell differentiation.
Alexander Rauch,Susanne Mandrup +1 more
TL;DR: In this article, mesenchymal stromal cells (MSCs) were found to exhibit a marked plasticity to differentiate into many different cell lineages and showed the involvement of specific signalling pathways and key transcriptional regulators.

High‐fat diet‐induced obesity augments the deleterious effects of estrogen deficiency on bone: Evidence from ovariectomized mice
Dalia Ali,Florence Figeac,Atenisa Caci,Nicholas Ditzel,Clarissa Schmal,Greet Kerckhofs,Jesper F. Havelund,N. Faergeman,Alexander Rauch,Michaela Tencerova,Moustapha Kassem +10 more
TL;DR: Obesity and estrogen deficiency exert combined deleterious effects on bone resulting in accelerated cellular senescence, expansion of BMAT and impaired bone formation leading to decreased bone mass, suggesting that obesity may increase bone fragility in postmenopausal women.
